The preparation of L-lactyl-L-valine from L-lactic acid and L-valine, by methods used for the synthesis of peptides, was investigated. Initial experiments with O-benzoyl-DL-lactic acid and DL-valine benzyl ester unexpectedly gave N-benzoyl-DL-valine benzyl ester. However, coupling between O-benzyloxycarbonyl-DL-lactic acid and either DL-valine, or DL-valine benzyl ester, occurred in good yield. A simpler method, which was reported to give a 39% yield of L-lactyl-L-valine, gave only 10% of product but was modified to give L-lactyl-L-valine-1-14C in 90% yield.

Background: Mucopolysaccharidosis (MPS) is a rare congenital lysosomal storage disorder with complex airways. High anterior larynx is assessed by thyromental distance (TMD) nasendoscopy. A simpler method to assess this hyoid bone is described. The distance between the central-hyoid and symphysis of the mandible (hyo-mental distance; HMD) and inclination of this line to the horizontal axis (hyo-mental angle; HMA) in neutrally positioned patients is investigated. Methods: HMA, HMD in MPS, and non-MPS were compared, and their correlation with height and weight were assessed. Results: 50 adult MPS patients (M = 32, F = 18, age range = 19–66 years; mean BMI = 26.8 kg/m2) of MPS I, II, III, IV, and VI were compared with 50 non-MPS (M = 25, F = 25; age range = 22–84 years; mean BMI = 26.5 kg/m2). Mean HMA in MPS was 25.72° (−10 to +50) versus 2.42° (−35 to +28) in non-MPS. Mean HMD was 46.5 (25.7–66) millimeters in MPS versus 41.8 (27–60.3) in non-MPS. HMA versus height and weight showed a moderate correlation (r = −0.4, p < 0.05) in MPS and no significant correlation (r < 0.4, p > 0.05) in non-MPS. HMD versus height and weight showed no correlation (r < 0.4, p > 0.05) in both groups. Conclusions: HMA seems more acute in MPS despite nearly the same HMD as non-MPS, signifying a high larynx, which may be missed by TMD.

Abstract We describe a radioimmunoassay of serum cortisol in which 125I-tracer and preconjugated double antibody are used. This simpler method involves direct serum assay, and the majority of reagents are prepared in-house. Within-assay precision (coefficient of variation) of assay of Ortho Diognostic RIA Cortisol I and II was 3.2% (mean = 131 microgram/L, n = 10) and 2.2% (mean = 405 microgram/L, n = 10), respectively. Between-assay precision (coefficient of variation) for the same controls was 6.7% (mean = 128 microgram/L, n = 62) and 5.6% (mean = 395 microgram/L, n = 62), respectivley. By using automatic pipetters and a gamma counter with a data-reduction package, a technologist can complete 100 assays in less than 3 h. The operating cost is approximately 10 cents per tube.

Globe artichoke is an intriguing source of indigestible sugar polymers such as inulin-type fructans. In this study, the effect of ultrasound in combination with ethanol precipitation to enhance the extraction of long chain fructans from artichoke wastes has been evaluated. The inulin-type fructans content both from bracts and stems was measured using an enzymatic fructanase-based assay, while its average degree of polymerization (DP) was determined by HPLC-RID analysis. Results show that this method provides artichoke extracts with an inulin-type fructans content of 70% with an average DP between 32 and 42 both in bracts and in stems. The prebiotic effect of long chain inulins from artichoke extract wastes was demonstrated by its ability to support the growth of five Lactobacillus and four Bifidobacterium species, previously characterized as probiotics. Besides, we considered the possibility to industrialize the process developing a simpler method for the production of inulin-type fructans from the artichoke wastes so that the artichoke inulin preparation could be suitable for its use in synbiotic formulations in combination with different probiotics for further studies including in vivo trials.
