Under the stress of ultraviolet radiation some cyanobacteria synthesize scytonemin, a protective pigment against DNA photodamage. In addition to photoprotection, scytonemin has been shown to have an anti-proliferative effect on various types of malignant cells. In this study the effect of scytonemin on melanoma and spleen cells was assessed both in vitro using tissue cultures and in vivo in mice models. Melanoma and spleen cells were exposed to 0.08 to 10 μM of scytonemin, and cell proliferation was measured using tritiated thymidine uptake. The data suggest that scytonemin acts as an inhibitor for melanoma cells in a concentration-dependent manner while enhancing the proliferation of spleen cells, suggesting that it can potentially augment the immune response. Furthermore, mice injected with melanoma cells and scytonemin produced fewer tumors than mice that did not receive scytonemin, although the data were not significant. This study adds to the growing body of research that scytonemin may be beneficial as a future anticancer agent to prevent tumor cell growth.

Skin cells are affected by UV-induced oxidative stress resulting in photoaging and diseases. The objective of this work was to assess the effect of an aqueous extract of Larrea divaricata(Aq) and a flavonoid rich fraction (EA), obtained by liquid/liquid fractionation, on the proliferation of keratinocytes and fibroblasts by the tritiated thymidine uptake assay and on cell viability by the trypan blue exclusion assay. The in vitro sun protection factor (SPF), peroxidase (Px)-like and superoxide dismutase (SOD)-like activities were determined by kinetic spectrophotometric assays, and the phytochemical composition was determined by HPLC-UV and HPLC MS/MS.Aq and EA induced keratinocytes and fibroblast proliferation, protected fibroblasts from H2O2-induced apoptosis and exerted SOD-like and Px-like activities. Aq and EA had a high sun UVB protection factor. So, L. divaricatacould be used in a future for the development of new dermocosmetic or phytotherapy adjuvant in skin oxidative damage.

Objectives. Interleukin-27 (IL-27) has been reported to inhibit type 2 T helper cell (Th2) response in allergic rhinitis (AR). However, its effects on group II innate lymphoid cells (ILC2) in AR are not fully understood. Methods. Nineteen patients with AR and nineteen controls were enrolled in this study. The effects of IL-27 on ILC2 differentiation and function as well as the regulation of the IL-27 receptor (IL-27R) were analyzed by tritiated thymidine incorporation, enzyme-linked immunosorbent assay (ELISA), and real-time polymerase chain reaction (PCR), respectively. AR mice were used to confirm the role of IL-27 in vivo. Results. The serum IL-27 protein expression in AR patients was significantly lower compared with controls. IL-27 decreased the ILC2 proliferation and type II cytokine secretion through the interaction with IL-27R. IL-27 also inhibited systemic and nasal ILC2 response of AR mice. Conclusion. IL-27 inhibited the proliferation and function of ILC2 in AR, implying that IL-27 may be used as new treatment target in AR.

Abstract Introduction AAES is a powdered form of Biostymina, herbal medicinal product of Phytopharm Klęka S.A., a water extract of Aloe arborescens Mill. leaves. Aloe arborescens Mill. (woody aloe, tree-like aloe) is known to have several traditional medicinal properties including anti-inflammatory, immunomodulatory, antiviral and antimicrobial activity. Objective The aim of this work was to study the in vivo effect of AAES on cellular (leukocyte-induced cutaneous angiogenesis, LIA test, and proliferative response to PHA) and humoral (anti-SRBC antibody response) immunity in mice. Methods Balb/c mice were fed AAES from 0.5 to 75 mg/kg body mass for seven days before grafting their splenocytes intradermally to F1 (Balb/cxC3H) recipients (LIA test). Neovascular reaction was evaluated 72 h later in dissection microscope. Spleen cell cultures were incubated with 0.5, 1 and 2 μg/ml of PHA. After 48 h of incubation, tritiated thymidine was added. After further 24 h, cells were harvested (Skatron) and incorporation of tritiated thymidine was measured using Beta-scintillation counter. Balb/c mice were fed for 7 days with AAES, then immunized intraperitoneally with 5% SRBC suspension and 7 days later the antibody response was measured with hemagglutination test. Results Neovascular reaction was significantly higher in groups grafted with splenocytes collected from all AAES fed donors than from the controls. The proliferation of splenocytes taken from mice fed AAES at doses ranging from 0.5 mg/kg to 7.5 mg/kg was stimulated in all cultures. Suppression of proliferation was observed in cell cultures derived from mice fed with higher doses of AAES. Stimulation of anti-SRBC antibody production was seen in mice fed both 2.5 and 7.5 mg/kg dose of AAES. Conclusion Powdered form of Biostymina (AAES) might be useful in the treatment of patients with ischaemia of tissues and organs (myocardial infarction, stroke, necrosis) and in deficiency in the production of immune cells and growth factors (infections, chronic wound healing, ulceration and bone fusion).

