An attempt was made to standardize the appropriate concentration of different growth regulators for successful in vitro growth of different explants (leaf, node and internode) of Aloe vera L. Results demonstrated best in vitro growth in leaf explants in MS medium supplemented with BAP (1.0 mg/l) and NAA (1.0 mg/l) at 26 ± 2ºC) with pH 5.70 using agar solidified medium and 16 hrs photoperiod.

Agrobacterium tumefaciens has been widely used in plant genetic transformation. Hormone-encoding genes residing in the T-DNA region have been removed, resulting in disarmed Agrobacterium strains that are used in various transformation experiments. Nopaline Agrobacterium strains, however, carry another hormone gene, trans-zeatin synthesizing (tzs), that codes for trans-zeatin in the virulence region of the tumor-inducing plasmids. We investigated the activity and function of the tzs gene of a nopaline Agrobacterium sp. strain GV3101 in plant in vitro regeneration. Leaf explants of tobacco and Nicotiana benthamiana co-cultured with strain GV3101 exhibited active shoot regeneration in media without added plant growth regulators. On medium without plant growth regulators, transgenic shoots were also induced from explants co-cultured with GV3101 containing a binary vector. Enzyme-linked immunosorbent assay showed that cell-free extracts of Agrobacterium sp. strain GV3101 culture contained the trans-zeatin at 860 ng/liter. Polymerase chain reaction using tzs-specific primers showed that the tzs gene was present in strain GV3101 but not in other Agrobacterium strains. The study showed that the tzs gene in GV3101 was actively expressed, and that trans-zeatin produced in the Agrobacterium strain can promote plant shoot regeneration.

Melia azedarach L. is one of the important plants because it’s a good source of natural compounds thathave insecticide and antimicrobial effect. The main aim of this research is to investigate the effect of explants source and plant growth regulators on in vitro callus induction and regeneration of organs from it. Callus was induced from nodes, internodes from one-year-old seedlings and seeds of Melia plant by culturing them on MS medium supplemented with α-naphthalene acetic acid (NAA) 0.0, 0.1, 0.2, 0.3, or 0.4 mg/L and 6- benzyl adenine (BA) 0.0, 1.0, 2.0, 3.0, or 4.0 mg/L, then shoot regeneration from callus was occurred. Results showed that there was a different response from explants towards callus induction and adventitious shoots formation according to plant growth regulators combination. Seeds gave superior percentage for callus induction 24.4% compared with node and internode 15.6, 12.8% respectively. Combination of 0.3 mg/L NAA + 3.0 mg/L BA was the best for callus induction in all explants 86.6% . Shoot regeneration was achieved in 0.3 mg/L NAA + 4.0 mg/L BA and 0.4 mg/L NAA + 4.0 mg/L BA for callus from seeds and internodes respectively, while the combination 0.3 mg/L NAA + 3.0 mg/L BA or 0.4 mg/L NAA + 3.0 mg/L BA was the best for node callus. The shoots were rooted well in MS + 0.25 mg/L NAA . Rooted plantlets were acclimatized in small plastic pots filled with peat moss: river soil (1 :1 v/v ), then transferred to the soil.

Dwarf hygro (Hygrophila polysperma) is an ornamental aquatic plant that changes its leaf colours to pinkish in high light. It is listed as a medicinal plant in medicinal plant lists of Indian states of West Bengal and Karnataka. It is also used as a screening tool for toxicities and a bioindicator to detect and control algae. The study reportedin vitroadventitious shoot regeneration from leaf explants cultured on MS medium containing 0.10–1.60 mg/L Kin/TDZ with or without 0.10 mg/L IBA and 500 mg/L Amoklavin to eradicate endogenic bacterial contamination. Direct adventitious shoot regeneration started within one week from both culture mediums followed by late callus induction which was more prominent on TDZ containing media compared to Kin containing media. Addition of 0.10 mg/L IBA with both Kin and TDZ increased shoot regeneration frequency, mean number of shoots per explant, and mean shoot length. Maximum number of 16.33 and 20.55 shoots per explant was obtained on MS medium containing0.80+0.10 mg/L Kin-IBA and0.10+0.10 mg/L TDZ-IBA, respectively. Regenerated shoots were rooted on MS medium containing 0.20–1.00 mg/L IBA followed by successfull acclimatization in aquariums. Regenerated plantlets were also tested in jars containing distilled water that showed the pH 6–9 for the best plant growth and development.
