Spencermartinsiella europaea gen. nov., sp. nov., a new member of the family Trichomonascaceae

Ten strains of a novel heterothallic yeast species were isolated from rotten wood collected at different locations in Hungary. Analysis of gene sequences for the D1/D2 domain of the large subunit rRNA, as well as analysis of concatenated gene sequences for the nearly complete nuclear large subunit rRNA, nuclear small subunit rRNA and translation elongation factor 1-α, placed the novel species in the family Trichomonascaceae, but showed that it was distinct from all currently recognized genera. The name Spencermartinsiella europaea gen. nov., sp. nov. is proposed to accommodate the new genus and novel species. The novel species could be distinguished from recognized species of neighbouring genera on the basis of standard phenotypic characteristics. The type and isotype strains of Spencermartinsiella europaea are NCAIM Y.01817T ( = NRRL Y-48265T =  CBS 11730T) and NCAIM Y.01819I ( = NRRL Y-48266I = CBS 11731I), respectively.

Specimens of a new pleosporalean taxon were obtained on the bark of Juniperus excels from the northern mountains of Oman; from the Jebel Akhdar (‘Green Hills’). Sequence analyses based on the regions of large subunit rRNA (LSU), small subunit rRNA (SSU), translation elongation factor 1-α (TEF) and internal transcribed spacers (ITS) were performed to resolve the phylogenetic relationships of the new taxon in Phaeosphaeriaceae. The data concluded that the taxon represents a novel genus of the family Phaeosphaeriaceae and the generic name Hydeomyces and the species name H. desertipleosporoides are introduced for the new taxon. An outline of the characters which differentiate the new genus from phylogenetically closely related genera Dematiopleospora and Dlhawksworthia is given and its morphology of asexual and sexual morphs is described.

During a survey of saprobic microfungi in Southwest China, a coelomycetous fungus was found on dead twigs of Jasminum nudiflorum in Kunming, Yunnan Province. Based on a detailed morphological characterization coupled with multi-locus phylogenetic analyses, the fungus was identified as a new species in the genus Dothidea. Phylogenetic analyses using a combined matrix consisting of internal transcribed spacer (ITS), large subunit rRNA (LSU), small subunit rRNA (SSU), beta tubulin (tub2) and translation elongation factor-1 alpha (tef1-α) confirmed its placement in Dothideaceae and revealed a sister relationship to Dothidea eucalypti. The new species is characterized by pycnidial conidiomata, ampulliform or doliiform conidiogenous cells as well as aseptate, subglobose to ovoid, hyaline to pale-brown conidia. Comprehensive descriptions and illustrations are provided. Morphological characteristics of asexual morph taxa in Dothideaceae are also summarized and discussed.

Ambrosiozyma oregonensis sp. nov. is described from two strains, one isolated from a mountain stream in Oregon, USA (NRRL Y-6106T = CBS 5560T), and a second (NRRL YB-4169) from an unknown substrate from Marion, Illinois, USA. The species forms four hat-shaped ascospores in each deliquescent ascus and appears to be homothallic. Abundant true hyphae are produced with some having apparent dolipore-like septa. Analyses of nuclear gene sequences for the D1/D2 domains of large-subunit rRNA, small-subunit rRNA, translation elongation factor-1α, and subunits B1 and B2 of RNA polymerase II show the proposed novel species to be distinct from other species of the Ambrosiozyma clade. Because of their placement in the Ambrosiozyma clade, Candida kashinagacola, Candida llanquihuensis, Candida maleeae, Candida pseudovanderkliftii and Candida vanderkliftii are reassigned to the genus Ambrosiozyma as new combinations, and the description of the genus Ambrosiozyma is emended to reflect the resulting changes in phenotypic characters.

