It is of crucial importance to evaluate the safety profile of the ingredients used in dermatological emulsions. A suitable equilibrium between safety and efficacy is a pivotal concern before the marketing of a dermatological product. The aim was to assess the safety and biological effects of a new cold processed silicone-based emulsion (SilEmulsion). The hazard, exposure, and dose-response assessment were used to characterize the risk for each ingredient. EpiSkin assay and human repeat insult patch tests were performed to compare the theoretical safety assessment toin vitroandin vivodata. The efficacy of the SilEmulsion was studied using biophysical measurements in human volunteers during 21 days. According to the safety assessment of the ingredients, 1,5-pentanediol was an ingredient of special concern since its margin of safety was below the threshold of 100 (36.53). EpiSkin assay showed that the tissue viability after the application of the SilEmulsion was 92 ± 6% and, thus considered nonirritant to the skin. The human studies confirmed that the SilEmulsion was not a skin irritant and did not induce any sensitization on the volunteers, being safe for human use. Moreover, biological effects demonstrated that the SilEmulsion increased both the skin hydration and skin surface lipids.

Aim. To determine whether 5-aminolevulinic acid-based photodynamic therapy (ALA-PDT) is effective in combating ultraviolet A- (UVA-) induced oxidative photodamage of hairless mice skin in vivo and human epidermal keratinocytes in vitro. Methods. In in vitro experiments, the human keratinocyte cell line (HaCaT cells) was divided into two groups: the experimental group was treated with ALA-PDT and the control group was left untreated. Then, the experimental group and the control group of cells were exposed to 10 J/m2 of UVA radiation. ROS, O2− species, and MMP were determined by fluorescence microscopy; p53, OGG1, and XPC were determined by Western blot analysis; apoptosis was determined by flow cytometry; and 8-oxo-dG was determined by immunofluorescence. Moreover, HaCaT cells were also treated with ALA-PDT. Then, SOD1 and SOD2 were examined by Western blot analysis. In in vivo experiments, the dorsal skin of hairless mice was treated with ALA-PDT or saline-PDT, and then, they were exposed to 20 J/m2 UVA light. The compound 8-oxo-dG was detected by immunofluorescence. Conclusion. In human epidermal keratinocytes and hairless mice skin, UVA-induced oxidative damage can be prevented effectively with ALA-PDT pretreatment.

Abstract Background – The skin barrier is important in the pathogenesis of atopic dermatitis and stratum corneum lipids have a critical role. Skin surface lipids have been largely overlooked but also contribute to barrier function. An untargeted approach was used to compare the skin surface lipids from atopic and non-atopic West Highland White terrier dogs. The primary hypothesis was that a difference in the lipidome of atopic and non-atopic dogs would be found and the secondary hypothesis was that affected and unaffected skin would differ in lipid profile.Results – Thirty-nine dogs were classified into one of four disease status groups based on strict criteria. Samples for lipid analysis were collected from affected and unaffected skin, and liquid chromatography/mass spectrometry found 421 lipid soluble features. Ten lipids were positively identified. Statistical analysis could not distinguish between non-atopic and atopic dogs. Partial least squares-discriminant analysis revealed a difference in the lipid profiles from affected and non-affected skin irrespective of disease status. Conclusions – An untargeted approach found a large array of unidentified lipids from the skin surface. There was a difference in the lipidome between affected and unaffected skin that was not related to disease status. Investigation into the lipidome of the skin surface in health and disease is an emerging area of research which could have clinical and therapeutic applications.

Contracting skeletal muscles emit pressure waves that are audible at the skin surface and are easily recorded with standard microphones both in vivo and in vitro. These muscle sounds are an intrinsic component of the contractile mechanism and are produced by mechanical vibrations at the resonant frequency of the muscle. The sounds are useful in measuring force, fatigue, and mechanical properties of muscle.
