Insecticidal activity of culture filtrates from liquid medium of Beauveria bassiana isolates from South Sumatra (Indonesia) wetland soil against larvae of Spodoptera litura. Biodiversitas 20: 2101-2109. The obstacle in utilizing entomopathogenic fungi to control pest insects in wetlands is the inability of their isolates to survive during the saprophytic phase due to the soil being very acidic. Therefore, the exploration of fungi being able to survive in the acidic soil was utilized in the acidic ecosystem. This study aimed to test the toxic activity of Beauveria bassiana culture filtrates from liquid medium at pH 2.5 and 6 against Spodoptera litura larvae. Total of ten isolates collected from South Sumatra, Indonesia was grown in liquid media at pH of 2.5 and 6 for six weeks. The result showed that the culture filtrate from liquid medium at pH 6 was more toxic against the larvae than that at pH 2.5. The mortality of the larvae at pH 6 reached 92%, while that at pH 2.5 it reached 13.33% only. The findings of present study revealed that the culture filtrate isolates of BPdR, BJgTs, BSwTd2, BSwTd3, BSwTd4, BKKPp2 from pH 2.5 media still caused high larvae mortality (6.67-13.33%). These seven isolates are superior due to their toxic nature at pH 2.5. Consequently, present study has increased the chances of success in utilizing these isolates for biological control in acidic ecosystems, such as peatlands, tidal lowlands, and freshwater swamps.

Activity of Melia azedarach (L.) seed extract against armyworm Spodoptera litura (F.) (Lepidoptera: Noctuidae). This study was conducted to evaluate the effectivenes and biological activity of Melia azedarach L. (Meliaceae) seed extract against armyworm, Spodoptera litura F. The first instar larvae were fed extract-treated cotton leaves for 2 days, then were maintained on untreated leaves until the third instar stage. Records were kept in regard to the larvae mortality and developmental time of surviving larvae from first instar to third instar. The result showed that Melia azedarach L. seed extract at consentration of 50 g of seeds/l of water (5%) exhibited moderate insecticidal activity against S. litura larvae (43.33 - 68.33% mortality). Addition of detergen at 0.2% to extract did not increase insecticidal activity of the extract. However, boiling seed extract at consentration of 50 g of seeds/l of water (5%) during 10 until 20 minutes increased insecticidal activity of extract (66.67 - 68.33% mortality). Generally, M. azedarach seed extract treatment did not affect  developmental time of  S. litura larvae.

Penelitian ini bertujuan untuk mempelajari karakter fisiologi isolat B. bassiana yang berasal dari berbagai inang dan virulensinya terhadap Spodoptera litura. Lima isolat B. bassiana yang diuji diisolasi dari hama Leptocorisa oratorius (F.) dan Hypothenemus hampei (Ferr.), rizosfer tanaman bawang daun buah kakao dan batang gandum. Karakter fisiologi yang diamati adalah daya kecambah konidia, pertumbuhan koloni, dan sporulasi, Cendawan diperbanyak pada media SDAY dan uji virulensi dilakukan terhadap larva instar II S. litura. Hasil penelitian menunjukkan bahwa daya kecambah konidia, pertumbuhan koloni dan sporulasi bervariasi antar isolat. Isolat B. bassiana yang diisolasi dari L. oratorius dan buah kakao memiliki karakter fisiologi yang lebih baik dibandingkan dengan isolat lain. Kedua isolat ini juga lebih virulen terhadap larva instar II S. litura dengan mortalitas sebesar 80 – 81,67%.

Beauveria bassiana is an important entomopathogenic fungus that used as a biocontrol agent of insect pests. Maintaining and preserving B. bassiana cultures is essential for the effective evaluation of its potential as microbial agent against insect pest, for biodiversity studies and also for exchange of fungal material between laboratories. In the present work we evaluated the suitability of four preservation materials based on gelatin and rice to maintain the viability of B. bassiana to be used as baits for insect’s control. The gelatin amended with sugar recorded the maximum viability after 70 days of storage as 98.3% compared to 63.3% on rice media. Effective control of 80% of the ants was observed after 10 days of feeding on gelatin amended with sugar and rice bait. The results demonstrated that treatment with formulations containing conidia of B. bassiana presents insecticidal activity against ant in addition of acting as preservation materials.

Bacillus thuringiensis (Bt) Vip3A proteins are important insecticidal proteins used for control of lepidopteran insects. However, the mode of action of Vip3A toxin is still unclear. In this study, the amino acid residue S164 in Vip3Aa was identified to be critical for the toxicity in Spodoptera litura. Results from substitution mutations of the S164 indicate that the insecticidal activity of Vip3Aa correlated with the formation of a >240 kDa complex of the toxin upon proteolytic activation. The >240 kDa complex was found to be composed of the 19 kDa and the 65 kDa fragments of Vip3Aa. Substitution of the S164 in Vip3Aa protein with Ala or Pro resulted in loss of the >240 kDa complex and loss of toxicity in Spodoptera litura. In contrast, substitution of S164 with Thr did not affect the >240 kDa complex formation, and the toxicity of the mutant was only reduced by 35%. Therefore, the results from this study indicated that formation of the >240 kDa complex correlates with the toxicity of Vip3Aa in insects and the residue S164 is important for the formation of the complex.