Four strains of a novel heterothallic yeast species were isolated from rotten wood collected in or near the Pilis Mountains in Hungary. The strains produced riboflavin in liquid culture. Analysis of gene sequences for the D1/D2 domains of the LSU nuclear rRNA, as well as analysis of concatenated gene sequences for the D1/D2 nuclear LSU rRNA, mitochondrial SSU rRNA and cytochrome oxidase II placed the novel species in a small clade including only two recognized species, Candida santjacobensis and Candida transvaalensis, in the family Trichomonascaceae. DNA sequence analyses demonstrated that the novel species was distinct from all currently recognized teleomorphic yeast genera. The name Diddensiella caesifluorescens gen nov., sp. nov. is proposed to accommodate the novel genus and species. The new genus proposed here can be recognized only from gene sequence analysis, because the characters of its asexual reproduction and ascospore formation are shared by several members of the genera Trichomonascus, Sugiyamaella and Spencermartinsiella. The type and isotype strains of D. caesifluorescens are NCAIM Y.01949T ( = NRRL Y-48781T = CBS 12613T) and NCAIM Y.01956I ( = NRRL Y-48782I = CBS 12614I), respectively. In view of their close relatedness to D. caesifluorescens, C. santjacobensis and C. transvaalensis are transferred to the genus Diddensiella as new combinations in accordance with changes in the International Code of Nomenclature for algae, fungi and plants.

Citeromyces hawaiiensis sp. nov. (NRRL Y-11581T  = CBS 12303T, type strain) is described from 12 strains isolated from flux of the sandalwood (Myoporum sandwicense) and adjacent soil in Hawaii, USA. Analyses of gene sequences from the D1/D2 domains of nuclear large subunit rRNA, internal transcribed spacer (ITS), mitochondrial small-subunit rRNA and translation elongation factor-1α each separated the proposed novel species from Citeromyces matritensis and Citeromyces siamensis, the other known species of the genus Citeromyces. The three species are morphologically similar but they can be separated by growth reactions in standard assimilation tests. An additional strain of Citeromyces siamensis (NRRL Y-11788), a species previously known only from Thailand, was obtained from spoiled condensed milk in Ohio, USA.

Analysis of sequences of the D1/D2 domain of the large subunit rRNA gene placed the novel species in the genus Spencermartinsiella. The novel species can be distinguished from Spencermartinsiella europaea, the single species of the genus, and from Candida cellulosicola, the only recognized anamorphic species of the Spencermartinsiella clade, on the basis of standard phenotypic characteristics. The relatedness among the four strains of the novel species and two closely related strains representing undescribed yeast species is discussed. The name Spencermartinsiella ligniputridi sp. nov. is proposed to accommodate the four novel strains. The type and isotype strains of Spencermartinsiella ligniputridi sp. nov. are NCAIM Y.01992T ( = CBS 12585T = NRRL Y-48818T) and NCAIM Y.01936I ( = CBS 12586I = NRRL Y-48819I), respectively. Two additional strains are NCAIM Y.01991 and NCAIM Y.01993.

Four strains of a novel yeast species were isolated from laboratory nests of the leaf-cutting ant Atta sexdens in Brazil. Three strains were found in older sponges and one was in a waste deposit in the ant nests. Sequencing of the D1/D2 region of the large-subunit rRNA gene showed that the novel species, named Sympodiomyces attinorum sp. nov., is phylogenetically related to Sympodiomyces parvus. Unlike Sympodiomyces parvus, Sympodiomyces attinorum can ferment glucose, assimilate methyl α-d-glucoside, salicin and citrate, and grow at 37 °C, thus enabling these two species to be distinguished. Differentiation from other related species is possible on the basis of other growth characteristics. The type strain of Sympodiomyces attinorum is UNESP-S156T (=CBS 9734T=NRRL Y-27639T).

