AbstractThree bacterial species pertaining to the red complex (Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia) and Aggregatibacter actinomycetemcomitans were investigated in relation to the incidence and severity of periodontal disease. A total of 259 patients were included in this study, 179 being diagnosed with periodontal disease. The gingival crevicular fluid samples were obtained from periodontal pockets and the presence and levels of target bacteria were assessed following DNA extraction and real-time quantitative PCR. Our results account for significant positive associations between the number of bacterial species from the red complex coexisting within a patient and several clinical signs (gingival bleeding, inflammation and bone deterioration). A similar positive association was found between bacterial load of the red complex species and the clinical Case Type diagnostic of the periodontal disease, as well as the probing depth with the most evident results for T. denticola. In conclusion, our study, a first for the Romanian population, confirms previous results found elsewhere and finds a possible regional pathogenic specificity for T. denticola as a major factor for periodontitis severity.

These bacteria are usually found together in periodontal pockets, suggesting that they may cause destruction of the periodontal tissue in a cooperative manner. This article discusses the interspecies pathogenic interactions within the red complex.

Objectives. The study aimed to detect the presence/quantification of the 12 bacterial strains more frequently involved in the occurrence and aggravation of periodontal disease in a group of patients. Material and method. The study included 55 patients of both sexes, from the portfolio of the „Dr. Vodiță“ Dental Clinic, with chronic marginal periodontitis, who had not been under regular dental control and hygiene for at least 2 years. During the first treatment session, fluid was collected from each patient from the periodontal bags using sterile paper cones from the collection putty. Subsequently, fluid-soaked paper cones from periodontal bags were sent to the Genetic Lab in Bucharest for DNA extraction and quantification of 12 bacterial species, more common in the etiology of periodontal disease. For the correctness of the results obtained, it is necessary that patients have not taken antibiotics in the last 3 weeks. The data were analyzed and statistically processed with the Microsoft Excel 2016 program. Results. The least common bacteria found were from the species: Capnocytophaga ochracea, Aggregatibacter actinomycetemcomitans, Eikanella corrodens and Campylobacter rectus, closely followed by Capnocytophaga gingivalis, Prevotella intermedia and Capnocytophaga sputigena. The rest of the bacterial species were detected much more frequently. Discussions. In the analyzed group there was no monoinfection with Aggregatibacter actinomycetemcomitans. Porphyromonas gingivalis had an important presence in the studied group, registering increased levels in 89% of cases. In our group, Treponema denticola was present in increased amounts in 76.36% of cases. Tannerella forsythia was present in 92.72% of cases in elevated concentrations. Eikanella corrodens and Campylobacter rectus were present in 18.18% of the cases studied in high quantities. Prevotella intermedia was present in significant amounts in 34.54% of cases. Fusobacterium nucleatum was present in high concentrations in 98.18% of the cases studied. Prevotella nigrescens was present in increased amounts in 56.36% of cases. Capnocytophaga ochracea was present in 9.09% of cases in high amounts, Capnocytophaga sputigena in 38.18% of cases, and Capnocytophaga gingivalis was detected in 24.45% of cases. Conclusions. From the class of bacteria with high pathogenicity, we most frequently encountered in the studied group Tannerella forsythia. From the class of bacteria with moderate pathogenicity, we encountered the most common Fusobacterium nucleatum. From the class of bacteria with low pathogenicity, we encountered the most common Capnocytophaga sputigena. In order to validate the results obtained, it is necessary to extend the study to a larger number of patients.

Porphyromonas gingivalis and Tannerella forsythia have been identified as pathogenic bacteria associated with periodontal disease. Also, there are reports describing possible association of periodontal disease with other systemic diseases and some studies have revealed the presence of these bacterial species or their genomic content in sites distal to oral cavity in human body. There have been various studies investigating on possible pathogenic mechanisms that these bacteria device in healthy and disease conditions. Majority of the studies have been focused on possible molecular mechanisms and investigating the end products that are released as a result of bacteria coming into contact with the human tissues/ cells. Up to date only a limited number of studies have visualized direct histo-pathological and cellular events of these bacteria with time lapse information. In this mini review we tried to narrow down our focus on the visualization P. gingivalis and T. forsythia, starting from the oldest possible publications to current advancements, their contribution on understanding the histo-morpho-pathological changes during bacterial invasion, the pros and cons in these methods and how future research should be shaped to reveal the possible translocation of these bacteria from periodontal tissues to distal sites of the human body.

