Attenuation of Schistosoma mansoni cercariae with a molluscicide derived from Millettia thonningii

A chloroform extract of molluscicidal plant secondary compounds from the seeds of a West African legume Millettia thonningii was used to attenuate cercariae of Schistosoma mansoni prior to infection of NMRI female mice. Exposure of cercariae to 0·3, 0·6, 1·3, 2·5, 10 or 20 mg/l concentrations of extract for 30 mm, immediately before standardized infection, was associated with a concentration-dependent decline in worm establishment at 55 days post-infection. The mean numbers of adult worms established declined from about 17 worms/mouse with control cercariae and those exposed to 0·3 mg/l Millettia to 0·1 worms/mouse after 10 mg/l pre-treatment. Mice exposed to cercariae after 20 mg/l pre-treatment had no adult worms at 55 days post-infection. The activities of cercariae 30 mm after exposure to Millettia extract at concentrations up to 2·5 mg/l were similar to those of control larvae: none was immobile. Exposure to higher concentrations of Millettia progressively reduced swimming activity and increased the proportion of immobile cercariae. After pretreatment with 20 mg/l Millettia the majority of larvae were immobile. Levels of anti-parasite antibodies (estimated by ELISA assay using an adult worm-based antigen preparation) rose between 21 and 55 days p.i. with control cercariae and those pre-treated with 2·5 mg/l Millettia. After 5 mg/l pre-treatment, cercariae induced a reduced antibody rise, while mice exposed to cercariae after 10 mg/l pre-treatment showed no rise in anti-parasite antibody levels. These results are discussed in the context of protocols which could possibly use Millettia-attenuated cercariae to induce useful levels of protection in mice towards further cercarial challenge.

AbstractThe study describes methods developed for using video-imaging technology to record and measure the velocity of Schistosoma mansoni miracidia. The efficacy of the classical bioassay procedure (a qualitative behavioural assay) was compared with that of the new quantitative protocol, for assessing the sub-lethal impact of a larvicidal dichloromethane extract of the seeds of Millettia thonningii on miracidia. The new technique confirmed the efficacy of the classical bioassay for rapid determination of the lethal and sub-lethal impact of larvicides but also provided quantitative information on sub-lethal impacts on miracidial velocity and shape.

We propose a hypothesis for digestive constraints on the browsing and grazing options available to ruminants: that the diet-niche range (maximum and minimum grass intake) of a species is dependent upon its predisposition to stratified rumen contents, based on observations that this characteristic is a critical step towards enhanced fibre digestion and greater fluid throughput. We compare a physiological (heterogeneity of ingesta fluid content) and an anatomical (the intraruminal papillation pattern) measure with dietary evidence for a range of African and temperate species. Both measures are strongly related to the mean percentage of grass in species’ natural diets, as well as to the maximum and minimum levels of grass intake, respectively. The nature of these effects implies a stratification-level threshold, below which a species will not use a grass-based diet, but above which grass consumption can increase exponentially. However, above this threshold, a minimum percentage of grass in the diet is a prerequisite for optimal performance. We argue that this second constraint is crucial, as it depicts how a greater fluid throughput reduces potential for detoxification of plant secondary compounds, and therefore limits the maximum amount of browse a stratifying species will consume.

The vectorial capacity of Biomphalaria pfeifferi from Ndiangue, Senegal, was investigated with an allopatric isolate of Schistosoma mansoni from Nkolbisson, Cameroon. The snail infection rate after exposure to a single miracidium per snail (MD1) was 56·3%, and 91·6% for snails exposed to 5 miracidia per snail (MD5). The minimum pre-patent period was 21 days. The mean total cercarial production for the MD1 group was 18511 cercariae per snail, and 9757 cercariae for the MD5 group. The maximum production of cercariae for 1 day was 4892 observed in a snail from the MD1 group at day 43 post-infection. The mean longevity of snails was higher in group MD1 (88 days p.i.) than in group MD5 (65 days p.i.). The chronobiological emergence pattern revealed a circadian rhythm with one shedding peak at mid-day. Comparisons are made with the vectorial capacity of the sympatric combination of B. pfeifferi Senegal/S. mansoni Senegal.

