Flavobacterium yonginense sp. nov. and Flavobacterium myungsuense sp. nov., isolated from a mesotrophic artificial lake

Two non-motile, Gram-staining-negative, yellow-pigmented bacterial strains designated HMD1001T and HMD1033T were isolated from the water of a mesotrophic artificial lake in Korea. A phylogenetic tree based on 16S rRNA gene sequences indicated that both strains could be assigned to the genus Flavobacterium ; strain HMD1001T appeared most closely related to Flavobacterium fluvii H7T (96.8 % sequence similarity), F. succinicans DSM 4002T (96.6 %) and F. hydatis DSM 2063T (96.6 %) whereas strain HMD1033T appeared most closely related to Flavobacterium psychrolimnae LMG 2201T (96.2 %), F. segetis AT1048T (96.2 %) and F. weaverense AT1042T (96.2 %). The major fatty acids of strain HMD1001T were iso-C15 : 0 (21.5 %), summed feature 3 (comprising C16 : 1ω6c and/or C16 : 1ω7c; 18.0 %) and iso-C15 : 1 G (7.6 %), whereas those of HMD1033T were summed feature 3 (23.8 %), iso-C15 : 0 3-OH (16.9 %), iso-C15 : 0 (15.3 %) and anteiso-C15 : 0 (12.1 %). The genomic DNA G+C contents of strains HMD1001T and HMD1033T were 35.9 and 32.2 mol%, respectively. Phylogenetic and phenotypic evidence indicates that strains HMD1001T and HMD1033T represent two novel species of the genus Flavobacterium , for which the names Flavobacterium yonginense sp. nov. (type strain HMD1001T  = KCTC 22796T  = CECT 7594T) and Flavobacterium myungsuense sp. nov. (type strain HMD1033T  = KCTC 22825T  = CECT 7649T) are proposed.

A Gram-staining-negative, rod-shaped bacterium, strain HMD2169T, was isolated from a mesotrophic artificial lake in Korea. Strain HMD2169T grew in the presence of 0–3.0 % (w/v) NaCl, at pH 5–10 and at 20–37 °C. The predominant quinone of strain HMD2169T was ubiquinone (UQ)-8. The major fatty acids were summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c), C16 : 0 and summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c). The major polar lipids were phosphatidylethanolamine, diphosphatidylglycerol, two unidentified aminolipids and two unidentified lipids. The DNA G+C content was 59.8 mol%. A phylogenetic tree based on 16S rRNA gene sequences showed that strain HMD2169T was a representative of a lineage within the genus Chitinimonas . Strain HMD2169T was closely related to Chitinimonas taiwanensis (95.8 % sequence similarity) and Chitinimonas koreensis (94.6 %). On the basis of the evidence presented in this study, strain HMD2169T is a representative of a novel species of the genus Chitinimonas , for which the name Chitinimonas viridis sp. nov. is proposed with the type strain HMD2169T ( = KCTC 22839T = CECT 7703T).

Two bacterial strains, designated KV-962T and KV-963, were isolated from soil collected from a field in Japan. Cells of both strains were Gram-staining-positive, non-spore-forming, short rod-shaped and motile. Phylogenetic analysis based on 16S rRNA gene sequences indicated that these strains were related to Conexibacter woesei DSM 14684T, with a similarity value of 98.6 %. These strains possessed MK-7 (H4) as the sole menaquinone and contained C18 : 1ω9c, C17 : 1ω6c and iso-C16 : 0 as the major fatty acids. On the basis of genotypic and phenotypic characteristics, strain KV-962T and KV-963 were indicated as a novel species of the genus Conexibacter , for which the name Conexibacter arvalis sp. nov. is proposed. The type strain is KV-962T ( = DSM 23288T  = NBRC 106558T)

A Gram-staining-negative, rod-shaped, gliding and pale-pink-pigmented bacterium, designated strain ZLM-10T, was isolated from a soil sample collected from an arid area in Xinjiang province, China, and characterized in a taxonomic study using a polyphasic approach. The novel strain grew optimally at 30–37 °C and in the presence of 2 % (w/v) sea salts. The only respiratory quinone detected was MK-7 and the major cellular fatty acids were summed feature 3 (iso-C15 : 0 2-OH and/or C16 : 1ω7c), iso-C15 : 0 and iso-C17 : 0 3-OH. The polar lipids consisted of diphosphatidylglycerol, phosphatidylethanolamine, an unidentified aminolipid and two unidentified aminophospholipids. The DNA G+C content was 45.4 mol%. Flexirubin-type pigments were not produced. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain ZLM-10T was a member of the phylum Bacteroidetes and appeared most closely related to Cesiribacter roseus 311T (90.2 % sequence similarity), Marivirga sericea LMG 13021T (89.2 %), Cesiribacter andamanensis AMV16T (89.1 %) and Marivirga tractuosa DSM 4126T (89.1 %). On the basis of phenotypic and genotypic data and phylogenetic inference, strain ZLM-10T should be classified as a novel species of a new genus in the family Flammeovirgaceae , for which the name Nafulsella turpanensis gen. nov., sp. nov. is proposed. The type strain of the type species is ZLM-10T ( = CCTCC AB 208222T = KCTC 23983T).

