Complement, a complex protein system, plays an essential role in host defense through bacterial lysis, stimulation of phagocytosis, recruitment of immune cells to infected tissue, and promotion of the inflammatory response. Although complement is most well-characterized in serum, complement activity is also present in the lung. Here we further characterize the complement system in the normal and inflamed lung. By Western blot, C5, C6, and factor I were detected in bronchoalveolar lavage (BAL) at lower levels than in serum, whereas C2 was detected at similar levels in BAL and serum. C4 binding protein (C4BP) was not detectable in BAL. Exposure to lipopolysaccharide (LPS) elevated levels of C1q, factor B, C2, C4, C5, C6, and C3 in human BAL and C3, C5, and factor B in mouse and rat BAL. Message for C1q-B, C1r, C1s, C2, C4, C3, C5, C6, factor B, and factor H, but not C9 or C4BP, was readily detectable by RT-PCR in normal mouse lung. Exposure to LPS enhanced factor B expression, decreased C5 expression, and did not affect C1q-B expression in mouse and rat lung. BAL from rats exposed to LPS had a greater ability to deposit C3b onto bacteria through complement activation than did BAL from control rats. In summary, these data demonstrate that complement levels, expression, and function are altered in acute lung injury and suggest that complement within the lung is regulated to promote opsonization of pathogens and limit potentially harmful inflammation.

O παράγοντας SAFB1 (Scaffold Attachment Factor B) είναι πρωτεΐνη της πυρηνικής μήτρας και απομονώθηκε πριν από είκοσι περίπου χρόνια με βάση το χαρακτηριστικό του να προσδένεται σε στοιχεία S/MAR (Scaffold/Matrix Attachment Region) του DNA. Ο SAFB1 με τον εξελικτικά συγγενή του SAFB2, έχουν μεταξύ τους ομολογία 74% και αποτελούν αντικείμενο έρευνας της παρούσας διδακτορικής διατριβής. Αποτελούνται από την περιοχή πρόσδεσης στο DNA, SAF-box στο αμινοτελικό τους άκρο, την περιοχή δέσμευσης με το RNA, RRM, ενώ στο καρβοξυτελικό τους άκρο έχουν μια περιοχή πλούσια σε επαναλήψεις Arg-Glu (R/E) που ακολουθείται από μια περιοχή πλούσια σε κατάλοιπα Arg και Gly (R, G), υπεύθυνη για τις γνωστές ως τώρα πρωτεϊνικές τους αλληλεπιδράσεις. Στο πρώτο μέρος της διατριβής εξετάστηκε η ικανότητα πρόσδεσης των SAFB στη χρωματίνη μέσω της αμινοτελικής τους περιοχής, υπό την επίδραση δυο μεταβολικών στρεσσογόνων συνθηκών, της υποξίας και της στέρησης γλυκόζης. Για κάθε συνθήκη εξετάστηκαν τα πρωτεϊνικά επίπεδα και ο υποκυτταρικός εντοπισμός τους. Τα αποτελέσματα έδειξαν ότι και οι δύο συνθήκες δεν επηρεάζουν τον υποκυτταρικό εντοπισμό τους αλλά τα πρωτεϊνικά επίπεδά τους. Συγκεκριμένα, η υποξία αυξάνει τα πρωτεϊνικά επίπεδα των SAFB1/2 όχι όμως στο επίπεδο της μεταγραφής και με μηχανισμό ανεξάρτητο των μεταγραφικών παραγόντων HIFs (Hypoxia-inducible factors), αλλά προκαλώντας αύξηση του διαλυτού κλάσματός τους στο πυρηνόπλασμα. Επίσης η υποξία δεν επηρέασε την δράση τους ως συγκαταστολείς της μεταγραφικής ενεργότητας του ERα (Estrogen Receptor α). Αντίθετα, η στέρηση γλυκόζης φάνηκε να μειώνει τα πρωτεϊνικά επίπεδα των SAFB1/2, χωρίς όμως να αποκλείεται αυτό να συμβαίνει στο επίπεδο της μεταγραφής.