AbstractWe report a high-throughput technique for characterising the motility of spermatozoa using differential dynamic microscopy. A large field of view movie (~ 10mm2) records thousands of cells (e.g. ≈ 5000 cells even at a low cell density of 20 × 106 cells/ml) at once and yields averaged measurements of the mean (υ) and standard deviation (σ) of the swimming speed, a head oscillation amplitude (A0) and frequency (f0), and the fraction of motile spermatozoa (α). Interestingly, the measurement of α relies on the swimming spermatozoa enhancing the motion of the non-swimming population. We demonstrate the ease and rapidity of our method by performing on-farm characterisation of bull spermatozoa motility, and validate the technique by comparing laboratory measurements with tracking. Our results confirm the long-standing theoretical prediction that for swimming spermatozoa.

Objective lens is the core part of a collimator, its quality has a direct impact on the collimators quality. Due to the small field of view and low image quality requirement, general collimator objective lens is difficult to achieve the requirement of testing an aerial camera s performance. So designing a large field of view, apochromatic collimator objective lens has an important practical significance. In this paper, the major qualifications were first proposed to design the collimator objective lens,according to which the initial configuration was confirmed. Then we used the ZEMAX optical design software to optimize the initial structure and normalized the optimized structural parameters. Finally the image quality of collimator objective lens was evaluated to make it achieve the requirement of qualifications.

A wide holographic field of view (up to 15 μm in the Hitachi-HF2000) is achieved in a TEM by switching off the objective lens and imaging the sample by the first intermediate lens. Fig.1 shows the corresponding ray diagram for low magnification image plane off-axis holography. A coherent electron beam modulated by the sample in its amplitude and its phase is superimposed on a plane reference wave by a negatively biased Möllenstedt-type biprism.Our holograms are acquired utilizing a Hitachi HF-2000 field emission electron microscope at 200 kV. Essential for holography are a field emission gun and an electron biprism. At low magnification, the excitation of each lens must be appropriately adjusted by the free lens control mode of the microscope. The holograms are acquired by a 1024 by 1024 slow-scan CCD-camera and processed by the “Holoworks” software. The hologram fringes indicate positively and negatively charged areas in a sample by the direction of the fringe bending (Fig.2).
