The Box-Behnken design was used to investigate the effect of three independent variables – time, temperature and solvent-to-solid ratio on the responses of total phenolics, total flavonoids, 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and cupric ion reducing antioxidant capacity (CUPRAC) of date fruit methanolic extracts. Response surface analysis showed that the optimal ultrasound extraction parameters that maximized the responses were 30 min, 298 K and 74.4 ml/g. Under optimum conditions, UHPLC-DAD-MS/MS was used to tentatively characterize 11 phenolic compounds. The experimental values for the quantification of phenolic compounds and antioxidant activities are in accordance with the predicted values, indicating the suitability of the model and the success of response surface methodology in optimizing the ultrasound extraction conditions.

AbstractSeveral methods have been described for the determination of phenolic compounds in animal and plant products using the Folin-Ciocalteu (FC) assay. Most of these methods describe the use of this reagent and sodium carbonate in spectrophotometric methods. The macro FC assay was compared with two micro FC assays carried out on a microplate reader. Excellent correlation was obtained among the three assays with a molar extinction coefficient of 5.228±0.187x103 M−1 cm−1. The micro assay may serve as a high throughput method for the rapid determination of polyphenols in various samples.

This experiment was conducted to determine the equations relating the Hue digital values of the fruits surface of the oil palm with maturity stage of the fruit in plantation. The FFB images were zoomed and captured using Nikon digital camera, and the calculation of Hue was determined using the highest frequency of the value forR,G, andBcolor components from histogram analysis software. New procedure in monitoring the image pixel value for oil palm fruit color surface in real-time growth maturity was developed. The estimation of day harvesting prediction was calculated based on developed model of relationships for Hue values with mesocarp oil content. The simulation model is regressed and predicts the day of harvesting or a number of days before harvest of FFB. The result from experimenting on mesocarp oil content can be used for real-time oil content determination of MPOB color meter. The graph to determine the day of harvesting the FFB was presented in this research. The oil was found to start developing in mesocarp fruit at 65 days before fruit at ripe maturity stage of 75% oil to dry mesocarp.
