Herbiconiux moechotypicola sp. nov., a xylanolytic bacterium isolated from the gut of hairy long-horned toad beetles, Moechotypa diphysis (Pascoe)

A novel Gram-positive, non-motile, rod-shaped bacterium, designated strain RB-62T, was isolated during a study of culturable bacteria from the gut of Moechotypa diphysis (Pascoe) and its taxonomic position was investigated. Strain RB-62T grew at 15–30 °C and pH 5.0–8.5. The isoprenoid quinones were menaquinones MK-11 (77.1 %), MK-10 (11.7 %) and MK-12 (11.2 %). The major cellular fatty acids were anteiso-C15 : 0 (34.6 %), anteiso-C17 : 0 (29.8 %), iso-C16 : 0 (17.0 %) and cyclohexyl-C17 : 0 (11.4 %). The diagnostic diamino acid of the cell-wall peptidoglycan was 2,4-diaminobutyric acid. The G+C content of the genomic DNA of strain RB-62T was 70.3 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain RB-62T was affiliated with the genus Herbiconiux cluster within the family Microbacteriaceae, and was related most closely to Herbiconiux ginsengi wged11T (98.08 % similarity). The level of DNA–DNA relatedness between strain RB-62T and H. ginsengi wged11T was 43.2 % (reciprocal 66.7 %). Phenotypic and phylogenetic characteristics clearly distinguished strain RB-62T from recognized species of the genus Herbiconiux. Based on data from the present polyphasic study, strain RB-62T is considered to represent a novel species of the genus Herbiconiux, for which the name Herbiconiux moechotypicola sp. nov. is proposed. The type strain is RB-62T ( = KCTC 19653T = JCM 16117T).

In the context of studying the effects of transgenic fructan-producing potatoes on the community structure of phyllosphere bacteria, a group of strains closely related to the species Leifsonia ginsengi was isolated. Phylogenetic analysis based on 16S rRNA gene sequences revealed that the new isolates and L. ginsengi DSM 19088T formed a lineage at the genus level and this finding was supported by chemotaxonomic characterization. The peptidoglycan type of the representative isolate, K134/01T, and L. ginsengi DSM 19088T was B2γ, with d- and l-diaminobutyric acid as the diagnostic diamino acid and glycine, alanine and threo-3-hydroxyglutamic acid. The almost-complete substitution of glutamic acid by threo-3-hydroxyglutamic acid supported the differentiation of the new strains from recognized species of the genus Leifsonia. Furthermore, the detection of substantial amounts of the fatty acid cyclohexyl-C17 : 0 in the new isolates and L. ginsengi DSM 19088T was a prominent chemotaxonomic feature for a clear demarcation of these strains from all genera of the family Microbacteriaceae that display the B2γ cell-wall type. Comparative phylogenetic and phenotypic analyses of the isolates and L. ginsengi DSM 19088T revealed the separate species status of the isolates. On the basis of these results, it is proposed that L. ginsengi should be classified as the type species of a novel genus, Herbiconiux gen. nov., with the name Herbiconiux ginsengi gen. nov., comb. nov. (type strain wged11T = CGMCC 4.3491T = JCM 13908T = DSM 19088T = NBRC 104580T). The phyllosphere isolates are assigned to a novel species, Herbiconiux solani sp. nov. (type strain K134/01T = DSM 19813T = LMG 24387T = NBRC 106740T).

A novel strain of the class Actinobacteria was isolated from a seawater sample collected in the South China Sea using modified R2A agar plates. The strain was a Gram-stain-positive, non-motile, non-spore-forming, catalase-positive, irregular rod-shaped bacterium. The strain grew at 4–45 °C and pH 5.0–10.2, and tolerated 5 % (w/v) NaCl. Based on its 16S rRNA gene sequence, the organism was related phylogenetically to members of the genera Fodinibacter (96.7 % similarity), Lapillicoccus (96.5 %), Knoellia (95.0–95.8 %), Oryzihumus (95.6 %) and Humibacillus (95.6 %). The cell-wall contained l-ornithine as the major diagnostic diamino acid in the peptidoglycan. MK-8(H4) was the predominant menaquinone. Major cellular fatty acids were iso-C18 : 1 ω9c, iso-C16 : 0, iso-C15 : 0 and C17 : 0. The G+C content of the DNA was 69.6 mol%. Phenotypic and phylogenetic data revealed that this strain represents a novel species in a new genus of the family Intrasporangiaceae, for which the name Ornithinibacter aureus gen. nov., sp. nov. is proposed; the type strain of Ornithinibacter aureus is HB09001T (=CGMCC 1.10341T =DSM 23364T).

