FORMATION OF POLYTHIONATES AND THEIR INTERRELATIONSHIPS DURING OXIDATION OF THIOSULFATE BY THIOBACILLUS FERROOXIDANS

The metabolic pathway for the utilization of thiosulfate by the acidophilic chemoautotroph, Thiobacillus ferrooxidans, has been examined by chromatographic and radiographic techniques. With suitable chemical controls, it was found that the major pathway involved 2 moles of thiosulfate passing to tetrathionate to trithionate regenerating 1 mole of thiosulfate concurrent with the production of 2 moles of sulfate. Thus, previous findings which demonstrate this pathway as far as trithionate, made with other thiobacilli, are sound despite recent documentation of the chemical interaction of various polythionates. The cyclical pathway, proposed here, explains some findings of other workers.

There are two strains of bacteria viz. Thiobacillus thiooxidansand Thiobacillus ferrooxidanswidely mentioned to play an important role in the leaching process of low-grade ores. Another strain used in this study is a thermophile and is designated Caldariella .These microorganisms are acidophilic chemosynthetic aerobic autotrophs and are capable of oxidizing many metal sulfides and elemental sulfur to sulfates and Fe2+ to Fe3+. The necessity of physical contact or attachment by bacteria to mineral surfaces during oxidation reaction has not been fairly established so far. Temple and Koehler reported that during oxidation of marcasite T. thiooxidanswere found concentrated on mineral surface. Schaeffer, et al. demonstrated that physical contact or attachment is essential for oxidation of sulfur.

SummaryThe analysis of normal human plasma by fibrin autography revealed four species of plasminogen activator (PA) activity related to tissue-type PA, factor XII, prekallikrein and urokinase-type PA (u-PA). The u-PA activity increased significantly by incubating plasma with dextran sulfate. This increase was coincident with both the cleavage of factor XII and the complex formation of activated factor XII with its plasma inhibitors, which were determined by immunoblotting procedure. The dextran sulfate-dependent activation of u-PA required both factor XII and prekallikrein, but did not require either plasminogen or factor XI. High molecular weight kininogen was required only at a low concentration of dextran sulfate. Thus the results indicate that the factor XII and prekallikrein-mediated activation of single chain u-PA (scu-PA) operates as a major pathway of scu-PA activation in whole plasma in contact with dextran sulfate.

Enterobacter aerogenes AY-2 mutant is known for hydrogen gas producer which ws obtained from the sludge of methane fermentation and the yield is 1.5 fold higher than wildtype. Hydrogen gas production can be gain via NADH oxidation in anaerobic metabolic pathway by blocking organic acid production. Metabolic pathway can be changed by mutagenesis. Enterobacter aerogenes AY-2 mutated with ethyl methane sulfonate in logarithmic phase with consentration 10, 11, 12, 13, 14 and 15 μl/ml cell suspention during 120 minute. Mutation that result lowest survival ratio (0,01%) was 14 μl EMS/ml cell suspention is repeated with variation incubation time, 30, 60, 90 and 120 minute. 166 double mutant colony has been collected and choosen randomly. The choosen 43 colony was fermented in glycerol complex medium for determining ten double mutant with the highest H2 production. Double mutant AD-H43 is a highest H2 producer that increase 20% H2 production from AY-2 and has a decrease lactid acid production, 31% less from AY-2. Increasing H2 production in double mutant AD-H43 is caused by lactate dehydrogenase deffi cient.Keywords: Enterobacter aerogenes AY-2, ethyl methane sulfonate (EMS), H2 and methane sludge

Administration of an antiviral drug, acyclovir via the oral route leads to low and variable bioavailability (15-30%). Therefore, this research work was aimed to enhance bioavailability of acyclovir by nanocrystallization technique. The drug nanocrystals were prepared by anti-solvent precipitation method in which different stabilizers were used. The formed nanocrystals are subjected to biopharmaceutical characterization including solubility, particle size and in-vitro release. SEM studies showed nano-crystals were crystalline nature with sharp peaks. The formulated drug nanocrystals were found to be in the range of 600-900nm and formulations NC7 and NC8 showed marked improvement in dissolution velocity when compared to pure drug, thus providing greater bioavailability. FT-IR and DSC studies revealed the absence of any chemical interaction between drug and polymers used.
