Anthocyanin composition and nutritional properties of a blue-grained wheat-Triticum boeoticum substitution line

Durum wheat selection 56-1 was crossed with an accession of Triticum boeoticum Boiss.; and a F2 plant had 13 pairs plus 2 univalents (13″ + 2′) at metaphase I. In subsequent generations, plants with the chromosome configuration 14″ + 1′ were selected. Plants with 14″ + 1′ produced two types of seed. Approximately 78% of the seeds had purple pericarp and the rest were amber. Purple seeds failed to germinate unless they were soaked overnight in a one mg/l solution of gibberellic acid (GA3). The seeds with purple pericarp had 28 chromosomes including a pair of T. boeoticum chromosomes which were substituted for a pair of 4B chromosomes from durum wheat. The disomic-substitution plants were dwarfs and male-sterile. Amber seeds had the chromosome constitution 13″ + X″ + 4B′ (X = the T. boeoticum chromosome) and produced male-fertile, normal height plants. Chromosome pairing relationships indicated that the T. boeoticum chromosome had no more homology with wheat chromosomes 4A, 4B, and 4D than they had with each other. The T. boeoticum chromosome compensated for the loss of durum chromosome 4B except for a locus affecting GA.

Triticum boeoticum Boiss (AbAb, 2n = 2x = 14) is one of the sources of the blue grain trait controlled by blue aleurone layer 2 (Ba2). However, the underlying genes have not been cloned. In this study, a transcriptomic comparison between a blue-grained wheat-T. boeoticum substitution line and its wheat parent identified 41 unigenes related to anthocyanin biosynthesis and 29 unigenes related to transport. The bHLH transcription factor gene TbMYC4A showed a higher expression level in the blue-grained substitution line. TbMYC4A contained the three characteristic bHLH transcription factor domains (bHLH-MYC_N, HLH and ACT-like) and clustered with genes identified from other wheat lines with the blue grain trait derived from other Triticeae species. TbMYC4A overexpression confirmed that it was a functional bHLH transcription factor. The analysis of a TbMYC4A-specific marker showed that the gene was also present in T. boeoticum and T. monococcum with blue aleurone but absent in other Triticeae materials with white aleurone. These results indicate that TbMYC4A is a candidate gene of Ba2 controlling the blue aleurone trait. The isolation of TbMYC4A is helpful for further clarifying the genetic mechanism of the blue aleurone trait and is of great significance for breeding blue-grained wheat varieties.

Abstract:  Manabí and Ecuadorian population in general are consumers of tomato paste in different forms, so that producers and industrial product processing always looking to have a high quality  and  good  prices  for  the  benefit  of its  customers.  This research based on the results obtained, it leaves a number of very valuable conclusions for determining the carrot pulp will have a positive impact on the physical, chemical and organoleptic, when combined with the tomato paste features, without diminishing or provide a look that could adversely affect its commercial presentation to the client. The objective of this research was to get the development of a tomato paste mixed with carrot paste, with the purpose of obtaining an improved nutritional properties (fiber and beta-carotene) for the benefit of the consumer.