Abstract Background We investigated frequency of reinfection with seasonal coronaviruses (HCoV) and serum antibody response following infection over 8 years in the Household Influenza Vaccine Evaluation cohort. Methods Households were followed annually for identification of acute respiratory illness with RT-PCR confirmed HCoV infection. Serum collected before and at two time points post infection were tested using a multiplex binding assay to quantify antibody to seasonal, SARS-CoV-1, and SARS-CoV-2 spike proteins and SARS-CoV-2 spike sub-domains and N protein. Results Of 3418 participants, 40% were followed for ≥3years. A total of 1004 HCoV infections were documented; 303 (30%) were reinfections of any HCoV type. The number of HCoV infections ranged from 1 to 13 per individual. The mean time to reinfection with the same type was estimated at 983 days for 229E, 578 days for HKU1, 615 days for OC43, and 711 days for NL63. Binding antibody levels to seasonal HCoVs were high, with little increase post-infection, and were maintained over time. Homologous, pre-infection antibody levels did not significantly correlate with odds of infection, and there was little cross response to SARS-CoV-2 proteins. Conclusions Reinfection with seasonal HCoVs is frequent. Binding anti-spike protein antibodies do not correlate with protection from seasonal HCoV infection.

AbstractThis study consisted of two experiments. In experiment 1, stress responses of sheep which were restrained either in a laparoscopy cradle or a roll-over cradle were compared. The results of this experiment indicated that restraint in roll-over cradle is less (P < 0·05) stressful than that in a laparoscopy cradle when assessed in terms of the elevation and duration of both the mean heart rate and plasma cortisol responses. Experiment 2 compared the stress responses of sheep subjected to restraint in a laparoscopy cradle, restraint in a laparoscopy cradle with intrauterine artificial insemination (AI) by laparoscopy, minimal restraint with cervical AI or restraint in a roll-over cradle plus foot-trimming. All treatments resulted in significant elevations in both heart rate and plasma cortisol concentrations (F < 0·001). The peak heart rate was significantly (P < 0·05) higher in ewes subjected to cervical AI than in those subjected to intrauterine insemination, with other treatments intermediate. The peak cortisol response did not differ among different treatments. The duration over which both the mean heart rate and -plasma cortisol concentrations remained significantly elevated above pre-treatment concentrations did not differ among treatment groups. The results of this study suggest that while restraint using a laparoscopy cradle is more stressful than that using a rollover cradle, the stress inflicted by intrauterine insemination by laparoscopy itself is no greater than that due to restraint using the laparoscopy cradle alone, cervical AI or the management practice offoot-trimming using a rollover cradle.

SUMMARYEnzyme-linked immunosorbent assay (ELISA) tests were used to measure IgG antibody levels in 2638 New Zealand children who had been immunized with the triple vaccine DTP. The percentage of children immune to diphtheria decreased with age. The percentage of children immune to tetanus varied from 67.1 to 55.0%. The percentage of children with measurable antibody to pertussis increased with age. The mean percentages of children with measurable antibody or immunity to one or more DTP components were 34.2% (with 3 components), 34.4% (2 components), and 78.1% (1 component). It appears the immunization strategy for diphtheria and tetanus is satisfactory for herd immunity in New Zealand children. However, the current pertussis strategy may not be providing adequate immunity to 5-year-olds in this country.

More instability (MSSD) and variability (SD) of negative affect (NA) have been related to current and future depressive symptoms. We investigated whether MSSD and SD of NA were predictive of the rate of symptom improvement during treatment and of reaching remission status. Forty-six individuals with major depressive disorder completed six days of ecological momentary assessments (10 beeps per day) before starting a combination of pharmacotherapy and supportive therapy. During and after treatment, the Hamilton Depression Rating Scale (HDRS) diagnostic interview was performed monthly for 18 months. Using multilevel modeling and logistic regression, a linear decrease in HDRS scores as well as reaching remission status (HDRS of ≤7 within or after five months) were predicted by the mean, SD and MSSD of NA in momentary assessments, and relevant baseline predictors. Mean NA, but not the SD or MSSD of NA, predicted rates of depressive symptom reduction over five months. The odds of remitting during treatment were not associated with any predictors. Our results suggest that pre-treatment assessments of NA instability and variability may not give an indication of the treatment response over time. Clinically, the mean of NA may be more promising as a baseline indicator of response potential.
