Factors influencing the accuracy of early pregnancy detection in cattle by real-time ultrasound scanning of the uterus

Hemorrhage into areas of periventricular leukomalacia may occur and range in size from microscopic to massive. Hitherto, the definitive diagnosis has been made only at autopsy. A case is described in which the diagnosis of hemorrhagic periventricular leukomalacia was made antemortem by real-time ultrasound scanning and confirmed at autopsy. Periventricular/intraventricular hemorrhage, a more common hemorrhagic lesion, may extend into periventricular white matter in a location ventral and medial to hemorrhagic periventricular leukomalacia. Real-time ultrasound scanning is a safe and reliable means of defining the topography of these two hemorrhagic lesions and, thereby, of suggesting the correct diagnosis.

Abstract Charolais x Angus steers (n = 80) were evaluated across a 378-day feeding period in response to administration of a growth promoting implant containing trenbolone acetate (TBA) and estradiol (E2) using A-mode and B-mode ultrasound. Steers were scanned at 42-d intervals (0, 42, 84, 126, 168, 210, 252, 294, 336) during 10 separate ultrasound scanning sessions 24-h prior to harvest. Ultrasound B-mode rump fat images (BRFU) were obtained by a certified Ultrasound Guidelines Council (UGC) technician using an ALOKA 500V console equipped with a 17.2 cm carcass probe. Ultrasound A-mode images (ARFU) were obtained by a non-UGC certified technician using a RENCO Lean Meater. Images of BRFU were analyzed by the CUP lab whereas ARFU image results were determined chute side by the scanning technician. Carcasses were dissected into total lean, fat and bone post-harvest. Rump fat thickness ultrasound measurements were compared to carcass traits used to determine carcass value. Carcass endpoints included 12th–rib fat thickness (FTC), 12th rib-eye area (REAC), marbling score (MARB), and total percentage of carcass fat (PCF). The correlation between BRFU and ARFU was 0.70 (P < 0.01). Positive correlations (P < 0.01) between ultrasound rump fat measures taken the day before harvest and carcass measurements indicated moderate relationships between BRFU and FTC (r=0.80); BRFU and REAC (r=0.66); BRFU and MARB (r=0.68); BRFU and PCF (r=0.75); ARFU and FTC (r=0.69); ARFU and REAC (r=0.76); ARFU and MARB (r=0.79); and ARFU and PCF (r=0.80). These results indicate A- and B-mode ultrasound measures of rump fat may be adequate measures for estimation of total percent carcass fat.

To implant and establish the connections that are vital for further development, the early embryo must attach to and then breech the barrier posed by the epithelium of the maternal tract. Expression of adhesion and anti-adhesion molecules in the luminal epithelium of the endometrium are thought to fluctuate in a temporal pattern to 'frame' the implantation site, with their expression regulated by endocrine and paracrine factors. Anti-adhesion molecules, such as members of the mucin family, provide a barrier to implantation in sites or at times unsuitable for embryo development. Expression of adhesion molecules, or specific integrins, are thought to aid in the adhesion of the embryo, allowing it to induce changes in the underlying tissue promoting embryo invasion and pregnancy. The aim of this study was to quantitate the expression of mRNA encoding the integrins αυ, α4 and β3 and MUC1 and MUC4 from Day 0 (oestrous) to Day 4 of pregnancy (implantation) using quantitative real time RT-PCR. Uterine tissues were collected at oestrous and at Days 1, 2, 3 and 4 of pregnancy (Day 1 corresponding to the presence of a vaginal plug), total RNA was extracted, DNAse treated, reverse transcribed into cDNA, and quantified by real-time PCR using SYBR Green chemistry. All specific primers were designed using GenBank sequences and data were normalised to β-actin mRNA expression. Expression of MUC1 and MUC4 mRNAs was dramatically reduced, with mean values 20-fold and 100-fold less than at oestrous respectively, by Day 4 of pregnancy. In contrast, expression of mRNAs encoding integrins αυ, α4 and β3 was detected throughout early pregnancy. These data demonstrate that adhesion and anti-adhesion molecules are differentially expressed in the murine uterus during early pregnancy and may be key mediators in embryo implantation, promoting attachment of the embryo to the luminal epithelium in an environment conducive to embryo growth and development. Technological advancements have enabled people to communicate with each other at the convenience of their homes and in real time. This change, however, also means the changes in human behaviors, such as computer-mediated communication, can be shaped by the richness of the media that people can use to convey their thoughts and opinions. The existence of different chatting applications has fulfilled the needs of human beings to be connected and to interact with each other, and the interactions that take place can be in the form of gossiping and spreading information that may not necessarily be accurate.
