Results are presented of 130-145 generations of selection for low scutellar bristle number in four lines of D. melanogaster derived directly from an Oregon-RC wild-type stock and in one derived from an Oregon-RC line selected for low sternital bristle number. The most rapid initial response and the lowest mean scutellar bristle number ultimately reached, just below 2 bristles, occurred in a line in which the response was due to a new recessive gene located at approximately 17�4 on the X chromosome. Three of the other four lines reached a plateau just above a mean of 2 bristles after different patterns of response. These plateaux reflected a new canalization or threshold phenomenon at 2 bristles in these lines. The remaining line reached a mean of about 2� 5 bristles after some 50 generations and remained at that level or slightly higher thereafter, but had no indication of canalization at 2 bristles. Two relaxed lines were derived from each selection line at different times and showed variable patterns of regression towards the base population level.

ABSTRACT Results are presented of 135 generations of selection for high scutellar bristle number in two lines M and M3 derived from the same original mating of one female with 5 bristles by one male with 4 bristles, the latter being the wild-type canalised phenotype. Results are also given of two relaxed lines per line and of a reselection line M2 derived from the first relaxed line of line M which had regressed almost to base population level. The effect of introducing the sc1 allele into the M and M3 selected backgrounds was studied at generations 39–44. At the end of selection the effect of an extra dose of sc+ was also studied in males of all selected backgrounds. The correlated responses in abdominal bristles were followed in all lines.—Considering their common origin, the selection lines differed markedly in pattern of scutellar response and in most other aspects observed, namely correlated responses in abdominals and p.c. scutellars, sex differences, and behaviour on relaxation. Selection limits for scutellar bristles in lines M and M2 were equal to or greater than the most extreme reported in the literature.—The probit span of the canalised 4 bristle class decreased in each selection line as the mean scutellar bristle number increased, and increased again in the relaxed lines as the mean bristle number decreased. In the context of an hypothesis that canalisation at 4 bristle is due to regulation of the scute locus, this result is now interpreted as being due mainly to selection for poor regulators of sc+, in contrast to a previous interpretation that only the minor gene background was altered by selection, the canalisation (regulation) genotype not being affected.—Introducing the sc  1 allele into the selected backgrounds M and M3 showed a reduced effect on sc  1 flies compared with sc+ flies, and an interaction of sc  1 and sc+ with selected background. sc  1 flies had about the same number of bristles in both backgrounds though the mean of sc+ flies in line M was about 3? higher than in line M3. Dominance of sc+ to sc  1 was reduced slightly in M3. However, the effect of an extra dose of sc+ at the end of selection was about the same as in unselected in all lines, so the first or dominance level of regulation of the scute locus was not significantly affected by selection, though the second or canalisation level of regulation was.—A large positive correlated response in abdominal bristles occurred in all lines. The response in line M was about twice that in M2 and M3 and was in fact as large as can be obtained from direct selection on abdominals. In line M some genes may have been selected with a proportionately greater effect on abdominals than on scutellars. This is supported by the further observation in line M that the abdominal scores of flies with particular scutellar bristles scores increased as the scutellar mean increased. An attempt was made to apply to these results Rendel's (1962) model of competition between scutellars and abdominals for common bristle-making resources. This could not be done satisfactorily mainly because the assumptions in the model about the similarity of effects in scute and wild-type flies were not met in the present material.

SUMMARYA major difference in developmental stability has been demonstrated between two populations produced by artificial selection for supernumerary scutellar bristles. The test system involves the substitution of an X-ray induced partial revertant of sc1 for the wild-type allele at the scute locus, enabling direct comparisons to be made of the degree of canalization at the wild-type level of expression of the character. One population is comparable with the unselected Canberra stock in stability, though it differs appreciably in mean bristle number: the other population shows a marked reduction in the level of regulation of bristle number variability. The alleles responsible for the reduced level of canalization are rare in the base population, and are of particular importance in the determination of limits to directional selection. Their effects on developmental stability have been shown to depend on the activity of the allele at the scute locus.

SUMMARYIndividual and within-full-sib family selection for low sternopleural bristle number was carried out for 17 generations, with six replicate lines for each selection method. Our results can be summarized as follows: (1) the response to selection was exhausted very quickly, (2) the additive variance of the selected lines declined rapidly, (3) the variation in response to selection decreased as selection progressed, (4) genetic differences among replicates at the selection limit were small, (5) individual selection resulted in a higher initial response than within-family selection, but similar limits were achieved with both procedures. These observations are consistent with the hypothesis that the pattern of response to selection is due to the segregation in the base population of only a few loci with large effects, at intermediate frequencies.

Abstract Following mutagenesis with ethyl methanesulfonate, selection in a susceptible strain with a concentration of the insecticide diazinon (0.0004%, w/v) above that required to kill 100% of the susceptible strain, the LC100 of that strain, resulted in a single gene response. The resultant four mutant resistant strains have equivalent physiological, genetical and biochemical profiles to a diazinon-resistant strain derived from a natural population and homozygous for the Rop-1 allele. Modification of the microsomal esterase E3 is responsible for resistance in each case. The Rop-1 locus maps approximately 4.4 map units proximal to bu on chromosome IV. Selection within the susceptible distribution, at a concentration of diazinon [0.0001% (w/v)] less than the LC100, resulted in a similar phenotypic response irrespective of whether the base population had been mutagenized. The responses were polygenically based, unique to each selection line and independent of Rop-1. The relevance of the results to selection for insecticide resistance in laboratory and natural populations is discussed.

