Insecticidal activity of culture filtrates from liquid medium of Beauveria bassiana isolates from South Sumatra (Indonesia) wetland soil against larvae of Spodoptera litura. Biodiversitas 20: 2101-2109. The obstacle in utilizing entomopathogenic fungi to control pest insects in wetlands is the inability of their isolates to survive during the saprophytic phase due to the soil being very acidic. Therefore, the exploration of fungi being able to survive in the acidic soil was utilized in the acidic ecosystem. This study aimed to test the toxic activity of Beauveria bassiana culture filtrates from liquid medium at pH 2.5 and 6 against Spodoptera litura larvae. Total of ten isolates collected from South Sumatra, Indonesia was grown in liquid media at pH of 2.5 and 6 for six weeks. The result showed that the culture filtrate from liquid medium at pH 6 was more toxic against the larvae than that at pH 2.5. The mortality of the larvae at pH 6 reached 92%, while that at pH 2.5 it reached 13.33% only. The findings of present study revealed that the culture filtrate isolates of BPdR, BJgTs, BSwTd2, BSwTd3, BSwTd4, BKKPp2 from pH 2.5 media still caused high larvae mortality (6.67-13.33%). These seven isolates are superior due to their toxic nature at pH 2.5. Consequently, present study has increased the chances of success in utilizing these isolates for biological control in acidic ecosystems, such as peatlands, tidal lowlands, and freshwater swamps.

This research reported the in vitro pathogenicity of Verticillium lecanii strains, L2 and L5, and Beauveria bassiana strains, B76 and B252, against Megoura japonica using leaf-dip method. The virulence potential of these four entomopathogenic fungal strains of V. lecanii and of B. bassiana were compared between fungi conidia (concentrations 1 × 106, 1 × 107, and 1 × 108 conidia mL−1) and culture filtrate. Moreover, binary combination of four different fungal strains (L2 + B76, B76 + L5, L2 + B252, and L2 + B76 + B252 + L5) were evaluated against M. japonica under control condition. Aphid mortality was recorded after two, four, six, and eight days of post-treatment. In the conidial concentration bioassay, strain B76 showed maximal mortality (85.3%) against bean aphid, and strain L5 showed the lowest effect (60.0%) at the highest concentration (1 × 108 conidia mL−1) at eight days post-treatment. Whereas, binary combinations of B76 and L2 strains showed the highest effect against M. japonica (90.5%) than other combinations. Moreover, in comparison with the effect of filtrate and conidia bioassay, 91.4% and 84.1% were achieved in strain B76, and the lowest effect (63.8% and 55.1%) was recorded in strain L5.

<p>The influence of certain nitrogen compounds - components of glucose-peptone medium (GPM) on the accumulation of carotenoids by some strains was investigated by surface cultivating basidiomycetes. The total carotenoid content was set in acetone extracts of mycological material spectrophotometrically and calculated using the Vetshteyn formula.</p> <p>As the nitrogen-containing components used GPM with 9 compounds, such as peptone, DL-valine, L-asparagine, DL-serine, DL-tyrosine, L-proline, L-alanine, urea, NaNO<sub>3</sub>. The effect on the accumulation of specific compounds both in the mycelium and in the culture fluid of carotenoids by culturing certain strains of Basidiomycetes was identified.</p> <p>Adding to standard glucose-peptone medium peptone at 5 g/l causes an increase of carotenoid accumulation by strain <em>L. sulphureus</em> Ls-08, and in a concentration of 4 g/l by strains of <em>F. hepatica </em>Fh-18 and <em>F. fomentarius</em> Ff-1201.</p> <p>In order to increase the accumulation of carotenoids in the mycelium  we suggested to make a standard glucose-peptone medium with proline or valine for cultivating of <em>L. sulphureus</em> Ls- 08 strain; alanine for <em>F. fomentarius</em> Ff-1201 strain; proline, asparagine and serine - for strain Fh-18 of <em>F. hepatica</em>. The results can be implemented in further optimization of the composition of the nutrient medium for culturing strains of Basidiomycetes wich producing carotenoids.</p> <p><em>Keywords: nitrogen-containing substances, Basidiomycetes, mycelium</em><em>,</em><em> culture filtrate, carotenoids</em></p>

El psílido de la papa Bactericera cockerelli afecta a las solanáceas en México, por el daño directo que causa, además, de que es transmisor de patógenos procariotes como Candidatus Liberibacter solanacerum (psyllaurous). Este insecto es combatido principalmente con productos químicos. Sin embargo, en la actualidad, se buscan nuevas estrategias para su manejo como el uso de hongos entomopatógenos. El objetivo de este trabajo fue evaluar la susceptibilidad del psílido de la papa a dos cepas comerciales y dos nativas de Metarizhium anisopliae y Beauveria bassiana, así como confirmar la identificación de las mismas mediante técnicas moleculares. Los bioensayos se realizaron con la cepa nativa MA25 de M. anisopliae y las comerciales Metabich® (MA28) y Bassianil® (BB09) y la nativa BB42 de Be. bassiana. Se probaron cinco concentraciones sobre ninfas del tercer estadio de B. cockerelli y se obtuvieron sus CL50 y CL95. Todas las cepas resultaron patógenas para el psílido de la papa, las más tóxicas fueron BB09, BB42 y MA28. La mortalidad fluctuó entre 90 a 100% con las concentraciones más altas de cada cepa: la más virulenta fue BB09, con una CL50 de 2,99 x 104 conidias mL-1 y MA25 la menos virulenta tuvo una CL50 de 6,34 x 105 conidias ml-1. Mediante la clonación y la secuenciación de la región ITS de los rADN 18S, se corroboró que las cepas nativas identificadas por morfología microscópica y claves taxonómicas corresponden a Be. bassiana y M. anisopliae.

En busca de una alternativa de control del mosquito Culex quinquefasciatus en áreas urbanas se desarrolló el proyecto: “Evaluación de microorganismos biocontroladores sobre el mosquito Culex quinquefasciatus” cuyo objetivo fue evaluar la capacidad biocontroladora de diferentes hongos entomopatógenos sobre larvas del mosquito a nivel de laboratorio. Se evaluaron 16 aislamientos de hongos en diferentes concentraciones. Se constataron diferencias en el porcentaje de mortalidad producido en larvas de Cx. quinquefasciatus dependientes del tipo de aislamiento, de la concentración utilizada y del tiempo de exposición. Los aislamientos que presentaron el nivel de mortalidad más alto pertenecen a la especie Metarhizium anisopliae, seguidos por aislamientos de la especie Beauveria bassiana. El aislamiento que presentó el mayor nivel de mortalidad (100%) en menor tiempo de exposición (24 horas) fue Metarhizium anisopliae 99068 en una concentración de 1x106 e/ml. Por lo tanto este aislamiento es recomendado para su evaluación en campo y desarrollo de un producto comercial.
