Arginine vasopressin (AVP) plays an important role in the expression of aquaporin (AQP-2) in the collecting duct. The present study was undertaken to determine whether there is an AVP-independent regulation of AQP-2 gene expression in homozygous Brattleboro rats in which endogenous AVP is absent. Exogenous administration of 1-deamino-8-d-AVP produced an antidiuresis and expressed AQP-2 mRNA and AQP-2 protein in the renal medulla of the homozygous Brattleboro rats. Twelve hours of water deprivation produced severe dehydration in the homozygous Brattleboro rats, such that urinary osmolality increased from 200 to 649 mosmol/kgH2O. However, no increase in AQP-2 mRNA expression was observed after this dehydration, and the medullary tissue content and urinary excretion of AQP-2 also remained unchanged. Increases in AQP-2 mRNA expression and AQP-2 protein were evident in Long-Evans rats after 64 h of water deprivation, with a severity of dehydration almost equal to the 12-h dehydrated, homozygous Brattleboro rats. These results indicate the lack of an AVP-independent mechanism for upregulating AQP-2 mRNA expression in renal collecting duct cells.

This study evaluated the Nephrotoxic effect of water soluble fraction (WSF) of Bonny Light Crude Oil (BLCO). After preparation of the WSF and a range finding test, the Wistar albino rats were administered three concentrations (25%, 50% and 100%) of WSF of BLCO for 30 and 60days. Data from the study showed that Urea concentration increased significantly (p≤0.05) with increasing dose of BLCO ranging from 14.71 mg/dl in the control to 35.28 mg/dl in the 100% group after 30days and 14.28 mg/dl in the control to 41.08mg/dl in the 100% group after 60days, Creatinine concentration increased significantly (p≤0.05) from 0.22 mg/dl in the control to 0.82mg/dl in the 100% group after 60 days administration while electrolyte (Na, K, Cl) concentration increased significantly (p≤0.05) with increasing dose of BLCO after 60days administration. Histopathological examination of the kidney was characterized by partial partitioning of the glomerular tufts, obliteration of the Bowman’s capsule and distortion of the renal tubules. The findings in this research suggest that WSF of BLCO induced nephrotoxicity.

In aldosterone target tissues, 11β-hydroxysteroid dehydrogenase type 2 (11βHSD2) is coexpressed with mineralocorticoid receptors (MR) and protects the receptor from activation by glucocorticoids. Null mutations in the encoding gene, HSD11B2, cause apparent mineralocorticoid excess, in which hypertension is thought to reflect volume expansion secondary to sodium retention. Hsd11b2−/− mice are indeed hypertensive, but impaired natriuretic capacity is associated with significant volume contraction, suggestive of a urine concentrating defect. Water turnover and the urine concentrating response to a 24-h water deprivation challenge were therefore assessed in Hsd11b2 −/− mice and controls. Hsd11b2 −/− mice have a severe and progressive polyuric/polydipsic phenotype. In younger mice (∼2 mo of age), polyuria was associated with decreased abundance of aqp2 and aqp3 mRNA. The expression of other genes involved in water transport ( aqp4, slc14a2, and slc12a2) was not changed. The kidney was structurally normal, and the concentrating response to water deprivation was intact. In older Hsd11b2 −/− mice (>6 mo), polyuria was associated with a severe atrophy of the renal medulla and downregulation of aqp2, aqp3, aqp4, slc14a2, and slc12a2. The concentrating response to water deprivation was impaired, and the natriuretic effect of the loop diuretic bumetanide was lost. In older Hsd11b2 −/− mice, the V2 receptor agonist desmopressin did not restore full urine concentrating capacity. We find that Hsd11b2 −/− mice develop nephrogenic diabetes insipidus. Gross changes to renal structure are observed, but these were probably secondary to sustained polyuria, rather than of developmental origin.

Drive level affected reversal rather than non-reversal-shift learning during initial shift-discrimination trials. Animals under high water deprivation during the original simultaneous discrimination and reversal-shift discrimination made more initial (first trial-block) errors during reversal-shift than animals that were maintained on moderate deprivation during either or both discrimination tasks.
