1. The keratinocytes in human epidermis are constantly turned over and replaced by a population of stem cells located in the basal epidermal layer. Until recently there were no markers allowing the isolation of viable epidermal stem cells. However, it has now been shown that epidermal stem cells can be isolated both in vitro and direct from the epidermis as they express high levels of functional β1 integrin family receptors for extracellular matrix proteins. 2. The evidence for integrins as stem cell markers and the insights that have been gained into stem cell behaviour are reviewed.

The procedure for aspiration of bone marrow from the femur of patients undergoing total knee arthroplasty (TKA) or total hip arthroplasty (THA) may vary from an OR (operating room) to OR based on the surgeon’s skill and may lead to varied extent of clotting of the marrow and this, in turn, presents difficulty in the isolation of mesenchymal stem cells (MSCs) from such clotted bone marrow. We present a simple detailed protocol for aspirating bone marrow from such patients, isolation, and characterization of MSCs from the aspirated bone marrow specimens and show that the bone marrow presented no clotting or exhibited minimal clotting. This represents an economical source and convenient source of MSCs from bone marrow for use in regenerative medicine. Also, we presented the detailed protocol and showed that the MSCs derived from such bone marrow specimens exhibited MSCs characteristics and generated micromass cartilages, the recipe for regenerative medicine for osteoarthritis. The protocols we presented can be used as standard operating procedures (SOPs) by researchers and clinicians.

Although the European rabbit is an "endangered" species and a notorious biological model, the analysis and comparative characterization of new tissue sources of rabbit mesenchymal stem cells (rMSCs) has not been well studied. Here we report for the first time the isolation and characterization of rMSCs derived from an animal belonging to a natural rabbit population within the species native region. New rMSC lines were isolated from different tissues: oral mucosa (rOM-MSC), dermal skin (rDS-MSC), subcutaneous adipose tissue (rSCA-MSC), ovarian adipose tissue (rOA-MSC), oviduct (rO-MSC), and mammary gland (rMG­MSC). The six rMSC lines showed plastic adhesion with fibroblast-like morphology and were all shown to be positive for CD44 and CD29 expression (characteristic markers of MSCs), and negative for CD34 or CD45 expression. In terms of pluripotency features, all rMSC lines expressed NANOG, OCT4, and SOX2. Furthermore, all rMSC lines cultured under osteogenic, chondrogenic, and adipogenic conditions showed differentiation capacity. In conclusion, this study describes the isolation and characterization of new rabbit cell lines from different tissue origins, with a clear mesenchymal pattern. We show that rMSC do not exhibit differences in terms of morphological features, expression of the cell surface, and intracellular markers of pluripotency and in vitro differentiation capacities, attributable to their tissue of origin.

Τα καρκινικά βλαστικά κύτταρα (ΚΒΚ) αποτελούν έναν υποπληθυσμό των καρκινικών κυττάρων με ιδιότητες αυτο-ανανέωσης και διαφοροποίησης, που θεωρείται ότι ευθύνονται για την ανάπτυξη του όγκου και την υποτροπή μετά από θεραπεία. Η ικανότητα των ΚΒΚ να επιβιώνουν μετά από εφαρμογή συμβατικών μορφών αντικαρκινικής θεραπείας συνέβαλε στην προσπάθεια ανάπτυξης νέων θεραπευτικών στρατηγικών που στοχεύουν τα συγκεκριμένα κύτταρα. Πρόσφατες μελέτες, υποδεικνύουν ότι οι μοναδικές ιδιότητες των ΚΒΚ φαίνεται να υπόκεινται σε επιγενετική ρύθμιση. Η LSD1 / KDM1A, μία απομεθυλάση ιστονών, ρυθμίζει τις βλαστικές ιδιότητες των κυττάρων, ενώ επίσης υπερεκφράζεται σε διαφορετικούς τύπους καρκίνου συμπεριλαμβανομένου και του καρκίνου του μαστού. Στην παρούσα εργασία, στόχος μας ήταν να διερευνήσουμε τον ρόλο της LSD1 στην χημειο-ανθεκτικότητα των ΚΒΚ του μαστού. Πειράματα ανοσοϊστοχημείας σε δείγματα ασθενών με καρκίνο του μαστού επιβεβαίωσαν τη σύνδεση της έκφρασης της LSD1 με λιγότερο διαφοροποιημένους όγκους του μαστού. Επιπλέον, ο γονιδιακός χειρισμός της έκφρασης της LSD1 με διαφορετικές πειραματικές προσεγγίσεις, συσχέτισε την απομεθυλάση με την χημειο-ανθεκτικότητα των καρκινικών κυττάρων του μαστού. Επιπροσθέτως, έχοντας εγκαθιδρύσει και χαρακτηρίσει εκτενώς ένα in vitro σύστημα καλλιέργειας σφαιρικών δομών εμπλουτισμένο σε καρκινικά βλαστικά κύτταρα του μαστού, πειράματα γονιδιακής αποσιώπησης και υπερέκφρασης, έδειξαν ότι το ένζυμο ρυθμίζει τις βλαστικές ιδιότητες του υπο-πληθυσμού των καρκινικών βλαστικών κυττάρων του μαστού CD44+/CD24-/low.. Επιπλέον, η φαρμακολογική αναστολή της LSD1 οδήγησε σε μείωση των ΚΒΚ και εξασθένιση του δυναμικού βλαστικότητας in vitro, ενώ ήταν σε θέση να οδηγήσει σε συρρίκνωση των όγκων και να μειώσει τον υποπληθυσμό ΚΒΚ in vivo. Εν κατακλείδι, η ανθεκτικότητα στη χημειοθεραπεία των καρκινικών βλαστικών κυττάρων του μαστού επιβεβαιώθηκε στο σύστημά μας μέσω των αντίστοιχων πειραματικών προσεγγίσεων. Ωστόσο, η χορήγηση ενός αναστολέα της LSD1 κατέστησε τα καρκινικά βλαστικά κύτταρα του μαστού πιο ευαίσθητα στις ευρέως χρησιμοποιούμενες θεραπευτικές μεθόδους, όπως δείχνουν οι ποσοτικοί προσδιορισμοί του ρυθμού πολλαπλασιασμού των κυττάρων και σχηματισμού σφαιρικών δομών. Πειράματα με ξενομοσχεύματα σε επίμυες βρίσκονται σε εξέλιξη για να επιβεβαιωθεί ότι η στόχευση της LSD1 σε συνδυασμό με χημειοθεραπευτικούς παράγοντες αναστέλλει αποτελεσματικότερα την ανάπτυξη όγκων. Τα παραπάνω δεδομένα υποδηλώνουν έντονα ότι η LSD1 αποτελεί έναν πιθανό θεραπευτικό στόχο στα καρκινικά βλαστικά κύτταρα του μαστού, καθώς βρέθηκε ότι ρυθμίζει τις βλαστικές ιδιότητες των κυττάρων αυτών και έτσι προσδίδει χημειο-ανθεκτικές ικανότητες.
