Abstract Background Nanocolloidal human serum albumin radiolabelled with 99mTc provides a diagnostic radiopharmaceutical for sentinel node lymphoscintigraphy. NanoHSA (Nanotop), a commercially available kit, enables the simple preparation of this radiopharmaceutical via reconstitution with pertechnetate eluted from a generator. Thin-layer chromatography is widely used for determining radiochemical purity in clinical nuclear medicine. Quality control methods recommended by the manufacturer were sometimes reported to yield variable results. Therefore, we proposed and evaluated three alternative thin-layer chromatography methods for the quality control of [99mTc]Tc-NanoHSA from a commercially available kit. Results The radiochemical purity of [99mTc]Tc-NanoHSA determined with all methods was reproducible and met the requirements of the SPC and the European Pharmacopoeia (≥ 95%). Our quality control using iTLC-SG chromatographic paper in methyl ethyl ketone mobile phase identified only free pertechnetate as impurity, resulting in > 99% RCP. The quality control using iTLC-SG in 85% methanol or iTLC-SA in 0.9% NaCl identified an additional small fraction of a hydrophilic impurity, resulting in 95–97% RCP. Glucose was identified as a potential 99mTc-carrying hydrophilic species contributing to hydrophilic impurities. Conclusion Our quality control of [99mTc]Tc-NanoHSA with non-polar mobile phase tended to underestimate the amount of hydrophilic impurities, although without compromising the final quality of the radiopharmaceutical. Alternative TLC methods using aqueous mobile phases enabled a more accurate determination of hydrophilic impurities.

Anguilla bicolor is the fish that have high level economic and that was export commodity. One of potential that have in protein content of albumin more high than another fish, this matter can be used as substitute alternative Human Serum Albumin (HSA) which market so expensive. The aim of this research is to know quality of Anguilla bicolor albumin, to compare between albumins of fresh Anguilla bicolor and have steamed. The used method this study is experiment with sampling purposive and technic Duplo test analysis. This research use four times test and each sample take two quotation top (around the head) and bottom (around tail). Process sterilization and deposition albumin doing in two sections the first, using NaOH 10% to promote pH to 11.5 and discharge pH use H2SO410 % to 5,5. For test albumin quantity itself use spectrophotometer with 420nm spectrum. Based on outcome research indicate quality of Anguilla bicolor albumin steamed more high than fresh Anguilla bicolor however difference obtained is not significant.ABTRAKIkan sidat (Anguilla bicolor) merupakan ikan yang memiliki potensi dalam bidang ekonomi yang tinggi dan merupakan komoditas ekspor yang menjanjikan. Salah satu potensi yang dimiliki adalah kandugan protein albumin ikan sidat yang tinggi jika dibandingkan dengan ikan lainnya, hal ini dapat dimanfaatkan sebagai alternatif pengganti HSA (Human Serum Albumin) yang dipasaran harganya relatif mahal. Penelitian ini bertujuan untuk mengetahui kadar albumin ikan sidat, juga membandingkan antara albumin ikan sidat segar dan dikukus. Metode yang digunakan dalam penelitian ini adalah ekperimen dengan purposive sampling serta teknik analisa uji rerata duplo. Penelitian ini menggunakan empat kali ulangan dan masing-masing sampel diambil dua cuplikan bagian atas (sekitar kepala) dan bagian bawah (sekitar ekor). Proses penyeterilan dan pegendapan albumin dilakukan dengan 2 tahap yakni menggunakan NaOH 10% untuk menaikkan pH hingga 11,5 dan penurunan pH menggunakan H2SO410 % hingga 5,5. Untuk uji kuantitas albumin sendiri menggunakan spektrofotometer dengan panjang gelombang  450 A. Berdasalkan hasil penelitian menunjukkan bahwa kadar albumin ikan sidat yang dikukus lebih tinggi daripada ikan sidat yang segar namun perbedaan yang didapatkan tidak signifikan.Kata Kunci : Ikan Sidat (Anguilla bicolor), Albumin, Segar, Kukus

The effect of Fe3+ on the interaction between nitrofurazone (NF) and human serum albumin (HSA) was studied by fluorescence spectroscopy and UV-vis spectroscopy. The results from fluorescence spectra showed that Fe3+ increased the binding constant of NF to HSA, indicating longer storage time of NF in blood plasma. The effect model of Fe3+ was researched by binding distance and the UV-vis spectra, which showed that in the HSA-NF-Fe ternary system Fe-NF complex formed via metal ion bridge and enhanced the binding capability of NF to HSA.

ZusammenfassungNach tierexperimentellen Voruntersuchungen wurde die Placentographie mit trägerfreiem 113Inm -HSA als klinische Methode eingeführt. Vor Amniocentesen und bei Verdacht auf Placenta praevia werden Placentographien geschrieben. Den Schwangeren wird eine Aktivität von 500 μCi in die Cubitalvene injiziert. Die der Aktivität entsprechende Indiummenge ist kleiner als 0,1 ng. Die fetale Strahlenbelastung liegt unter lOmrad. Bei Anwendung von 113Inm-HSA entfällt eine Blockade der mütterlichen und fetalen Schilddrüsen. Die genaue Abgrenzung einer Placenta praevia wird nicht durch eine Blasenaktivität beeinträchtigt.Es wurden bisher 19 Placentalokalisationen durchgeführt. In allen Fällen konnte der Placentasitz eindeutig festgestellt werden. Bedingt durch die lange Liegezeit beim Aufnehmen eines Szintigramms kam es in zwei Fällen zu einem Vena-Cava-Kompressions-Syndrom. Zur Verhinderung dieser klinischen Zwischenfälle werden inzwischen Placentographien mit der Anger-Kamera aufgenommen. Mit Hilfe des divergierenden Kollimators konnte der gesamte Abdominalbereich erfaßt werden. Die Aufnahmezeit konnte auf 7 — 10 Minuten verkürzt werden. Die intravenöse injizierte Aktivität betrug bei dieser Methode ebenfalls 500 μCi. Der diagnostische Aussagewert der Kamerabilder ist szintigraphischen Aufnahmen gleichwertig.