SummaryJapanese fancy mouse, mini mouse or pet mouse are common names used to refer to strains of mice that present with different colour varieties and coat types. Although many genetic studies that involve spotting phenotype based on the coat have been performed in these mice, there are no reports of quantitative data in the literature regarding testis structure and spermatogenic efficiency. Hence, in this study we researched testis function and spermatogenesis in the adult Japanese fancy mouse. The following values of 68 ± 6 mg and 0.94 ± 0.1% were obtained as mean testis weight and gonadosomatic index, respectively. In comparison with other investigated mice strains, the fancy mouse Leydig cell individual size was much smaller, resulting in higher numbers of these cells per gram of testis. As found for laboratory mice strains, as a result of the development of the acrosomic system, 12 stages of the seminiferous epithelium cycle have been described in this study. The combined frequencies of pre-meiotic and post-meiotic stages were respectively 24% and 64% and very similar to the laboratory mice. The more differentiated germ cell types marked at 1 h or 9 days after tritiated thymidine administration were preleptotene/leptotene and pachytene spermatocytes at the same stage (VIII). The mean duration of one spermatogenic cycle was 8.8 ± 0.01 days and the total length of spermatogenesis lasted 37.8 ± 0.01 days (4.5 cycles). A high number of germ cell apoptosis was evident during meiosis, resulting in lower Sertoli cell and spermatogenic efficiencies, when compared with laboratory mice strains.

β-Adrenergic receptors (β-ARs) and catecholamines are present in rodents as early as embryonic day (E)10.5. However, it is not known whether β-AR signaling plays any role in the proliferation and differentiation of ventricular cells in the embryonic heart. Here, we characterized expression profiles of β-AR subtypes and established dose-response curves for the nonselective β-AR agonist isoproterenol (ISO) in the developing mouse ventricular cells. Furthermore, we investigated the effects of ISO on cell cycle activity and differentiation of cultured E11.5 ventricular cells. ISO treatment significantly reduced tritiated thymidine incorporation and cell proliferation rates in both cardiac progenitor cell and cardiomyocyte populations. The ISO-mediated effects on DNA synthesis could be abolished by cotreatment of E11.5 cultures with either metoprolol (a β1-AR antagonist) or ICI-118,551 (a β2-AR antagonist). In contrast, ISO-mediated effects on cell proliferation could be abolished only by metoprolol. Furthermore, ISO treatment significantly increased the percentage of differentiated cardiomyocytes compared with that in control cultures. Additional experiments revealed that β-AR stimulation leads to downregulation of Erk and Akt phosphorylation followed by significant decreases in cyclin D1 and cyclin-dependent kinase 4 levels in E11.5 ventricular cells. Consistent with in vitro results, we found that chronic stimulation of recipient mice with ISO after intracardiac cell transplantation significantly decreased graft size, whereas metoprolol protected grafts from the inhibitory effects of systemic catecholamines. Collectively, these results underscore the effects of β-AR signaling in cardiac development as well as graft expansion after cell transplantation. NEW & NOTEWORTHY β-Adrenergic receptor (β-AR) stimulation can decrease the proliferation of embryonic ventricular cells in vitro and reduce the graft size after intracardiac cell transplantation. In contrast, β1-AR antagonists can abrogate the antiproliferative effects mediated by β-AR stimulation and increase graft size. These results highlight potential interactions between adrenergic drugs and cell transplantation.