Στο δεύτερο μέρος διερευνήθηκαν οι λειτουργίες των SAFB μέσω δύο αλληλεπιδράσεων που έχουν βρεθεί στο εργαστήριο Βιοχημείας και συμβαίνουν με το καρβοξυτελικό τους άκρο. Η πρώτη αλληλεπίδραση είναι με την κινάση SRPK1 (Serine/Arginine Protein Kinase 1). Καταρχάς εξετάστηκε δομικά, όπου με πειράματα χρωματογραφίας αγχιστείας εντοπίστηκε για πρώτη φορά, ότι τα 180 C-τελικά αμινοξέα της SRPK1(473-655) είναι ικανά και απαραίτητα για την αλληλεπίδραση της με τον SAFB1. Επίσης, διερευνήθηκε λειτουργικά, μέσω εύρεσης συνθηκών όπου τα δύο μόρια SAFB και SRPK1, τα οποία κατά τ’ άλλα έχουν διαφορετικές υποκυτταρικές κατανομές, είναι πιθανόν να βρεθούν σε κοινό υποκυτταρικό εντοπισμό. Εξετάστηκαν τα πρωτεϊνικά επίπεδα και ο υποκυτταρικός εντοπισμός της SRPK1 υπό την επίδραση της υποξίας, του οσμωτικού στρες, της στέρησης αμινοξέων και της στέρησης γλυκόζης. Τα αποτελέσματα έδειξαν ότι καμία από τις τέσσερις συνθήκες δεν ήταν ικανή να μεταβάλλει τα πρωτεϊνικά επίπεδα της SRPK1, ενώ μόνο οι συνθήκες του οσμωτικού στρες και της στέρησης γλυκόζης προκάλεσαν μια υποκυτταρική μετατόπιση της SRPK1, σε διακριτές πυρηνικές δομές. Η δεύτερη αλληλεπίδραση που εξετάστηκε ήταν με την πρωτεΐνη ERH (Enhancer of Rudimentary Homologue), η οποία εμπλέκεται σε ποικίλες βασικές κυτταρικές διεργασίες μεταξύ των οποίων και το μάτισμα. Με πειράματα ανοσοκατακρήμινσης και χρωματογραφίας αγχιστείας βρέθηκε ότι η ERH αλληλεπιδρά άμεσα μέσα σε κύτταρα θηλαστικών μέσω των 205 καρβοξυτελικών αμινοξικών καταλοίπων των SAFB, ενώ από πειράματα ανοσοφθορισμού και βιοχημικής κλασμάτωσης δείχθηκε ότι οι δυο πρωτεΐνες συν-εντοπίζονται στην πυρηνική μήτρα. Τέλος, η ERH δεν μεταβάλλει τη λειτουργία των SAFB1/2 ως συγκαταστολείς του υποδοχέα των οιστρογόνων ERα αλλά αναιρεί την καταστολή που προκαλούν στην ενζυμική δράση της SRPK1, λογικό επακόλουθο του γεγονότος ότι SRPK1και ERH προσδένονται στην ίδια επικράτεια των SAFB1/2.Συνολικά τα παραπάνω ευρήματα θα βοηθήσουν στην εμβάθυνση και την κατανόηση των λειτουργιών των SAFB1/2 και των μοριακών παρτενέρ τους, οι οποίοι εμπλέκονται σε βασικές κυτταρικές διεργασίες όπως είναι η οργάνωση της χρωματίνης, ο μεταβολισμός του RNA και η μεταγραφή.

Tunicamycin, an inhibitor of N-acetylglucosaminylpyrophosphopolyisoprenol-dependent glycosylation, was used to study the effect of glycosylation on the synthesis, post-translational modification, secretion and function of the complement proteins that are associated with the major histocompatibility complex in humans, mice and guinea pigs. Tunicamycin blocked glycosylation of pro-C4, C2 and factor B and inhibited secretion of the corresponding native complement proteins synthesized by guinea-pig peritoneal macrophages in tissue culture. In addition, underglycosylated pro-C4 was more rapidly catabolized intracellularly than the corresponding fully glycosylated pro-complement protein. C4 protein secreted by cells incubated with tunicamycin had approximately the same specific biological activity as the protein obtained from control culture media, suggesting that carbohydrate is not required for its activity in immune haemolysis. Direct studies of carbohydrate incorporation and the tunicamycin effect suggested an unequal distribution of sugar among the C4 subunits, with maximal incorporation of carbohydrate into alpha-, and less into the beta-chain of the native protein.