Isolates of Pneumocystis carinii f. sp.hominis were examined from six individuals who died ofP. carinii pneumonia between 1968 and 1981 and who had underlying immunodeficiencies which were not due to human immunodeficiency virus infection. DNA sequence variation was analyzed in the genes encoding the mitochondrial large subunit rRNA (mt LSU rRNA), the internal transcribed spacer (ITS) regions of the nuclear rRNA, the arom locus, and the mitochondrial small subunit rRNA. No major variations were observed when these isolates were compared to isolates from HIV-infected individuals. A small number of minor differences were detected. A new position at which variation occurred in the mt LSU rRNA was observed in one sample. Three new ITS sequence types were identified. A total of nine different ITS sequence types were found in the six samples. Mixed infection with different ITS sequence types of P. carinii f. sp. hominis was observed in four of the six samples. The ITS locus was the most informative of the four loci for distinguishing among the isolates ofP. carinii f. sp. hominis. The data suggest that isolates of P. carinii f. sp. hominis from before the AIDS pandemic are genetically very similar to those currently found in HIV-infected individuals.

ABSTRACT First characterized in Trypanosoma brucei, the spliced leader-associated (SLA) RNA gene locus has now been isolated from the kinetoplastids Leishmania tarentolae and Trypanosoma cruzi. In addition to the T. brucei SLA RNA, bothL. tarentolae and T. cruzi SLA RNA repeat units also yield RNAs of 75 or 76 nucleotides (nt), 92 or 94 nt, and ∼450 or ∼350 nt, respectively, each with significant sequence identity to transcripts previously described from the T. brucei SLA RNA locus. Cell fractionation studies localize the three additional RNAs to the nucleolus; the presence of box C/D-like elements in two of the transcripts suggests that they are members of a class of small nucleolar RNAs (snoRNAs) that guide modification and cleavage of rRNAs. Candidate rRNA-snoRNA interactions can be found for one domain in each of the C/D element-containing RNAs. The putative target site for the 75/76-nt RNA is a highly conserved portion of the small subunit rRNA that contains 2′-O-ribose methylation at a conserved position (Gm1830) in L. tarentolae and in vertebrates. The 92/94-nt RNA has the potential to form base pairs near a conserved methylation site in the large subunit rRNA, which corresponds to position Gm4141 of small rRNA 2 in T. brucei. These data suggest that trypanosomatids do not obey the general 5-bp rule for snoRNA-mediated methylation.

Abstract A nematode recovered from syconia of Ficus hirta from Guangzhou, P. R. China, during a survey of nematode biodiversity from 2007 to 2009, is described herein as Schistonchus hirtus n. sp. and is differentiated by a combination of morphological characters, including excretory pore (EP) located near the metacorpus, a short post-uterine sac (PUS) (0.5 vulval body diam. (VBD) long), rose thorn-shaped spicules, amoeboid sperm, absence of gubernaculum, three pairs of subventral papillae on the male tail, host-Ficus and host-wasp species and DNA sequence data. Morphologically, S. hirtus n. sp. is close to S. centerae, S. altermacrophylla, S. aureus, S. laevigatus and S. virens based upon the length of the PUS (about 0.5 VBD long). However, the relative position of the EP in S. hirtus n. sp. is very different from these species (near metacorpus vs near head). With regard to the EP character, S. hirtus n. sp. is very similar to S. macrophylla, S. guangzhouensis and S. caprifici where the EP is at metacorpus level. However, S. hirtus n. sp. differs from S. macrophylla and S. guangzhouensis by possessing a shorter PUS and smaller spicules, and differs from S. caprifici by a shorter female stylet and smaller spicules. Schistonchus hirtus n. sp. was easily differentiated from other sequenced species by the proportion of parsimony informative changes in the partial small subunit rRNA gene (SSU) and D2/D3 expansion segments of the large subunit rRNA gene (LSU). Phylogenetic analysis with SSU sequences suggests that S. hirtus n. sp. is in a highly supported monophyletic clade with Aphelenchoides and Laimaphelenchus and is polyphyletic to other sequenced Schistonchus species. With LSU sequence data, it forms a clade with S. caprifici and they appear polyphyletic relative to S. guangzhouensis, S. centerae, S. aureus, S. laevigatus and S. virens.
