Blood eosinophils in COPD: Friend or foe?

We have analysed the relationship of blood eosinophil count and serum eosinophil cationic protein (ECP) levels in patients with acute and chronic idiopathic urticaria. The ECP levels and eosinophil counts were measured in the peripheral blood of 15 patients with acute urticaria, 25 with chronic idiopathic urticaria and 10 normal healthy subjects. Blood eosinophil counts and serum ECP levels increased in all patients with acute urticaria. Concerning patients affected by chronic urticaria, taking into account the recrudescence of the disease at the moment of taking the blood sample, only symptomatic patients showed increased eosinophil blood values whereas serum ECP levels were increased both in symptomatic and asymptomatic patients. Furthermore, serum ECP levels in chronic urticaria did not correlate with the peripheral eosinophil counts, as they did in acute urticaria. The results of the present study indicate that eosinophils may play a role in the inflammatory mechanisms in patients with acute and chronic urticaria showing a positive correlation between serum ECP levels and disease activity.

AbstractAtopic dermatitis (AD) often presents more severely in African Americans (AAs) and with greater involvement of extensor areas. To investigate immune signatures of AD in AAs with moderate to severe pruritus, lesional and non-lesional punch biopsies were taken from AA patients along with age-, race-, and sex-matched controls. Histology of lesional skin showed psoriasiform dermatitis and spongiotic dermatitis, suggesting both Th2 and Th17 activity. Gene Set Variation Analysis showed upregulation of Th2 and Th17 pathways in both lesional versus non-lesional and lesional versus control (p < 0.01), while Th1 and Th22 upregulation were observed in lesional versus control (p < 0.05). Evidence for a broad immune signature also was supported by upregulated Th1 and Th22 pathways, and clinically may represent greater severity of AD in AA. Furthermore, population-level analysis of data from TriNetX, a global federated health research network, revealed that AA AD patients had higher values for CRP, ferritin, and blood eosinophils compared to age-, sex-, and race-matched controls as well as white AD patients, suggesting broad systemic inflammation. Therefore, AA AD patients may feature broader immune activation than previously thought and may derive benefit from systemic immunomodulating therapies that modulate key drivers of multiple immune pathways.

Counts of granulocytes were performed on thymic imprints from normal rabbits 3 days old, 2–3 weeks old, and young adults, and from young adult rabbits mildly and severely parasitized with coccidiosis. Analyses of percentages of eosinophils, heterophils, and basophils indicated no sex differences. Comparisons of relative numbers of granulocytes in normal rabbits showed that young adult rabbits possessed more eosinophils and fewer basophils than the younger groups (no differences in heterophils); 3-day-old rabbits had more eosinophils than the 2-to 3-week animals (no differences in heterophils and basophils). No differences in relative numbers of the various granulocytes were found in the mildly and severely parasitized rabbits, but there was a decrease in eosinophils in the parasitized rabbits when compared with the young normal adult rabbits. Eosinophilic leukocytes were divided into two groups: the mononuclear "tissue" line and the lobated "blood" line of development. In the 3-day-old rabbits about 95% of eosinophils were of the lobed variety but in the older normal rabbits, the two developmental lines were about equal in numbers. When compared with the normal young adult rabbits, parasitized rabbits have increased percentages of "blood" eosinophils as a result of greater numbers of band and polymorphonuclear cells.