Strain JC268T was isolated from pebbles collected from a dam located in Lalitpur, Uttar Pradesh, India. Cells of strain JC268T were coccoid, appeared in pairs/triads/tetrads or short chains and were Gram-stain-positive, non-spore-forming, non-motile and obligately aerobic. Strain JC268T was catalase- and oxidase-positive and utilized citrate for growth. The genomic DNA G+C content of strain JC268T was 65.3 mol%. The cell-wall peptidoglycan contained l-lysine–l-serine–d-aspartic acid as interpeptide bridge with the type A4α. The major menaquinone was MK-8(H4). Major (>10 %) fatty acids were iso-C16 : 0, iso-C16 : 1H and anteiso-C17 : 1ω9c. Diphosphatidylglycerol, phosphoglycolipid, phosphatidylinositol, glycolipid, four unidentified lipids, an amino lipid and phospholipid were the polar lipids of strain JC268T. EzTaxon-e blast search of 16S rRNA gene sequences showed that strain JC268T has highest similarity to Barrientosiimonas humi 39T (98.65 %) and Tamlicoccus marinus MSW-24T (97.8 %) of the family Dermacoccaceae. Genome reassociation (based on DNA–DNA hybridization) of strain JC268T with Barrientosiimonas humi CGMCC 4.6864T ( = 39T) and T. marinus KCTC 19485T ( = MSW-24T) yielded values of 32.5 ± 2 % and 27.3 ± 2 %, respectively. Based on the data from phylogenetic and polyphasic taxonomic analyses, strain JC268T represents a novel species of the genus Barrientosiimonas for which the name Barrientosiimonas endolithica sp. nov., is proposed. The type strain of Barrientosiimonas endolithica is JC268T ( = KCTC 29672T = NBRC 110608T). Our data suggest that T. marinus should be reclassified within the genus Barrientosiimonas. Thus, a reclassification is proposed for T. marinus, the type and only species of the genus Tamlicoccus, as Barrientosiimonas marina comb. nov., which implies the emendation of the description of the genus Barrientosiimonas.

A Gram-reaction-negative, strictly aerobic, motile, rod-shaped bacterium, designated strain BZ78T, was isolated from soil from an industrial site. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain BZ78T belonged to the family Rhodospirillaceae and formed a coherent cluster with the type strain of Tistrella mobilis (98.3 % pairwise similarity). The predominant cellular fatty acids of strain BZ78T were C18 : 1ω7c (58.3 %), C19 : 0ω8c cyclo (11.5 %), C18 : 1 2-OH (10.9 %) and C14 : 0 3-OH (6.4 %). The predominant ubiquinone was Q-10. The genomic DNA G+C content of strain BZ78T was 65.8 mol%. On the basis of phenotypic characteristics, phylogenetic analysis and DNA–DNA relatedness data, strain BZ78T is considered to represent a novel species of the genus Tistrella, for which the name Tistrella bauzanensis sp. nov. is proposed. The type strain is BZ78T ( = DSM 22817T  = CGMCC 1.10188T  = LMG 26047T).

A novel psychrotolerant, Gram-positive, yellow-pigmented, aerobic bacterium, strain 0549T, was isolated from the China No. 1 glacier. The cells of the isolate were catalase-positive, motile, irregular rods. The diamino acid content of the cell-wall peptidoglycan was determined to be 2,4-diaminobutyric acid. Strain 0549T was able to grow at 4–27 °C, with optimum growth occurring at 20–22 °C. The major fatty acids were anteiso-C15 : 0, anteiso-C15 : 1, iso-C16 : 0 and anteiso-C17 : 0. The genomic DNA G+C content was 67 mol%. A phylogenetic analysis based on 16S rRNA gene sequences indicated that strain 0549T belonged to the genus Cryobacterium and exhibited 96.5 % similarity to Cryobacterium psychrophilum JCM 1463T. On the basis of the phenotypic, chemotaxonomic and phylogenetic properties and the DNA–DNA relatedness data, strain 0549T represents a novel species of the genus Cryobacterium, for which the name Cryobacterium psychrotolerans is proposed. The type strain is 0549T (=CGMCC 1.5382T=JCM 13925T).

A strictly aerobic, Gram-stain-negative, yellow-pigmented, non-spore-forming, motile (by gliding), rod-shaped bacterium, designated strain 15F3T, was isolated from leaf-and-branch compost. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain 15F3T was most closely related to Flavobacterium reichenbachii WB 3.2-61T and formed a distinct phyletic lineage within the genus Flavobacterium, the type genus of the family Flavobacteriaceae. Growth was observed at 10–34 °C (optimum, 30 °C) and pH 6.0–8.0 (optimum, pH 7.0). No growth occurred in the presence of ≥2 % (w/v) NaCl. Strain 15F3T reduced nitrate to nitrogen and showed catalase activity but no oxidase activity. The predominant cellular fatty acids were iso-C15 : 0 and summed feature 3 (comprising C16 : 1ω7c and/or iso-C15 : 0 2-OH). The major isoprenoid quinone was menaquinone-6. The G+C content of the genomic DNA was 31.1 mol%. On the basis of data from this polyphasic study, strain 15F3T may be classified as a representative of a novel species within the genus Flavobacterium, for which the name Flavobacterium banpakuense sp. nov. is proposed; the type strain is 15F3T ( = KACC 14225T  = JCM 16466T).

