Acaricidal Activity of Pla.nt Essential Oils against Tyrophagus putrescentiae (Acari: Acaridae)

Baccharis dracunculifolia, native to Brazil and the main source of “green propolis”, has been reported with several biological activities, and may be a source of bovine tick control substituting synthetic acaricides. Objective: to evaluate the in vitro and ex situ acaricidal activity of B. dracunculifolia leaf and flower essential oils against Rhipicephalus microplus. Methodology: the essential oils were extracted by hydrodistillation and analyzed by a gas chromatography coupled to mass spectrometry; the acaricidal activity of the essential oil was evaluated in vitro against adult females and against the egg hatchability; moreover, the acaricidal activity against tick larvae was evaluated in vitro and ex situ. Results: the major class of the essential oils was oxygenated sesquiterpene (55.1% leaves 50.4% flowers) and the main compounds were (21.5% leaves; 20.6% flowers) and spathulenol (21.8% leaves; 20.3% flowers). The essential oil at 500 mg/mL was effective to control egg hatchability with a reduction of egg laying capacity and decrease of number of adult ticks and larvae. The larvicidal activity of the essential oil had LC99.9 from 35 to 37 mg/mL by probit analysis, and the essential oil from 11 to 14 mg/mL presented 85 to 95% of treatment efficiency in the ex situ test. Conclusion: B. dracunculifolia leaf and flower essential oils are stable and have application potential to control bovine ticks.

The present study aimed to investigate the acaricidal properties of six essential oils. They were extracted from some plant species (Lamiaceae and Myrtaceae) using the technique of hydrodistillation with the Clevenger apparatus. The chemical compositions of the essential oils under study were determined by gas chromatography–mass spectrometer (GC-MS). An Adult Immersion Test (AIT) and a Larval Immersion Test (LIT) were used to evaluate the acaricidal activity of these essential oils against the adults and larvae of Hyalomma scupense. GC-MS analysis showed the major constituents of each essential oil: 25.49% of α-thujone (lavender); 46.82% of carvacrol (oregano); 78.78% of carvacrol (thyme); 40.27% of 1,8-cineole (blue gum); 17.45% of p-cymene (river red gum); and 26.96% of 1,8-cineole (rosemary). The biotests on the essential oils revealed that they inhibit the reproduction of H. scupense engorged females at a rate of 100 % with doses of 0.781 μl/ml of rosemary, 1.562 μl/ml of thyme, 3.125 μl/ml of lavender and oregano, and 6.250 μl/ml of blue gum and river red gum. After a treatment that lasted for 24 hours, essential oils showed a larvicidal activity with respective values of lethal concentrations (LC): LC50, LC90, and LC95 (0.058, 0.358, and 0.600 μl/ml for thyme; 0.108, 0.495, and 0.761 μl/ml for rosemary; 0.131, 0.982, and 1.740 μl/ml for oregano; 0.155, 2.387, and 5.183 μl/ml for blue gum; 0.207, 1.653, and 2.978 μl/ml for river red gum; and 0.253, 2.212, and 4.092 μl/ml for lavender). This is the first report on the acaricidal activity of these essential oils against H. scupense. The results obtained showed that the essential oils with chemotype carvacrol, 1,8-cineole, α-thujone, and p-cymene are highly acaricidal, and they can be used for ticks control. However, further studies on their toxicity in nontarget organisms are required.

Abstract The aim of this study was to evaluate the acaricidal activity of essential oils from three species of plants with intermediary concentrations of 1,8-cineole against the tick species Rhipicephalus (Boophilus) microplus. For this purpose, five serial concentrations (100.0, 50.0, 25.0, 12.5, 6.2 mg/mL) of essential oils from Mesosphaerum suaveolens (L.) Kuntze, Ocimum gratissimum L. and Alpinia zerumbet (Pers.) B. L. Burtt & R. M. Sm. were used on larval packet and adult immersion tests. The essential oils were analysed by gas chromatography-mass spectrometry (GC/MS) and gas chromatography-flame ionization detection (GC-FID), being detected 35.8, 24.7 and 24.0% of 1.8-cineol in the oils of M. suaveolens, O. gratissimum and A. zerumbet, respectively. The lethal concentration (LC 50) of each oil for larvae and engorged females was calculated through Probit analysis. All essential oils showed high efficacy (≥ 95.0%) on engorged females at the 100.0 mg/mL concentration. In regards to larvae, O. gratissimum (LC 50 = 11.9 mg/mL) was the most potent, followed by the A. zerumbet (LC50 = 19.7 mg/mL) and the M. suaveolens (LC50 = 51.6 mg/mL) essential oils. These results show that other compounds interfere with 1,8-cineole action.

AbstractAcaricidal activities and color alterations of 5-methylfurfural derived from Valeriana fauriei essential oil and its structural analogues against Dermatophagoides farinae, D. pteronyssinus, Haemaphysalis longicornis and Tyrophagus putrescentiae were evaluated in the present study. Based on the LD50 values of 5-methylfurfural and its analogues, 4,5-dimethylfurfural showed the highest acaricidal activity (LD50; 9.95, 9.91, and 7.12 μg/cm2), followed by 5-methylfurfural (11.87, 11.00, and 8.59 μg/cm2), furfural (12.94, 13.25, and 10.36 μg/cm2), and V. fauriei essential oil (15.15, 13.64, and 10.14 μg/cm2) against D. farinae, D. pteronyssinus and T. putrescentiae, respectively. However, all tested compounds did not show the acaricidal activities against H. longicornis. Interestingly, the color alterations of the mites and ticks were observed by furfural, 5-methylfurfural, and 4,5-dimethylfurfural from colorless to red brown during the acaricidal experiments. Furthermore, 4,5-dimethylfurfural which exhibited the highest acaricidal activity was formulated as nanoemulsion. The nanoemulsion of 4,5-dimethylfurfural showed higher acaricidal activity than it was emulsified in ethanol. The nanoemulsion was also found to show color changes of the mites and ticks from colorless to red brown. The results suggest that 5-methylfurfural and its analogues could be developed as an effective and easy-to-recognize acaricides to mites and ticks.