The results of short runs of disruptive and high selection for scutellar bristles in wild-type Drosophila are explained in terms of the hypothesis that canalization at four bristles is due to regulation of the major gene in the developmental system (Rendel, Sheldon, and Finlay 1965). Selection response has probably been due to selection for modifier (minor) genes rather than for isoalleles of the major gene or weak regulator alleles. Some environmental effects on the character, short runs of selection for low bristle number or different bristle types, and effects of relaxing selection are also reported.

SUMMARYThe effect of subdivision of a population on response to artificial directional selection for abdominal bristle number in Drosophila melanogaster was compared using large, replicated lines. Three different population structures were compared: (i) selection in an Undivided, large population with 50 pairs of parents (treatment U); (ii) selection in each of 10 sublines which were reconstituted every 6th generation by Crossing after Culling the 5 lowest sublines (treatment CC); and (iii) selection in each of 10 sublines which were reconstituted every 6th generation by Crossing after Retaining all 10 sublines (treatment CR). At the end of three cycles of selection and crossing, neither CR nor CC was superior to U; sublining did not increase response to selection. These results agree with the predictions arising from an entirely additive model and provide no evidence for the presence of epistasis.A comparison of 50-pair lines (U) with several 5-pair lines was made over 31 generations. For the 50-pair lines, there was close agreement between response predicted from the base population (using ih2σp) and observed response throughout all 31 generations of selection. Although the best of the 5-pair lines exceeded the 50-pair lines in the early generations, average response to directional selection in the 5-pair lines soon fell behind that predicted from ih2σp, and soon reached a plateau.

Selection for and against the canalized phenotype in scutellar bristles was attempted in two selection lines and a randomly selected line was used as control. The selection lines were the Decanalization line (D) and the Canalization line (N). The D line was maintained by matings of scute males (scwbl) with three scutellars with wild-type females (scwbl/yw) with five bristles, in the N line scute males with four bristles were mated with wild-type females also with four bristles, while in the C line males and females of the above genotypes were selected at random. The lines were established from a sample of flies taken from a line selected for high scutellar numbers.After eighteen generations of selection the C line was characterized by a regression of mean bristle number without appreciable change in variance. Relative to the N line, the D population showed a lower proportion of flies having four scutellars, a higher variance in bristle numbers, and a higher proportion of four-bristle scute flies having abnormal patterns.Two alternative hypotheses were advanced to account for the results of this experiment. The first postulated a relative change in the widths of the four-bristle canalization zones in the selection lines, while the second suggested a relative change in frequencies of specific modifier genes for scutellars in scute and in wild-type genotypes of the lines. The evidence favours the latter hypothesis.

The first dose effect of sc+ has been measured for abdominal bristle number in six replicate lines of D. melanogaster being selected upward for this character in the sc homo-and hemizygote. The within-line regression coefficient of the heterozygote on the sc homozygote showed considerable variation among the lines with a range of 0-47-1' 22, indicating the dependence of gene action on genetic background. But, on the average, the effect of sc+ was approximately additive for abdominal bristle number. From this average, the genetic correlation in the base population between the two genotypes was estimated to be considerably less than unity, which suggests some genetic variability that is dependent on the genotype of the major locus. A similar relationship was also obtained for the two male hemizygotes.

Although there is little experimental information on the effect of simultaneous selection for two quantitative characters on the magnitude of the genetic correlation between them, it is apparently generally expected that such selection for the two characters in the same direction will cause a negative change in the genetic correlation, and selection in opposite directions a positive change. Selection using independent culling levels was done for each of the four combinations of high or low third coxal bristle number with high or low sternopleural bristle number in Drosophila melanogaster for 22 generations. To estimate changes in the genetic correlation, realized genetic parameters were estimated from single-trait lines started from the base population, and from the two-trait lines after 10 and 22 generations of selection. Changes in the genetic correlation in individual two-trait selection lines were variable and unpredictable. At generation 22 concurrent two-trait selection had resulted in significantly larger realized genetic correlations than divergent two-trait selection, so that results were contrary to the generally accepted expectation.

Abstract Direct selection for increased resistance to a heat shock (41.9° for 90 min) was carried out using two replicate lines of Drosophila buzzatii that were derived from a large base population. Selected individuals were first acclimated to high temperature before selection, while control individuals were acclimated but not selected, and selection was performed every second generation. Resistance to heat shock with acclimation increased in selected lines. Without acclimation, a correlated smaller increase in heat-shock resistance was suggested. Survival of males was higher than that of females in all lines when tested with acclimation, but with direct exposure to high temperatures, survival of females was greater than that of males both in selection and control lines but not in the base population. From analysis of reciprocal cross progeny between lines, one selection line was found to possess a dominant autosomal factor that significantly increased resistance of males much more than resistance of females. Also suggestive was recessive traits on the X chromosome in both selection lines that increased thermotolerance. No cytoplasmic effects were found. After accounting for other effects, survival of F1 flies was intermediate, suggesting that additive variation is present for one or more of the autosomes.