Four Gram-staining-positive, catalase-negative, coccoid isolates, designated NTUH_1465T, NTUH_2196, NTUH_4957 and NTUH_5572T, were isolated from human specimens. The four isolates displayed more than 99.6 % 16S rRNA gene sequence similarity with Gemella haemolysans ATCC 10379T, and 96.7 to 98.6 % similarity with Gemella sanguinis ATCC 700632T, Gemella morbillorum ATCC 27824T or Gemella cuniculi CCUG 42726T. However, phylogenetic analysis of concatenated sequences of three housekeeping genes, groEL, rpoB and recA, suggested that the four isolates were distinct from G. haemolysans ATCC 10379T and other species. Isolates NTUH_2196, NTUH_4957 and NTUH_5572T clustered together and formed a stable monophyletic clade. DNA–DNA hybridization values among strains NTUH_1465T and NTUH_5572T and their phylogenetically related neighbours were all lower than 49 %. The four isolates could be distinguished from G. haemolysans and other species by phenotypic characteristics. Based on the phylogenetic and phenotypic results, two novel species Gemella parahaemolysans sp. nov. (type strain NTUH_1465T = BCRC 80365T = JCM 18067T) and Gemella taiwanensis sp. nov. (type strain NTUH_5572T = BCRC 80366T = JCM 18066T) are proposed.

Two Gram-stain-negative, aerobic, non-motile bacterial strains, 36D10-4-7T and 30C10-4-7T, were isolated from bark canker tissue of Populus × euramericana, respectively. 16S rRNA gene sequence analysis revealed that strain 36D10-4-7T shows 98.0 % sequence similarity to Sphingomonas adhaesiva DSM 7418T, and strain 30C10-4-7T shows highest sequence similarity to Sphingobacterium arenae H-12T (95.6 %). Average nucleotide identity analysis indicates that strain 36D10-4-7T is a novel member different from recognized species in the genus Sphingomonas . The main fatty acids and respiratory quinone detected in strain 36D10-4-7T are C18 : 1  ω7c and/or C18 : 1  ω6c and Q-10, respectively. The polar lipids are diphosphatidylglycerol, phosphatidylcholine, phosphatidylglycerol, aminolipid, phosphatidylethanolamine, sphingoglycolipid, two uncharacterized phospholipids and two uncharacterized lipids. For strain 30C10-4-7T, the major fatty acids and menaquinone are iso-C15 : 0, C16 : 1  ω7c and/or C16 : 1  ω6c and iso-C17 : 0 3-OH and MK-7, respectively. The polar lipid profile includes phosphatidylethanolamine, phospholipids, two aminophospholipids and six unidentified lipids. Based on phenotypic and genotypic characteristics, these two strains represent two novel species within the genera Sphingomonas and Sphingobacterium . The name Sphingomonas corticis sp. nov. (type strain 36D10-4-7T=CFCC 13112T=KCTC 52799T) and Sphingobacterium corticibacterium sp. nov. (type strain 30C10-4-7T=CFCC 13069T=KCTC 52797T) are proposed.

Two novel Gram-staining-negative, aerobic, cocci-shaped, non-motile, non-spore forming, pink-pigmented bacteria designated strains T6T and T18T, were isolated from a biocrust (biological soil crust) sample from the vicinity of the Tabernas Desert (Spain). Both strains were catalase-positive and oxidase-negative, and grew under mesophilic, neutrophilic and non-halophilic conditions. According to the 16S rRNA gene sequences, strains T6T and T18T showed similarities with Belnapia rosea CGMCC 1.10758T and Belnapia moabensis CP2CT (98.11 and 98.55% gene sequence similarity, respectively). The DNA G+C content was 69.80 and 68.96% for strains T6T and T18T, respectively; the average nucleotide identity by blast (ANIb) and digital DNA–DNA hybridization (dDDH) values confirmed their adscription to two novel species within the genus Belnapia . The predominant fatty acids were summed feature 8 (C18 : 1ω7c/C18 : 1ω6c), C16 : 0, C18 : 1 2-OH and summed feature 3 (C16 : 1ω7c/C16 : 1ω6c). According to he results of the polyphasic study, strains T6T and T18T represent two novel species in the genus Belnapia (which currently includes only three species), for which names Belnapia mucosa sp. nov. (type strain T6T = CECT 30228T=DSM 112073T) and Belnapia arida sp. nov. (type strain T18T=CECT 30229T=DSM 112074T) are proposed, respectively.

