Optical Density of Subretinal Fluid as a Predictive Biomarker for Eyes With Chronic Central Serous Retinopathy Treated With Eplerenone

Purpose: To investigate how the optical density ratio (ODR) of subretinal fluid (SRF) obtained by optical coherence tomography (OCT) in eyes with chronic central serous retinopathy (CSR), defined as symptoms >3 months, treated with eplerenone correlates with treatment response and visual outcomes. Methods: This retrospective, single-center observational study included patients with chronic CSR treated with eplerenone who had at least 6 weeks of follow-up. Eyes with poor-quality OCT scans, shallow SRF precluding sampling of optical density (OD), and macular pathology other than CSR were excluded. Optical density of the vitreous and SRF was measured using ImageJ and used to calculate ODR (ODR = ODSRF/ODVIT). Peak SRF height, foveal SRF height, central macular thickness (CMT), and Snellen visual acuity (VA) were measured. The Mann-Whitney U test was used to compare continuous variables. Pearson correlation coefficient was used to determine the statistical significance of applied linear regressions. Results: Twenty-three eyes of 23 patients met the inclusion criteria. Six (26%) eyes had complete resolution of foveal SRF (resolvers) with eplerenone treatment, and the remainder of patients had worsening or incomplete resolution (nonresolvers). Resolvers had significantly lower baseline logarithm of the minimum angle of resolution (logMAR) VA ( P < .01) than nonresolvers. There was a positive linear correlation between ODR and logMAR VA at baseline ( R2 = 0.208; P = .03) and follow-up ( R2 = 0.206; P = .03). A significant correlation between ODR and percentage change in CMT was found ( R2 = 0.263; P = .01). Linear correlations were not found between ODR and age, peak SRF height, and foveal SRF height (all P > .125). Conclusion: Lower baseline ODR was associated with improved percentage change in CMT and VA in patients treated with eplerenone for chronic CSR. Optical density ratio may represent a novel prognostic biomarker that may help determine which eyes with chronic CSR may respond better to eplerenone.

Background:Rheumatoid arthritis (RA) is a chronic inflammatory disease characterized by increased mortality and associated with metabolic disorders. Since the metabolomic profile is known to vary in response to different inflammatory conditions, metabolome analysis could substantially improve diagnosis and prognosis of RA.Objectives:To analyze the urine metabolome profile in RA patients and correlate it with disease activity changes over 12 monthsMethods:Seventy-nine RA patients, according to ACR/EULAR 2010 classification criteria, between 40 and 70 years old, were recruited and followed for 12 months. Metabolome analysis was performed by Nuclear Magnetic Resonance spectroscopy (NMR), resulting in the identification of 93 metabolites in urine collected at the baseline and after 12 months. Frequency analysis, Pearson Correlation and Multivariate data analysis with orthogonal projections to latent structures (OPLS) method were performed and a statistical significance was considered as p<0.05.Results:The study population was characterized by the majority of women (86.7%), mean age of 56 years old, around 80% with positive anti-CCP or Rheumatoid Factor. During the one year of follow-up, there was no substantial variation in the DAS28 measurement (baseline: 3.8, after 12 months: 4.0). There was no significant correlation between the metabolome pattern and DAS28 score (p>0.05) over time. However, multivariate analysis (OPLS-DA) demonstrated an adequate differentiation of the population with 0.92 of accuracy (Q2: 0.72 and R2: 0.89).There was a significant increase of L-cysteine, choline, L-Phenylalanin, creatine, L-histidine, oxalacetic acid and xanthine, and a decrease of L-threonine, taurine, butyric and gluconic acid (p<0.05) during the follow-up, metabolites that are involved in the skeletal muscle metabolism.Conclusion:The observed biomarkers indicate,as expected, that the RA metabolic profile is associated with inflammation injury and skeletal muscle amino acid metabolism. Correlations with disease activity changes was compromised by the stable disease status during the 12 months. Characterization of modified humic substances based binders for iron ore agglomeration was examined by chemical analysis, optical density, Fourier transform infrared spectrum (FTIR), and thermogravimetry and differential scanning calorimetry (TG–DSC). Chemical analysis displays the proportion of fulvic acid (FA) to humic acid (HA) in the binder is 1:10. Compared with the HA, the FA possesses more functional groups. Meantime, optical density ratio analysis shows that the molecular weight and aromatization degree of the FA are smaller than those of the HA. FTIR spectra further confirm aromatic and aliphatic fractions are associated with various types of oxygen-rich groups including carboxyl and hydroxyl groups. TG–DSC and chemical analysis indicate structural changes of the binder including thermal decomposition, dehydroxylation and/or decarboxylation are caused during heating. The structural characterization of the binder ensures its good performance in the field of iron ore agglomeration.

ABSTRACT Introduction Ample radiopacity in order to distinguish from the surrounding tissues is a desirable property of dental graft materials. A total of 15 bone graft materials’ (BGMs) opacities were analyzed in this study. Materials and methods Graft materials were placed in the implant cavity (5 × 10 mm) in cadaver's mandible respectively. Cavity was exposed by using periapical film and a dental X-ray machine at 70 kVp and 8 mA. The optical density of the radiographic images was measured with a transmission densitometer. One-way analysis of variance (ANOVA) was conducted for statistical analysis. Results Among the materials tested, the most radiolucent bone grafts were Grafton and Allogenix with a statistical significance of p ≥ 0.05. 4Bone and Bego Oss exhibited the highest radiopacity with a statistical significance of p ≥ 0.05. Inadequate radiopacity of the dental graft materials may lead to confusion among clinicians in the radiographical follow-up. Among 15 BGMs tested, only three had higher density than bone tissue. In this study, an azimuthal-rotation sample holder compatible with scanning transmission X-ray microscopy was developed. This holder exhibits improvement in the accuracy of rotation angles and reduces the displacement of the rotation axes during azimuthal rotation by using a crossed roller bearing. To evaluate the performance of the holder, the authors investigated the dependence of the optical density around the C K-edge absorption of π-orbital-oriented domains in natural spherical graphite on the rotational angle by using linearly horizontally and vertically polarized undulator radiation. Based on the dependence of the optical density ratio between C 1s → π* and 1s → σ* excitation on the polarization angle of the X-rays, the average two-dimensional orientation angle of the π orbital in each position in a natural spherical graphite sample was visualized.

A new method is applied to the determination of percent oxyhemoglobin (percent saturation) in whole blood. The method is based on a theoretical treatment of light absorption and scattering by particulate suspensions and requires the measurement of optical densities (D) of blood at three closely spaced wavelengths. The optical density of red blood cell (RBC) suspensions was measured with a video microdensitometer and a linear relationship was found between percent saturation and the corrected optical density ratio, (D555 B)/(D546 B). For a given saturation, this ratio was independent of optical path length (12 mum-2 mm), hematocrit (3–50%), and RBC velocity (1.5–17 mm/s). The applicability to microvascular measurements has been assessed through the use of TV microdensitometry on micropipettes with flowing RBCs and on microvessels in the hamster cheek pouch.
