AbstractTrans-10, cis-12 conjugated linoleic acid (CLA) decreases milk fat synthesis in lactating sows and involves, at least in part, the down-regulation of lipogenic genes. The objective was to evaluate the effect of CLA on milk composition and lipogenic gene expression. Twenty multiparous sows were randomly assigned to one of the two treatments for 18 d (from day 7 to day 25 of lactation): (1) control (no CLA added) and (2) 1 % of CLA mixed into the ration. CLA treatment decreased milk fat and protein content by 20 % (P = 0·004) and 11 % (P = 0·0001), respectively. However, piglet weight did not differ between treatments (P = 0·60). Dietary CLA increased the concentration of SFA in milk fat by 16 % (P < 0·0001) and decreased MUFA by 17·6 % (P < 0·0001). In the mammary gland, CLA reduced gene expression of acetyl-CoA carboxylase-α by 37 % (P = 0·003), fatty acid synthase by 64 % (P = 0·002), stearoyl-CoA desaturase 1 by 52 % (P = 0·003), lipoprotein lipase by 26 % (P = 0·03), acyl glycerol phosphate acyltransferase 6 by 15 % (P = 0·02) and diacylglycerol acyltransferase 1 by 27 % (P = 0·02), whereas the expression of fatty acid binding protein 3 was not altered by CLA treatment (P = 0·09). Mammary expression of casein-β and α-lactalbumin was reduced by CLA by 68 % (P = 0·0004) and 62 % (P = 0·005), respectively. Additionally, CLA had no effect on the expression of lipogenic genes evaluated in adipose tissue. In summary, CLA reduced milk fat content without negatively affecting litter performance and it affected mammary expression of genes involved in all lipogenic pathways studied.

AbstractTwenty pregnant Comisana ewes were divided into two groups of 10. One group was allowed to graze a vetch pasture (grass). The second group of animals was housed collectively in a pen and was given hay and concentrates (concentrate). After lambing, all the ewes were allowed to stay with the respective lambs between 18:00 h and 07:00 h of the following day in two different pens. Therefore all the lambs were raised exclusively on maternal milk. The lambs were slaughtered at 38 days of age. Milk and lamb meat (longissimus dorsi muscle) fatty acids were analysed. Ewes on grass produced milk with a lower (P < 0·001) proportion of saturated fatty acids and with a higher proportion of both monounsaturated (P < 0·05) and polyunsaturated fatty acids (P < 0·01) than ewes given concentrates. Trans-vaccenic acid was significantly higher (P < 0·001) in milk from grass-fed animals compared with ewes given concentrates. Linoleic acid (C18: 2 n-6) tended to be higher (P = 0·06) in milk from ewes on concentrates while linolenic acid (C18: 3 n-3) was significantly higher (P < 0·001) in milk from animals grazing pasture. Conjugated linoleic acid (cis-9, trans-11 C18: 2) was almost double in milk from grass-fed ewes compared with animals given concentrates (P < 0·001). Regarding lamb tissue, trans-vaccenic acid (C18: 1 trans-11) was higher (P = 0·01) in the fat from lambs raised by grazing ewes. Linoleic acid (C18: 2 n-6) was at higher concentration (P < 0·001) in the fat from lambs raised by ewes given concentrates. Linolenic acid (C18: 3 n-3) was increased three-fold (P < 0·001) in the fat of lambs from the grass group compared with lambs suckled by ewes given concentrates. The isomer cis-9, trans-11 of conjugated linoleic acid was present at double concentration (P < 0·001) in the fat from animals raised by grazing ewes. Eicosapentaenoic (C20: 5 n-3; EPA) and docosaesaenoic (C22: 6 n-3; DHA) acids were higher (respectively P < 0·001 and P = 0·01) in the intramuscular fat from lambs from the grass group compared with animals from the concentrate group. The n-6/n-3 ratio was lower (P < 0·001) in the meat from lambs raised by grazing ewes. Overall this trial showed that ewe feeding system strongly affects intramuscular fatty acids even in lambs raised exclusively on maternal milk.

Fatty acid profiling, meat and organoleptic quality assessments were conducted on 160 carcasses of crossbred steers born into one of two calving systems and later assigned to one of two postweaning feeding systems. The steers were weaned from either an early calving or late calving system and later assigned to either a rapid-gain feeding (RF) or a slow-gain feeding (SF) system. The RF steers received a silage–hay diet during the backgrounding period prior to finishing, while the SF steers received a hay diet at backgrounding and then grazed alfalfa–meadow bromegrass pasture and annual cereal swaths prior to finishing. All treatment groups received a conventional diet during finishing until the steers attained a target backfat thickness or body weight. Fatty acid analyses were conducted on longissimus dorsi muscle (LDM) and subcutaneous fat samples. Other analyses included LDM composition, tenderness and taste panel evaluations. Total saturated fatty acid was greater (P<0.02) in samples from RF steers, while total monounsaturated fatty acid was greater (P<0.01) in SF steers. The SF steers had greater (P<0.01) conjugated linoleic acid concentration. There was no main or interaction effect (P>0.05) on beef aroma, flavour and tenderness but the SF steaks had lower (P=0.02) cooking losses than RF steaks. The SF strategy has the potential to create a value chain that would lead to finished steers with higher backfat omega-3, conjugated linoleic acid and trans vaccenic acid and less cooking moisture losses.
