A study was carried out to compare the efficiency of vaginal electrical impedance with serum progesterone profile, cytological examination and clinical findings. Vaginal electrical impedance values varied at the different stages of the estrous cycle, with the highest value during the transition into estrous and the lowest in diestrus (P< 0.05). There was a negative statistical correlation between serum progesterone values and the impedance values (36%) (P< 0.001). Vaginal electrical impedance was a faster and cheaper method than progesterone assessment. It was more reliable than vaginal cytology and clinical evaluation. In conclusion, a combination of vaginal electrical impedance measurements and progesterone evaluation was a more useful method for determination of the optimal breeding time in bitches..

During the proestrous and estrous periods in 12 beagles, the following parameters were measured daily: the horizontal dimensions of the vulva, vaginal cytology, and serum luteinizing hormone (LH) and progesterone concentrations. Measurements of serum LH concentrations allowed for the identification of the LH surge and the optimal time for artificial insemination (AI). Nine out of the 12 beagles became pregnant through AI and completed a gestation. Shrinkage in the horizontal dimensions of the vulva (i.e., vulvar shrinkage) was primarily observed prior and subsequent to the LH surge. In six of the nine (66.7%) beagles that became pregnant, vulvar shrinkage was observed on the days in which the LH surge was confirmed, and the rate of vulvar shrinkage tended to be greater at higher serum LH concentrations. Further vulvar shrinkage was identified in all nine beagles within two days of the LH surge. An increase in the serum progesterone concentration was observed after the LH surge in each of the beagles that became pregnant, together with clinical signs of estrous behavior (i.e., standing heat) as well as a change in vulva condition from swollen to soft. This demonstrates that vulvar shrinkage is induced in response to the onset of the LH surge and that the LH surge can be predicted through the measurement of the horizontal dimensions of the vulva, vaginal cytology, and the assessment of serum progesterone concentrations in beagles.

The luteolytic action of prostaglandin F2α (PGF2α), 15-keto-PGF2α, 15-methyl-PGF2α, and cloprostenol was evaluated in cycling gilts and sows after intramuscular injection on day 13 of the estrous cycle. Only cloprostenol significantly shortened the mean cycle length (18.5 vs. 20.3 d, P < 0.05). Cloprostenol also caused a more rapid decline in serum progesterone concentrations than did the other prostaglandins. Serum concentrations of the prostaglandin metabolite 13,14-dihydro-15-keto-PGF2α (PGFM), showed rapid transitory peaks after PGF2α or 15-keto-PGF2α and a lower, later rise after cloprostenol. A second experiment examined luteal luteinizing hormone (LH) receptors and luteal membrane ultrastructure during the estrous cycle and pregnancy and the effect of cloprostenol on these parameters during the estrous cycle. The number of unoccupied luteal LH receptors, as measured by specific 125I-hCG binding, dropped significantly from mid to late pregnancy and from mid to late cycle. Cloprostenol lowered serum progesterone concentrations but did not affect hCG binding. X-ray diffraction showed no correlation of gel or liquid-crystalline phase lipids in luteal microsomes with the stage of the estrous cycle or pregnancy or cloprostenol treatment. Objectives The purpose of this study was to assess efficacy of deslorelin, a gonadotropin-releasing hormone (GnRH) agonist marketed in Europe for the control of male dog reproduction, for the postponement of puberty in queens. Methods Nine prepubertal queens aged 3–9 months were selected for this study; their general and reproductive health was checked through clinical, haematological, vaginal cytology and hormonal tests. Following treatment with a 4.7 mg deslorelin implant, each cat received a monthly clinical examination and blood was collected for hormonal assay every third month. Cats were monitored for 14.1 ± 5.2 (range 7–23) months. Results All cats were in good body condition and normal health prior to treatment. Their health status remained unchanged throughout the study and no significant variation was observed with regard to serum progesterone or oestradiol. Seven days post-treatment, 1/9 queens showed signs of heat, and one other queen showed complete vaginal keratinisation. No other signs of heat were subsequently observed in any other queen. Five queens were lost during the study after 7, 7, 16, 17 and 18 months of observation (during which time they did not show signs of heat). By the end of the study, no sign of puberty was observed in the four remaining queens at 21–36 months of age. Conclusions and relevance A 4.7 mg deslorelin implant was able to suppress the feline pituitary–gonadal axis, leading to postponement of puberty for up to 21–36 months in the four queens that completed the study. Deslorelin can be considered as a safe method to postpone puberty in queens.

Rat estrous cycle determination or vaginal smear staining is paramount for studies related to endocrinology and reproduction; in the present study; we have reported a simple and rapid method for estrous cycle determination in rats using crystal violet. With this technique, the identification of stages can be done even on the next day. Hormonal variation in blood and histomorphological changes in ovaries at different stages of the estrous cycle were studied in female Wistar rats, which can be used to determine the hormone levels in works related to hormonal drugs, further ovarian morphology can be used to study changes in ovaries during the estrous cycle. This study aims to report a rapid and simple method for vaginal cytology using crystal violet and to report normal hormonal levels and histomorphology of ovaries in various phases of the estrous cycle.
