Effect of Extended Galvanonarcosis on Behavior of Rainbow Trout, Salmo gairdneri, and Channel Catfish, Ictalurus punctatus

Yearling rainbow trout, Salmo gairdneri, and channel catfish, Ictalurus punctatus, were subjected to 0.5- and 3-h periods of narcosis using continuous direct current. Galvanonarcosis treatments decreased the immediate individual and group activity in both species of fish. Twenty-four hours after treatment, all fish except trout narcotized for 3 h recovered to levels of individual and group activity equivalent to those of untreated fish. Photonegative response was strong in both fish species and was not influenced by galvanonarcosis treatments. Trout narcotized with direct current were more vulnerable to predation than untreated fish; susceptability increased with increasing treatment periods. The potential of extended galvanonarcosis as a fish immobilization technique is strong. Rainbow trout (Salmo gairdneri) homozygous for liver lactate dehydrogenase alleles ldhHαA and ldhHαB were artificially propagated and their swimming stamina compared. The time required to fatigue 50% of the HαAHαA phenotypes in fixed water velocity tests was 2.3 times greater on the average than that of HαAHαB phenotypes.Likewise, LDH phenotypes HαAHαA, HαAHαB, and HαAHαB of steelhead trout from the Thompson River were artifically propagated and their swimming stamina compared. In contrast to the rainbow trout, significant differences in stamina among the three phenotypes of steelhead were not evident in the stocks from this river nor between phenotypes HαAHαB and HαBHαB from another stream, the Vedder River, which has a very low frequency of the ldhHαA allele. The stamina of young steelhead from the Thompson River was, however, 3.8 times greater than that of those from the Vedder River. The pharmacokinetics, bioavailability, and disposition of oxolinic acid (OA), a quinolone antibacterial drug, were examined in a warmwater (channel catfish (Ictalurus punctatus)) and coldwater fish species (rainbow trout (Oncorhynchus mykiss)). Studies were performed at 24 and 14 °C for catfish and at 14 °C for trout to compare temperature- and species-related differences. Analysis of OA pharmacokinetics when determined by HPLC provided elimination half-life [Formula: see text], volume of distribution (Vss), and clearance (Clb) estimates for 24 °C catfish, 14 °C catfish, and 14 °C trout of 40.9, 69.3, and 81.3 h, 939, 880, and 1817 mL/kg, and 16.3, 8.9, and 16.9 mL∙kg∙h−1, respectively. Following oral administration, OA plasma concentrations peaked between 8 and 24 h for all treatments; however, 14 °C trout and 14 °C catfish sustained peak concentrations for a longer duration than 24 °C catfish. Oral OA bioavailabilities were 56.0, 91.8, and 90.7% for 24 °C catfish, 14 °C catfish, and 14 °C trout, respectively. OA distribution data for muscle of 14 °C catfish demonstrated an inordinately high peak concentration and delayed time to peak relative to other treatments. Elimination half-lives of OA in muscle were 33.1, 54.3, and 141 h for 24 °C catfish, 14 °C catfish, and 14 °C trout, respectively.

Exposure of rainbow trout (Salmo gairdneri) to 5.7 × 10−10 M (5 × 10−5 mg/L) morpholine during smoking period did not influence the subsequent neural response to this substance, when tested immediately and 12 mo after smoking. No significant difference in the morpholine-induced responses was obtained between the exposed and unexposed fish. Using N-ethylmaleimide, a sulfhydryl reagent, it was further confirmed that the morpholine-effect is caused by a mechanism not directly associated with the normal olfactory function. Perfusion of the olfactory organs with 1.0 × 10−1 and 1.1 × 10−1 M (1%) morpholine for 10 min irreversibly inhibited the olfactory responsiveness to odorants. Sensitization of the olfactory bulbar electrical response is discussed in connection with homing of morpholine-imprinted salmonid fishes. Rainbow trout (Salmo gairdneri), west-slope cutthroat trout (Salmo clarki), and their F1 hybrids were electrophoretically examined for 13 different proteins. Of the 13 proteins, only phosphoglucose isomerase (PGI) proved to be a reliable biochemical marker for the distinction of genetically pure samples of the two species. Only PGI provided an accurate means of identifying F1 hybrids between the species. This information could possibly be used to facilitate the stabilization and restoration of native populations of west-slope cutthroat trout in western Montana. The effects of mercury and copper on the olfactory response of rainbow trout (Salmo gairdneri) were studied by perfusing HgCl2 and CuSO4 solutions through the olfactory organs while recording the olfactory bulbar electrical responses to the standard stimulant, L-serine.The olfactory response was depressed during exposure to mercury and copper. The lowest concentrations of mercury and copper needed to cause appreciable effects within 2 h were estimated at 0.10 and 0.008 mg/liter, respectively. The depression increased with increase in the concentration and exposure time of the heavy metals. Recovery of the olfactory response was slower with higher concentrations and longer exposure.The method is rapid, accurate, and sensitive, and can be applied to other toxic chemicals and fish species.

