Winter survival for thousands of species of insects relies on adaptive strategies for cold hardiness. Two basic mechanisms are widely used (freeze avoidance by deep supercooling and freeze tolerance where insects endure ice formation in extracellular fluid spaces), whereas additional strategies (cryoprotective dehydration, vitrification) are also used by some polar species in extreme environments. This review assesses recent research on the biochemical adaptations that support insect cold hardiness. We examine new information about the regulation of cryoprotectant biosynthesis, mechanisms of metabolic rate depression, role of aquaporins in water and glycerol movement, and cell preservation strategies (chaperones, antioxidant defenses and metal binding proteins, mitochondrial suppression) for survival over the winter. We also review the new information coming from the use of genomic and proteomic screening methods that are greatly widening the scope for discovery of genes and proteins that support winter survival.

We investigated the hibernation ecology and cold hardiness of hatchling diamondback terrapins, Malaclemys terrapin (Schoepf, 1793), an estuarine species that reaches 42°N along the Atlantic Ocean. During 3 years of study, about 50% of the nests we monitored harboured hatchlings during winter, and the majority (87%) of these individuals survived despite being intermittently exposed to subfreezing temperatures. Most such exposures were brief (ca. 12 h) and mild (minimum temperature: ca. –1.2 °C); however, turtles were occasionally subjected to longer chilling episodes and lower temperatures. In laboratory experiments, hatchlings supercooled extensively, attaining ca. –15 °C before spontaneously freezing. However, they were highly susceptible to inoculative freezing through contact with external ice and (or) ice-nucleating agents, which occur in nesting soil. Therefore, freeze avoidance through supercooling does not appear to be a viable cold-hardiness strategy in these turtles. Hatchlings subjected to experimental freezing survived exposure to temperatures as low as –3.0 °C, suggesting that freeze tolerance may account for the high winter survival observed in natural nests. We conclude that freeze tolerance in hatchling M. terrapin is promoted by high susceptibility to inoculation, which is known to moderate freezing, allowing cells time to adapt to the attendant physical and osmotic stresses.

Loss of freeze tolerance, or deacclimation, is an integral part of winter survival in woody perennials because untimely mid-winter or spring thaws followed by a hard freeze can cause severe injury to dehardened tissues. This study was undertaken to investigate deacclimation kinetics, particularly the timing and speed, of five blueberry (Vaccinium L.) cultivars (`Bluecrop', `Weymouth', `Ozarkblue', `Tifblue', and `Legacy'), with different germplasm compositions and mid-winter bud hardiness levels, in response to an environmentally controlled temperature regime. Based upon bud cold hardiness evaluations in 2000 and 2001, `Tifblue', a Vaccinium ashei Reade cultivar, was one of the least hardy and the fastest to deacclimate; `Bluecrop', a predominantly V. corymbosum L. cultivar, was the most hardy and the slowest to deacclimate; and `Ozarkblue', a predominantly V. corymbosum cultivar but including southern species V. darrowi Camp. and V. ashei, was intermediate in speed of deacclimation. `Weymouth' (predominantly V. corymbosum) and `Legacy' (73.4% V. corymbosum and 25% V. darrowi) were slow to intermediate deacclimators. Deacclimation rates did not correlate strictly with mid-winter bud hardiness. Data suggest that the southern germplasm component V. ashei may be responsible for the observed faster deacclimation whereas both southern species, V. darrowi and V. ashei, may contribute genes for cold sensitivity. Strong positive correlations between stage of bud opening and bud cold hardiness existed in both years (r = 0.90 and 0.82 in 2000 and 2001 study, respectively). Previously identified major blueberry dehydrins, 65-, 60-, and 14-kDa, progressively decreased in their abundance during incremental dehardening in `Bluecrop', `Weymouth', and `Tifblue'. However, down-regulation of the 14-kDa dehydrin most closely mirrored the loss in cold hardiness during deacclimation, and, therefore, may be involved in regulation of bud dehardening. Because differences in deacclimation rate were clearly evident among the genotypes studied, rate of deacclimation of the flower buds of blueberry should be an important consideration in breeding to improve winter survival.

