Magnesium chelatase (Mg-chelatase) inserts magnesium into protoporphyrin during the biosynthesis of chlorophyll and bacteriochlorophyll. Enzyme activity is reconstituted by forming two separate preactivated complexes consisting of a GUN4/ChlH/protoporphyrin IX substrate complex and a ChlI/ChlD enzyme ‘motor’ complex. Formation of the ChlI/ChlD complex in both Chlamydomonas reinhardtii and Oryza sativa is accompanied by phosphorylation of ChlD by ChlI, but the orthologous protein complex from Rhodobacter capsulatus, BchI/BchD, gives no detectable phosphorylation of BchD. Phosphorylation produces a 1-N-phospho-histidine within ChlD. Proteomic analysis indicates that phosphorylation occurs at a conserved His residue in the C-terminal integrin I domain of ChlD. Comparative analysis of the ChlD phosphorylation with enzyme activities of various ChlI/ChlD complexes correlates the phosphorylation by ChlI2 with stimulation of Mg-chelatase activity. Mutation of the H641 of CrChlD to E641 prevents both phosphorylation and stimulation of Mg-chelatase activity, confirming that phosphorylation at H641 stimulates Mg-chelatase. The properties of ChlI2 compared with ChlI1 of Chlamydomonas and with ChlI of Oryza, shows that ChlI2 has a regulatory role in Chlamydomonas.

Abstract The bioaccumulation of trace elements (TEs) in crops consumed by humans can lead to a lower food production due to photosynthetic damages and several diseases in humans, but decreasing soil acidity could mitigate these problems by decreasing TEs bioavailability. Thus, we evaluate the effect of increasing soil base saturation (BS%) on photosynthesis, growth and bioaccumulation of barium (Ba), cadmium (Cd), copper (Cu), nickel (Ni) or zinc (Zn) in lettuce (Lactuca sativa L.), rice (Oryza sativa L.), sunflower (Helianthus annuus L.) and tomato (Solanum lycopersicum L.) grown in sandy Entisol. The crops were grown in uncontaminated or contaminated Entisol, under two BS% ratios: 30% for all crops or 50% for rice and 70% for lettuce, sunflower and tomato. The photosynthesis-related parameters varied depending on the metal and crop, but in general, increasing BS% did not attenuate photosynthetic damages induced by Ba, Cd, Cu, Ni and Zn in the crops. There was no strong correlation between the photosynthetic parameters measured and biomass production, which suggest us that the suppression on biomass induced by Ba, Cd, Cu, Ni or Zn is related to other metabolic disorders besides the impairment on CO2 assimilation or chlorophyll synthesis in the crops assayed, with exception of Ni and Zn in lettuce. In conclusion, increasing BS% was not consistent in decreasing Ba, Cd, Cu, Ni and Zn accumulation in the edible parts of lettuce, rice, sunflower and tomato grown in the sandy soil, which probably is related to the low capacity of this soil in control TEs bioavailability.

Abstract Background Strawberry (Fragaria) is regarded as a model plant for both Rosaceae and non-climacteric fruit ripening. Although much progress has been made in identification of gene function using stable and transient genetic transformation systems in strawberry, the limitation is, more or less, are present. To this end, development of a rapid, efficient, and stable transformation system is required for strawberry research and breeding. Results Here, using diploid Hawaii-4 (Fragaria vesca) seeds and a reporter gene of CHLH (the H subunit of magnesium chelatase magnesium chelatase) key to chlorophyll synthesis, we first develop a rapid, efficient, and stable infected system by the Agrobacterium-mediated seed infection to silence the reporter gene, reaching an infection frequency with 28.3% through a series of optimization elements, including seed full imbibition and initial germination, shaking infection for 24 h, dark cultivation on MS medium for 3 d at 24 ℃, light culture on MS-Tim medium for 1 week at 24 ℃, and vector construction tagged with fluorescence label. Taken together, radicle-emergence germination seeds, appropriate Agrobacterium concentration and infection time are critical for successful infection, finally obtaining the infected kanamycin-resistant seedlings of T1 generation by infected wild seeds within 1 month and T2 generation-infected plants within 4 months. Conclusions The Agrobacterium-mediated infection of germinating seeds (AMTGS) in diploid strawberry (F. vesca) is first established, providing a useful tool for gene function identification and improved agronomic traits in strawberry.