The upstream orientation and frequency of upstream movement of ripe male rainbow trout (Salmo gairdneri) in the presence of ovarian fluid obtained from spawning females of the same species were observed and quantified. In a stream tank, male trout moved upstream in the ovarian fluid odor plume. In an optomotor tank, male trout exhibited an increase in the frequency of upstream orientation and movement in the presence of diffuse, nondirectional ovarian fluid. It is concluded that water currents provide directional cues and olfactory stimuli regulate the orientational and kinetic components of the rheotropic response. The significance of this mechanism of orientation in fish migration is discussed. Key words: rheotropism, behavior, olfaction, orientation, migration, ovarian fluid

To standardize the sealed-jar bioassay for optimum sensitivity to whole bleached kraft pulpmill effluent, experiments were designed to assess the effects of test temperature, acclimation temperature, and fish species. Coho salmon (Oncorhynchus kisutch) conformed to a previously recognized test paradigm by showing progressively decreased oxygen utilization with increasing toxicant concentrations; however, rainbow trout (Salmo gairdneri) did not. In concentrations of effluent < 1.5 LC50, rainbow trout used significantly more oxygen than control groups, while coho salmon used significantly less oxygen. The sensitivity of these responses was influenced by test temperature and acclimation temperature. Results for coho confirmed that this species was most sensitive to effluent when tested at ambient room temperature. Both species showed significantly decreased oxygen utilization in effluent concentrations > 1.5 LC50 irrespective of test temperature or acclimation temperature. The significance of these responses is discussed, and the applicability of sealed-jar bioassays for assessing the acute toxicity of pulpmill effluents is reviewed. <em>Abstract.-</em>Demographic studies have indicated that, since 1990, Kentucky’s urban population has experienced an 18% increase while its rural population has expanded by less than 1%. These demographic changes, coupled with a decline in fishing license sales, focused the need to develop and increase fishing opportunities in and around urban areas. In 2005, the Kentucky Department of Fish and Wildlife Resources initiated a pilot urban fishing program aimed at providing urban residents with high quality fishing opportunities close to home. Five pilot lakes, located within four of Kentucky’s most populated counties, were selected to receive regular stockings of rainbow trout <em>Oncorhynchus mykiss</em>, channel catfish <em>Ictalurus punctatus</em>, largemouth bass <em>Micropterus salmoides</em>, and hybrid bluegill <em>Lepomis macrochirus </em>x <em>L. cyanellus</em>. Rainbow trout were stocked at each lake in April and October. Channel catfish, largemouth bass, and hybrid bluegill were stocked in May and June. Monthly stocking rates were 202 trout/ha, 34 channel catfish/ha, 12 largemouth bass/ha, and 81 hybrid bluegill/ha. Creel and angler attitude surveys were conducted at two of the pilot lakes from April through August 2006 to determine angler catch and satisfaction. Although fishing pressure at these two lakes was high, most anglers practiced catch and release. Catch rates were 1.03 fish/h at Stein Lake and 0.79 fish/h at Upper Sportsman’s Lake. Angler attitude survey results indicated that the majority of anglers interviewed were not aware of the program and did not know that regular fish stockings were occurring at the lakes. The typical angler fishing these lakes was a white male between the ages of 30 and 49, who was satisfied with his fishing experience regardless of the number or size of fish caught. Further studies need to be completed to compare results from this study, and to refine stocking and marketing techniques used in the program.
