In yeast mitochondria, most of the isoaccepting species of tyrosyl tRNA are coded by a mitochondrial gene, tyrA. A particular isoaccepting species is coded by a second mitochondrial gene, tyrB. This gene is not expressed in certain strains of yeast which show no deficient phenotype. Genetic crosses between strains expressing or not expressing the tyrB gene demonstrate that expression is controlled by specific nuclear genes and that a mutation of the tyrA gene can be bypassed when the tyrB gene is operative.

Barley cultivars show various patterns of resistance against isolates of Magnaporthe oryzae and M. grisea. Genetic mechanisms of the resistance of five representative barley cultivars were examined using a highly susceptible barley cultivar, ‘Nigrate’, as a common parent of genetic crosses. The resistance of the five cultivars against Setaria, Oryza, Eleusine, and Triticum isolates of M. oryzae was all attributed to a single locus, designated as Rmo2. Nevertheless, the Rmo2 locus in each cultivar was effective against a different range of isolates. Genetic analyses of pathogenicity suggested that each cultivar carries an allele at the Rmo2 locus that recognizes a different range of avirulence genes. One allele, Rmo2.a, corresponded to PWT1, which conditioned the avirulence of Setaria and Oryza isolates on wheat, in a gene-for-gene manner. The other alleles, Rmo2.b, Rmo2.c, and Rmo2.d, corresponded to more than one avirulence gene. On the other hand, the resistance of those cultivars to another species, M. grisea, was conditioned by another locus, designated as Rmo3. These results suggest that Rmo2 is effective against a broad range of blast isolates but is specific to M. oryzae. Molecular mapping revealed that Rmo2 is located on the 7H chromosome.

Abstract Previous studies of bacteriophage lambda recombination have provided indirect evidence that substantial sequence nonhomologies, such as insertions and deletions, may be included in regions of heteroduplex DNA. However, the direct products of heterology-containing heteroduplex DNA--heterozygous progeny phage--have not been observed. We have constructed a series of small insertion and deletion mutations in the cI gene to examine the possibility that small heterologies might be accommodated in heterozygous progeny phage. Genetic crosses were carried out between lambda cI- Oam29 and lambda cI+ Pam80 under replication-restricted conditions. Recombinant O+P+ progeny were selected on mutL hosts and tested for cI heterozygosity. Heterozygous recombinants were readily observed with crosses involving insertions of 4 to 19 base pairs (bp) in the cI gene. Thus, nonhomologies of at least 19 bp can be accommodated in regions of heteroduplex DNA during lambda recombination. In contrast, when a cI insertion or deletion mutation of 26 bp was present, few of the selected recombinants were heterozygous for cI. Results using a substitution mutation, involving a 26-bp deletion with a 22-bp insertion, suggest that the low recovery of cI heterozygotes containing heterologies of 26 bp or more is due to a failure to encapsulate DNA containing heterologies of 26 bp or more into viable phage particles.

The study examined the reproductive indices of albino and normal pigmented Clarias gariepinus fish from Katsina (KT) and Yola (YY), carried out under hatchery condition. The experiment compared the fecundity, testes, milt volume, percentage fertilization and hatchability of albino (AA) and normal pigmented (NN) Clarias gariepinus from Katsina (KT) and Yola (YY). Normal pigmented C. gariepinus from Katsina (KT) had the highest number of eggs (229,240), followed by YY (127,250) and the least was recorded in the Albino (AA) (124,750). The weights and lengths of the left and right lobes as well as the volumes of the milt were quantified . However, KT had the highest weight and length of the right testis (18g and 5.7cm respectively) while AA had the highest weight, length of the left testis and milt volume (30g, 11cm and 8.4ml respectively). The least length and weight of the testes was observed in YY. Meanwhile, KT and YY had milt volumes of 4.0ml and 4.7ml, respectively. The highest percentage fertilization and hatchability were recorded in KT × KT (98.7% and 98.5% respectively) among the purebred, while KT × AA had the highest percentage fertilization and hatchability (98.4% and 97.3% respectively) in the reciprocal hybrids. The percentage fertilization and hatchability among the genetic crosses showed significant differences (p<0.05). The results deduced the essentiality of fish hybridization. Furthermore, crossing of broodstocks from different regions have showcased the contingency of acquiring fish seed of improved reproductive potentials in the reciprocal hybrids in the aspects of fecundity, fertilization, hatchability rate, testis quality and faster growth.