Keragaan morfologi dan kandungan antosianin padi beras merah (Oryza sativa L.) pada Kecamatan Munte dan Kecamatan Payung di Kabupaten Karo. Penelitian ini bertujuan untuk mendapatkan informasi morfologi dan kandungan antosianin padi beras merah (Oryza sativa L.). Penelitian ini dimulai dari Juni 2017dan selesai pada Januari 2018 di kecamatan Munte dan Kecamatan Payung Kabupaten Karo.Metode survei deskriptif menggunakan panduan International Rice Research Institute (IRRI). Teknik penentuan lokasi secara sengaja dan pengamblan sampel secara kebetulan.Uji kandungan antosianin menggunakan metode analitik dengan menghomogenkan sampel. Hasil eksplorasi didapatkan 72 genotipe yang dibagi menjadi tiga lokasi lahan. Berdasaran uji kandungan antosianin didapatkan lahan A memiliki kandungan tertinggi yaitu 0,5 mg/100 g dan terendah yaitu lahan B 0,08 mg/100 g.

Experiments were carried out to assess some insecticide modules against major insect pests of rice. Each module consists of a basal application of carbofuran 3G @ 1 kg a.i ha-1 at 20 DAT and Rynaxypyr 20 SC @ 30 g a.i ha-1 at 45 DAT except untreated control. All modules differ with each other only in third treatment which was applied in 65 DAT. The third treatment includes: Imidacloprid 17.8 SL @ 27 g a.i ha-1, Pymetrozine 50 WG @ 150 g a.i ha-1, Triflumezopyrim 106 SC @ 27 g a.i ha-1, Buprofezin 25 SC @ 250 g a.i ha-1; Glamore (Imidacloprid 40+Ethiprole 40% w/w) 80 WG @ 100 g a.i. ha-1, Thiacloprid 24 SC @ 60 g a.i ha-1, Azadirachtin 0.03 EC @ 8 g a.i ha-1, Dinotefuran 20 SG@ 40 g a.i ha-1 and untreated control. All the treated plots recorded significantly lower percent of dead heart, white ear- head caused by stem borer and silver shoot caused by gall midge. Module with Pymetrozine 50 WG @ 150 g a.i ha-1 treated plot recorded significantly higher per cent reduction of plant hoppers (>80% over untreated control) and produced higher grain yield (50.75 qha-1) than the other modules. Among the different treated modules the maximum number of spiders was found in Azadirachtin 0.03 EC @ 8 g a.i ha-1 treated module plot followed by other treatments.

Tujuan dari penelitian ini adalah untuk mengetahui pengaruh aplikasi abu sekam terhadap kandungan prolin dan silikat daun serta kualitas hasil yaitu protein dan amilosa biji padi gogo dan hubungan korelasi antar keempat komponen tersebut pada kondisi pertanaman 80 persen kapasitas lapang pada skala pot. Penelitian dilakukan di polibag dalam screen house Fakultas Pertanian Unsoed dengan menggunakan rancangan acak kelompok pola faktorial dengan faktor varietas (Situ patenggang, Limboto, Towuti, Batutegi dan Aek sibundong) dan faktor abu sekam (0, 2, 4, 6 t/ha), diulang tiga kali. Hasil penelitian menunjukan bahwa pemberian abu sekam dosis 2−6 t/ha mampu meningkatkan kandungan silikat daun antara 18,49−29,43% dan menurunkan kandungan amilosa biji pada lima varietas sekitar 4,19−6,92%. Pemberian abu sekam dosis 2−6 t/ha mampu meningkatkan kandungan prolin daun antara 27,56−70,63% dan protein biji antara 2,35−16,71%. Antarvarietas menunjukan bahwa kandungan prolin tertinggi dihasilkan oleh varietas Batu tegi 18,58 persen dan protein biji pada varietas Situ patenggang 9,55%. Terdapat korelasi antar karakter fisiologis yaitu antara silikat-prolin (0,62) dan kandungan protein-amilosa biji (-0,78).
