AbstractStimulus images can be reconstructed from visual cortical activity. However, our perception of stimuli is shaped by both stimulus-induced and top-down processes, and it is unclear whether and how reconstructions reflect top-down aspects of perception. Here, we investigate the effect of attention on reconstructions using fMRI activity measured while subjects attend to one of two superimposed images. A state-of-the-art method is used for image reconstruction, in which brain activity is translated (decoded) to deep neural network (DNN) features of hierarchical layers then to an image. Reconstructions resemble the attended rather than unattended images. They can be modeled by superimposed images with biased contrasts, comparable to the appearance during attention. Attentional modulations are found in a broad range of hierarchical visual representations and mirror the brain–DNN correspondence. Our results demonstrate that top-down attention counters stimulus-induced responses, modulating neural representations to render reconstructions in accordance with subjective appearance.

Abstract Speech production requires temporal coordination between the actions of different functional groupings of muscles in the human body. Crucially, such functionally organized units, or “modules”, may be susceptible to disruption by an external stimulus such as a startling auditory stimulus (SAS; >120dB), enabling a possible window into the internal structure of learned speech movements. Following on the observation that SAS is known to accelerate the release of pre-planned actions, the current study examines lip kinematics in SAS-induced responses during speech movements to test whether this accelerated release applies on the scale of entire syllables or on the scale of smaller functional units. Production measures show that SAS-elicited bilabial movements in [ba] syllables are prone to disruption as measured by discontinuity in velocity profiles. We use a 3D finite element method (FEM) biomechanical model to simulate the temporal interaction between muscle groupings in speech. Simulation results indicate that this discontinuity can be accounted for as an instance of temporally decoupled coordination across neuromuscular modules. In such instances, the muscle groupings controlling lip compression and jaw opening, which normally fire sequentially, appear more likely to be activated synchronously.

Most flowering plants must defend themselves against herbivores for survival and attract pollinators for reproduction. Although traits involved in plant defence and pollinator attraction are often localised in leaves and flowers, respectively, they will show a diffuse evolution if they share the same molecular machinery and regulatory networks. We performed RNA-sequencing to characterise and compare transcriptomic changes involved in herbivory-induced defences and flower development, in tomato leaves and flowers, respectively. We found that both the herbivory-induced responses and flower development involved alterations in jasmonic acid signalling, suppression of primary metabolism and reprogramming of secondary metabolism. We identified 411 genes that were involved in both processes, a number significantly higher than expected by chance. Genetic manipulation of key regulators of induced defences also led to the expression changes in the same genes in both leaves and flowers. Targeted metabolomic analysis showed that among closely related tomato species, jasmonic acid and α-tomatine are correlated in flower buds and herbivory-induced leaves. These findings suggest that herbivory-induced responses and flower development share a common molecular machinery and likely have coevolved in nature.

Penicillium digitatum is the main postharvest pathogen of citrus fruit. Although the inner fruit peel part (albedo) is less resistant than the outer part (flavedo) to P. digitatum, the global mechanisms involved in their different susceptibility remain unknown. Here, we examine transcriptome differences between both tissues at fruit harvest and in their early responses to infection. At harvest, not only was secondary metabolism, involving phenylpropanoids, waxes, and terpenoids, generally induced in flavedo vs. albedo, but also energy metabolism, transcription factors (TFs), and biotic stress-related hormones and proteins too. Flavedo-specific induced responses to infection might be regulated in part by ERF1 TF, and are related to structural plant cell wall reinforcement. Other induced responses may be related to H2O2, the synthesis of phenylpropanoids, and the stress-related proteins required to maintain basal defense responses against virulent pathogens, whereas P. digitatum represses some hydrolase-encoding genes that play different functions and auxin-responsive genes in this peel tissue. In infected albedo, the repression of transport and signal transduction prevail, as does the induction of not only the processes related to the synthesis of flavonoids, indole glucosinolates, cutin, and oxylipins, but also the specific genes that elicit plant immunity against pathogens.

Abstract Iron (Fe) is an essential micronutrient whose uptake is tightly regulated to prevent either deficiency or toxicity. Cadmium (Cd) is a non-essential element that induces both Fe-deficiency and toxicity; however, the mechanisms behind these Fe/Cd-induced responses are still elusive. Here we explored Cd and Fe-associated responses in wildtype Arabidopsis and in a mutant that over-accumulates iron (opt3-2). Gene expression profiling revealed a large overlap between transcripts induced by Fe deficiency and Cd exposure. Interestingly, the use of opt3-2 allowed us to identify additional gene clusters originally induced by Cd in wildtype but repressed in the opt3-2 background. Based on the high levels of H2O2 found in opt3-2 we propose a model where reactive oxygen species prevent the induction of genes that are induced in wildtype by either Fe deficiency or Cd. Interestingly, a defined cluster of Fe-responsive genes was found to be insensitive to this negative feedback, suggesting that their induction by Cd is more likely the result of an impaired Fe sensing. Overall, our data suggest that Fe-deficiency responses are governed by multiple inputs and that a hierarchical regulation of Fe homeostasis prevents the induction of specific networks when Fe and H2O2 levels are elevated.

SummarySerosurveillance is an important epidemiologic tool for SARS-CoV-2, used to estimate burden of disease and degree of population immunity. Which antibody biomarker, and the optimal number of biomarkers, has not been well-established, especially with the emerging rollout of vaccines globally. Here, we used random forest models to demonstrate that a single spike or receptor-binding domain (RBD) antibody was adequate for classifying prior infection, while a combination of two antibody biomarkers performed better than any single marker for estimating time-since-infection. Nucleocapsid antibodies performed worse than spike or RBD antibodies for classification, but is of utility for estimating time-since-infection, and in distinguishing infection-induced from vaccine-induced responses. Our analysis has the potential to inform the design of serosurveys for SARS-CoV-2, including decisions regarding number of antibody biomarkers measured.