A Gram-stain-positive, ovoid to short rod-shaped, yellow-pigmented bacterium, designated strain DY66T, was isolated from tidal-flat sediment collected from Deukryang Bay (Republic of Korea), and its taxonomic position was investigated by using a polyphasic approach. Strain DY66T grew optimally at 30 °C and pH 8–9 and in 2 % (w/v) NaCl. The peptidoglycan type was A4α, l-Lys–l-Ala–d-Glu, and tyvelose and glucose were the major cell-wall sugars. The predominant menaquinones were MK-10 and MK-9. Major cellular fatty acids (>10 % of total) were anteiso-C15 : 0 and iso-C15 : 0. The polar lipids were diphosphatidylglycerol, phosphatidylcholine, phosphatidylglycerol, an unknown aminophospholipid and three unknown aminolipids. The DNA G+C content was 59.1 mol%. This chemotaxonomic profile supported the assignment of strain DY66T to the genus Zhihengliuella. Phylogenetic analysis based on 16S rRNA gene sequences also indicated that strain DY66T belonged to the family Micrococcaceae and was related to the genus Zhihengliuella. On the basis of the evidence presented in this study, strain DY66T represents a novel species of the genus Zhihengliuella, for which the name Zhihengliuella aestuarii sp. nov. is proposed. The type strain is strain DY66T ( = KCTC 19557T  = JCM 16364T).

A novel actinobacterium, designated strain C4-31T, was isolated from soil collected from a cave. Cells were aerobic, Gram-reaction-positive, oxidase-negative, catalase-positive and non-motile cocci. Comparison of 16S rRNA gene sequences showed that the organism occupied a distinct phylogenetic position within the suborder Frankineae, with sequence similarity values of less than 93.2 % to members of this suborder. The diagnostic diamino acid in the cell-wall peptidoglycan was meso-diaminopimelic acid. The major menaquinone was MK-9(H4). The polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, phosphatidylinositol mannoside, an unknown aminophospholipid and an unknown phospholipid. The major fatty acids were iso-C16 : 0, C17 : 1ω6c and C16 : 0. The G+C content of the DNA was 62.8 mol%. On the basis of morphological and chemotaxonomic data as well as phylogenetic evidence, strain C4-31T ( = KCTC 39556T = DSM 100065T) is considered to represent the type strain of a novel species of a new genus in the suborder Frankineae, for which the name Antricoccus suffuscus gen. nov., sp. nov. is proposed.

A Gram-staining-negative, non-gliding, orange-pigmented bacterial strain, designated HMF2925T, was isolated from fresh water in Korea. The phylogenetic tree based on 16S rRNA gene sequences showed that strain HMF2925T formed a distinct lineage within the genus Emticicia. Strain HMF2925T was closely related to Emticicia oligotrophica DSM 17448T (95.5 %) and Emticicia ginsengisoli Gsoil 085T (94.1 %). The major fatty acids of strain HMF2925T were summed feature 3 (C16 : 1ω6c and/or C16 : 1ω7c), iso-C15 : 0, C16 : 1ω5c and C16 : 0.The major polar lipids of strain HMF2925T were phosphatidylethanolamine, phosphatidylinositol, diphosphatidylglycerol, phosphatidylglycerol, an unidentified glycolipid, two unidentified amino lipids and three unidentified polar lipids. The DNA G+C content of strain HMF2925T was 36.5 mol%. On the basis of the evidence presented in this study, strain HMF2925T represents a novel species of the genus Emticicia, for which the name Emticicia aquatica sp. nov. is proposed. The type strain is HMF2925T ( = KCTC 42574T = CECT 8858T).

A Gram-stain-positive, aerobic, non-motile, psychrophilic bacterium, designated strain Cr6-08T, was isolated from alpine glacier cryoconite. Growth of strain Cr6-08T occurred at 1–25 °C. Phylogenetic analyses based on 16S rRNA gene sequences revealed that strain Cr6-08T is most closely related to members of the genus Arthrobacter. Strain Cr6-08T possessed chemotaxonomic properties consistent with those of the genus Arthrobacter, such as peptidoglycan type A3α (l-Lys–l-Ala4), MK-9(H2) as major menaquinone and anteiso- and iso-branched compounds (anteiso-C15 : 0 and iso-C15 : 0) as major cellular fatty acids. The polar lipid profile contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, one unknown glycolipid and three unknown polar lipids. The genomic DNA G+C content of strain Cr6-08T was 57.3 mol%. On the basis of phenotypic and chemotaxonomic characteristics, phylogenetic analysis and DNA–DNA relatedness data, strain Cr6-08T is considered to represent a novel species of the genus Arthrobacter, for which the name Arthrobacter cryoconiti sp. nov. is proposed. The type strain is Cr6-08T ( = DSM 23324T  = LMG 26052T  = CGMCC 1.10698T).