A novel strain designated SR18T was isolated from the rhizosphere soil of a ginseng in Korea. Cells were Gram-staining-negative, motile by gliding, catalase-positive and oxidase-negative, non-spore-forming rods. The isolate grew aerobically at 15–45 °C (optimum 28 °C), pH 5.5–7.5 (optimum pH 7.0) and with 0–3.0 % (w/v) NaCl (optimum 1.5 % NaCl). Phylogenetic analyses based on 16S rRNA gene sequences indicated that strain SR18T belongs to the genus Chitinophaga with sequence similarity of 97.2 % and 97.0 % to Chitinophaga japonensis 758T and Chitinophaga rupis CS5-B1T, respectively. Similarity to other species of the genus Chitinophaga was 92.8–95.5 %. The predominant menaquinone was MK-7. Major fatty acids were iso-C15 : 0 and C16 : 1ω5c. The polar lipids included phosphatidylethanolamine, unidentified phospholipids, unknown aminolipids and unknown lipids. The genomic DNA G+C content was 45.3 mol%. DNA–DNA relatedness between strain SR18T and C. japonensis NBRC 16041T was 29–32 %. On the basis of polyphasic analysis from this study, strain SR18T represents a novel species of the genus Chitinophaga , for which the name Chitinophaga ginsengihumi sp. nov. is proposed. The type strain is SR18T ( = KACC 17604T = NBRC 109832T).

Two bacterial strains, designated JAM1T and JAM7T, were isolated from a methanol-fed denitrification system treating seawater at the Montreal Biodome, Canada. They were affiliated within the genus Methylophaga of the Gammaproteobacteria by analysis of the 16S rRNA gene sequences. Strain JAM1T had the capacity to grow under denitrifying conditions by reducing nitrate into nitrite which is unique among the species of the genus Methylophaga . Major fatty acids were C16 : 1ω7c or ω6c, C16 : 0 and C18 : 1ω7c or ω6c. The major ubiquinone was Q8. Both strains required vitamin B12 and Na+ ions for growth. The genomes of strains JAM1T and JAM7T have been completely sequenced and showed a DNA G+C content of 44.7 mol% and 47.8 mol%, respectively. Growth occurred at pH 6–11 and at 0.5–8 % NaCl. Both genomes contained predicted ORFs encoding the key enzymes of the ribulose monophosphate pathway. Also, operons encoding two nitrate reductases (Nar), two nitric oxide reductases (Nor), one nitrous oxide reductase (Nos) and one truncated nitrite reductase (NirK) were clustered in a 67 kb chromosomal region in strain JAM1T. No such operons were found in strain JAM7T. These results supported the affiliation of the two strains as novel species within the genus Methylophaga . The names Methylophaga nitratireducenticrescens sp. nov. for type strain JAM1T ( = DSM 25689T = ATCC BAA-2433T) and Methylophaga frappieri sp. nov. for type strain JAM7T ( = DSM 25690T = ATCC BAA-2434T) are proposed.

A Gram-negative, aerobic and non-motile rod, designated Y4T, was isolated from a cucumber leaf from Pinggu District, east Beijing, PR China. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain Y4T was most closely related to Luteimonas aquatica RIB1-20T (96.7 % 16S rRNA gene sequence similarity). DNA–DNA relatedness between strain Y4T and L. aquatica RIB1-20T was 42.5±3.9 %. The predominant fatty acids were iso-C15 : 0, iso-C17 : 1ω9c, iso-C16 : 0 and iso-C17 : 0. The major ubiquinone was Q-8. The DNA G+C content of the type strain was 69.9 mol%. Based on the evidence above, strain Y4T represents a novel species of the genus Luteimonas , for which the name Luteimonas cucumeris sp. nov. is proposed. The type strain is Y4T ( = CGMCC 1.10821T = KCTC 23627T).

A novel haloalkaliphilic archaeon, strain B23T was isolated from the former lake Texcoco in Mexico. The strain was Gram-stain-negative, the cells coccoid to ovoid rods, red pigmented and aerobic. Strain B23T grew in 1.7–4.3 M NaCl, at pH 6.5–9.5 and at 25–45 °C with optimal growth at 2.6–3.4 M NaCl, pH 7.5–8.5 and 37 °C. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain B23T was most closely related to Natronobacterium gregoryi SP2T with 97.3 % sequence similarity. The polar lipids of strain B23T were phosphatidylglycerol and several unidentified phospholipids. The G+C content of the DNA of the strain was 62.5 mol%. Levels of DNA–DNA relatedness between strain B23T and Natronobacterium gregoryi DSM 3393T was 32.3 %. The name Natronobacterium texcoconense sp. nov. is proposed. The type strain is B23T ( = CECT 8068T = JCM 17655T).
