Estimating Production of Heterotrophic Bacterioplankton via Incorporation of Tritiated Thymidine

ABSTRACT Autoradiographic, enzymic and histologic studies on uteri of pregnant rats were carried out to follow the endometrial modifications which take place during progestation (days L0 – L4) and culminate in the state of uterine receptivity essential for ovum-implantation. Pulse labelling with tritiated thymidine (radioactive DNA precursor) on L0, L1 and L2 revealed a sequence of cell renewal in luminal and glandular epithelium and endometrial stroma. On L3 and L4 stromal cells showed extensive incorporation of tritiated thymidine. This synthetic activity was associated with endometrial preparation for decidualization and was evoked at least in part, by the surge of oestrogen on L3. All layers of the uterine wall were heavily infiltrated on L0 and resembled the site of an acute inflammatory reaction. Subsequently, polymorphonuclear infiltration diminished and monocytic cells predominated. On L3 a spatial arrangement was observed: eosinophiles were concentrated in the basal endometrium and monocytic cells in the subepithelial stroma. A comparison was made between such a shift in migratory cells in the uterus and similar phenomena which occur in inflammatory and immune reactions. Activities of acid and alkaline phosphatases, of ATP-ase and succinic dehydrogenase were low on L0 and L1 during the periods of infiltration, degeneration and regeneration of luminal and glandular epithelium. Enzymic activities increased on the following days, (L3 and L4). Vascular dilation and engorgement and endometrial oedema were observed near the blastocysts on L4. Most blastocysts incorporated tritiated thymidine after 14.00 h on L4, but some showed uptake before loss of the zona which occurs usually between 14.00 and 16.00 h; therefore, it was assumed that the permeability of the zona increases prior to being shed. Activities of succinic dehydrogenase and acid and alkaline phosphatases were demonstrable in blastocysts on L4 while they were still »free« in the uterine lumen.

Satellite cell activity was examined in the stretch-enlarge anterior latissimus dorsi muscle (ALD) of the adult quail. Thirty-seven birds had a weight equal to 10% of their body mass attached to one wing while the contralateral wing served as an intra-animal control. At various time intervals after application of the wing weight (from 1 to 30 days), the birds were injected with tritiated thymidine and killed 1 h later. Stretched muscle length was greater by day 1 and mass by day 3 when compared with the contralateral muscle. Satellite cells actively synthesizing DNA were quantitated in fiber segments of the control and stretched ALD. A minimum of 1,500 muscle nuclei (satellite cell nuclei and myonuclei) were counted in each muscle. Labeling in stretched muscle was expressed by the percent labeled nuclei per total nuclei counted. Satellite cell labeling was initiated by day 1, peaked between days 3 and 7, and was not statistically different from control values at day 30. These results demonstrate that satellite cells are induced to enter the cell cycle in the stretch-enlarged ALD muscle from the adult quail, and the peak of proliferative activity is within the first week of stretch.

Abstract Acute monoblastic leukemia (acute myeloid leukemia [AML], French-American-British type M5a) with leukemia cutis developed in a patient 6 weeks after the initiation of erythropoietin (EPO) therapy for refractory anemia with ringed sideroblasts. AML disappeared from both marrow and skin after the discontinuation of EPO. Multiparameter flow cytometric analysis of bone marrow cells demonstrated coexpression of the EPO receptor with CD45 and CD13 on the surface of blasts. The incubation of marrow cells with EPO, compared to without, resulted in 1.3- and 1.6-fold increases, respectively, in tritiated thymidine incorporation and bromodeoxyuridine incorporation into CD13+ cells. Clinical and laboratory findings were consistent with the EPO-dependent transformation of myelodysplastic syndrome (MDS) to AML. It is concluded that leukemic transformation in patients with MDS treated with EPO may be EPO-dependent and that management should consist of the discontinuation of EPO followed by observation, if clinically feasible.

SUMMARY Blastocysts were studied on the 5th and 8th day of pregnancy in lactating mice, in the fresh state, flushed from the uterus, in squash preparations and in serial sections. At the earlier period some mitosis was observed. Tritiated thymidine incorporation studies gave some evidence of DNA synthesis on the 5th and 6th days of pregnancy. By the 8th day the blastocysts were longer, contained more cells, and mitosis had ceased. They were located at the anti-mesometrial end of the uterine lumen, closely apposed to the uterine epithelium, and with their long axes parallel to the long axis of the uterine horn. Implantation could be induced, either by the removal of the litter, or by the injection of an appropriate dose of oestrogen on the 5th or 7th (but not the 4th) day of pregnancy. Both treatments were followed by the appearance of W-bodies in the neighbourhood of the blastocysts, the disappearance of the shed zonae, and the appearance of Pontamine Blue reactivity, oedema of the uterine stroma and formation of the primary decidual zone, in that order.