Введение. Условия космического полета приводят к изменению функциональных показателей многих органов и систем организма человека. Накопленный опыт показывает, что с увеличением длительности полета большое значение приобретают комплексные исследования состояния зубочелюстной системы. Цель исследования - изучение анаэробной составляющей микробиоты, иммунологических показателей ротовой жидкости и скорости кровотока в микроциркуляторном русле пародонта при моделировании невесомости. Методика. Наиболее адекватной моделью, воспроизводящей эффекты факторов полета в земных условиях, является «сухая» иммерсия. Было проведено комплексное обследование 20 мужчин добровольцев, в возрасте от 19 до 26 лет. В качестве модели микрогравитации использовали 5-суточную «сухую» иммерсию. Изучение состояния тканей пародонта включало микробиологические, иммунологические и микроциркуляторные исследования. Исследования выполняли в 1-е сут (фон), на 5-е сут иммерсии (выход) и на 7-е сут по окончании иммерсии. Качественный состав основных пародонтопатогенных видов микроорганизмов определяли методом полимеразной цепной реакцией. Концентрацию иммуноглобулинов (sIgA, IgA, IgМ) в ротовой жидкости - методом ИФА с помощью соответствующих наборов реагентов ЗАО «Вектор-Бест». Оценку кровотока в тканях пародонта проводили методом высокочастотной ультразвуковой допплеровской флуометрии ультразвуковым допплерографом «Минимакс-Допплер-К» (г. Санкт-Петербург) с датчиком непрерывного излучения, рабочая частота - 20 МГц. Результаты. Во время эксперимента в составе микробиоты пародонта обнаружено появление 5 основных пародонтопатогенных видов микроорганизмов (Aggregatibacter actinomycetemcomitans, Tannerella forsythia, Prevotella intermedia, Porphyromonas gingivalis, Treponema denticola). Отмечен рост концентрации в десневой жидкости sIgA, что связано с появлением пародонтопатогенной микробиоты. На 5-е сут и 7-е сут после «сухой» иммерсии у 12 человек отмечено статистически значимое снижение скорости кровотока в микроциркуляторном русле тканей пародонта как нижней, так и верхней челюсти. У 6 человек наблюдалась морфофункциональная перестройка микроциркуляторного русла в виде дилатации емкостных сосудов (венул) с частичным обеднением капиллярного звена, что свидетельствует о реакции венулярной компоненты на моделируемую микрогравитацию. Заключение. Комплексное исследование показало предрасположенность тканей пародонта в условиях «сухой» иммерсии к развитию воспалительных заболеваний и снижению барьерных функций пародонта.Comprehensive evaluation of 20 male volunteers aged 19-26 was conducted during exposure to 5-day “dry” immersion (DI). The aim was to examine periodontal tissues in the conditions of 5-day “dry” immersion, specifically, to study the anaerobic component of periodontal microbiota, immunological parameters of the oral fluid, and blood flow velocity in the periodontal microcirculation (PMC). Methods. Microgravity was modeled with the 5-day DI. Evaluation of the condition of periodontal tissues included microbiological, immunological, and microcirculatory studies. The studies were performed on day 1 (baseline), day 5 of immersion (completion), and day 7 upon the end of immersion. All studies were performed in the fasting state, before toothbrushing, in the supine position in the immersion bath, and in the sitting position 7 days after the immersion. PMC was studied at the same time points as in the immunoglobulin study. Blood flow in periodontal tissues was measured using the high-frequency ultrasonic Doppler fluorimetry (HFUDF) with a Minimax-Doppler-K ultrasonic dopplerograph (St. Petersburg) with a continuous radiation sensor at an operating frequency of 20 MHz. Results. Five major periodontopathogenic bacteria (Aggregatibacter actinomycetemcomitans, Tannerella forsythia, Prevotella intermedia, Porphyromonas gingivalis, and Treponema denticola) emerged as a part of the periodontal microbiota during the experiment. An increased concentration of sIgA was observed in the gingival fluid, which was associated with the emergence of periodontopathogenic microbiota. On day 5 of SI and day 7 following SI, 12 subjects had a statistically significant decrease in blood flow velocity in PMC of both the lower and upper jaws. 6 subjects showed morphological and functional rearrangement of the PMC, including dilation of capacitance vessels (venules) with partial capillary depletion, which indicated a venular component response to the conditions of modeled microgravity. Conclusion. This comprehensive study showed predisposition of periodontal tissues to development of inflammatory diseases and impaired functioning of the periodontal barrier under the conditions of DI.

Изучены синергия штаммов саливарного стрептококка BLIS K12 и M18 в ходе профилактики воспаления ротоглотки, а также возможности инвазии протективной микрофлоры в процессе межличностного общения в замкнутом пространстве искусственной среды обитания. Впервые в клинической практике изучено влияние пробиотиков на основе саливарного стрептококка на пародонтопатогены I и II порядков: Prevotella intermedia, Tanerella forsythia, Treponema denticola, Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis. Полученные результаты можно расценивать как положительные. По результатам наземного эксперимента «SIRIUS-19» планируется проводить закупки для космических экспериментов с шифрами «Пародонт» и «Пародонт-ЛОР».