This article gives an overview of the current state of cold hardiness research in fruit crops by reviewing the recently published studies on cold hardiness of both tree fruit and berry crops. Topics discussed include cold hardiness of fruit species, cultivars and different plant organs, biophysical and biochemical aspects of hardiness, evaluation of hardiness, as well as endogenous, cultural and environmental factors affecting cold hardiness in these species. Lack of cold hardiness is a major limiting factor for production of fruit crops in many regions of the world and improved cold hardiness one of the major objectives in numerous breeding programs and research projects. Screening cultivars or selections for cold hardiness is commonly done, and different methods applied to the evaluation of hardiness are discussed. The physical limit of deep supercooling may be a restricting factor for expanding the production of some fruit crops, such as Prunus species and pear. As for biochemical aspects, a relationship between carbohydrates and cold hardiness is most commonly found. Studies have also been made on different hardiness modifying cultural factors including rootstock, crop load, raised beds and application of growth regulators. The latter seems promising for some species. Cold hardiness is an extremely complex phenomenon and understanding different mechanisms involved is critical. Since hardiness is, however, primarily affected by genotype, developing cold-hardy fruit cultivars and effective screening methods for hardiness are essential. Finally, cultural practices may be improved to further enhance hardiness. Key words: Berries, cold hardiness, fruits, small fruits, stress, winter hardiness

AbstractIn Kings County, Nova Scotia, low temperatures in the coldest nights of winter can differ by as much as 10°F. from one area to another. This has an important bearing on winter survival of some arthropods. Overwintering sites of orchard arthropods range from exposed situations which remain at air temperature to well protected ones on the ground where temperatures rarely go below 20°F. The cold-hardiness of each of 24 species of arthropods was measured: seven were sufficiently cold-hardy to survive any winter conditions in Nova Scotia, five were less cold-hardy but overwinter in well protected sites and twelve had marginal cold-hardiness, their mortality varying with the winter and the locality.

At the colder peach production regions it is important to know the cold hardiness of peach cultivars at different phenological stages of flowering. In our experiment, artificial freezing tests were conducted in a climate chamber in five selected years between the period of 2007 and 2016 to determine the freeze tolerance of generative organs of three peach cultivars (‘Venus’, ‘Redhaven’, ‘Piroska’) at different phenological stages of bloom. Based on the results of the laboratory freezing tests LT<sub>50</sub> values were calculated. Our results showed that LT<sub>50</sub> values of examined peach cultivars in swelled bud stage averaged over five years were between –6.8 and –11.2°C according to cultivar, and as phenological phases progressed, cold hardiness of generative organs decreased. At the end of bloom LT<sub>50</sub> values varied between –1.7 and –4.1°C. Cultivar ‘Piroska’ had the highest freeze tolerance and cultivar ‘Venus’ showed the lowest in each year studied. This study shows that trees with delayed development are more prone to cold damage to flowers.

Abstract Background Survival to cold stress in insects living in temperate environments requires the deployment of strategies that lead to physiological changes involved in freeze tolerance or freeze avoidance. These strategies may consist of, for instance, the induction of metabolic depression, accumulation of cryoprotectants, or the production of antifreeze proteins, however, little is known about the way such mechanisms are regulated and the signals involved in their activation. Ascarosides are signaling molecules usually known to regulate nematode behavior and development, whose expression was recently found to relate to thermal plasticity in the Japanese pine sawyer beetle Monochamus alternatus. Accumulating evidence also points to miRNAs as another class of regulators differentially expressed in response to cold stress, which are predicted to target genes involved in cold adaptation of insects. Here, we demonstrate a novel pathway involved in insect cold acclimation, through miRNA-mediated regulation of ascaroside function. Results We initially discovered that experimental cold acclimation can enhance the beetle’s cold hardiness. Through screening and functional verification, we found miR-31-5p, upregulated under cold stress, significantly contributes to this enhancement. Mechanistically, miR-31-5p promotes production of an ascaroside (asc-C9) in the beetle by negatively targeting the rate-limiting enzyme, acyl-CoA oxidase in peroxisomal β-oxidation cycles. Feeding experiments with synthetic asc-C9 suggests it may serve as a signal to promote cold acclimation through metabolic depression and accumulation of cryoprotectants with specific gene expression patterns. Conclusions Our results point to important roles of miRNA-mediated regulation of ascaroside function in insect cold adaptation. This enhanced cold tolerance may allow higher survival of M. alternatus in winter and be pivotal in shaping its wide distribution range, greatly expanding the threat of pine wilt disease, and thus can also inspire the development of ascaroside-based pest management strategies.
