Abstract. A superfusion model for isolated ovarian cells was developed and characterized in detail. Granulosa cells isolated from pre-ovulatory rat ovarian follicles were placed in superfusion (perifusion) chambers with a volume of 125 μl. Culture medium was pumped through the chambers, collected in 20-min fractions of 600 μl and analysed for cAMP and steroids. Viability was confirmed by morphological examination. The use of polycarbonate membranes to retain the cells in the chambers was abandoned since the membranes caused severe cell damage. The temporal relationships between gonadotropic stimuli and the release of cyclic 3':5'-adenosine monophosphate (cAMP) and steroids was investigated. Within 10 min FSH elicited transient increase in the release of cAMP and progesterone but had no effect on testosterone or estradiol-17β release. Amplitude and duration of the response in cAMP and progesterone release were correlated to concentration and length of the FSH pulse when these parameters were varied within the ranges 1–100 μg/l and 30–270 min, respectively. Compared with the cAMP response, the progesterone response peaked up to 30 min later and lasted 1 to 2 h longer but could not be extended to more than approximately 6 h, not even with longer FSH pulses. These results could indicate a development of desensitization.

This paper is an exploration of preksha dhyana as a case study of modern yoga. Preksha is a system of yoga and meditation introduced by Acarya Mahaprajna of the Jain Svetambara Terapanth in the late twentieth century. I argue that preksha is an attempt to join the newly emerging transnational yoga market whereby yoga has become a practice oriented around the attainment of physical health and psychological well-being. I will evaluate the ways in which Mahaprajna appropriates scientific discourse and in so doing constructs a new and unique system of Jain modern yoga. In particular, I evaluate the appropriation of physical and meditative techniques from ancient yoga systems in addition to the explanation of yoga metaphysics by means of biomedical discourse. I will demonstrate how, in Mahaprajna’s preksha system, the metaphysical subtle body becomes somaticized. In other words, Mahaprajna uses the bio-medical understanding of physiology to locate and identify the functions of metaphysical subtle body parts and processes in the physiological body.

Reproductive indices like gonad size, fecundity, egg size and sex steroid levels of estradiol-17β and testosterone, vitellogenin in the blood as well as bile conjugates and liver 7-ethoxyresorufin O-deethylase (EROD) activity were studied in populations of perch (Perca fluviatilis L.) and roach (Rutilus rutilus L.) and experimentally exposed juvenile whitefish (Coregonus lavaretus L. s.l.) at the Southern Lake Saimaa (S.E. Finland). Our studies showed that the introduction of elemental chlorine free (ECF) bleaching and secondary treatment of effluents in a modern activated sludge plant at the mill in 1992 have substantially reduced the exposure of feral and caged fish to organochlorines. The liver EROD activity was noticeably lower in feral and caged fish near the mill indicating lesser impacts on the liver mixed function oxygenase (MFO) system. However, at the mill site, liver EROD activies in feral and caged fish still tend to be one to four times higher than at the reference sites. Significantly decreased plasma estradiol-17β and testosterone concentrations in perch and roach in the period of development of the gonads (autumn and winter) indicate that there were endocrine disrupting compounds present in the lake receiving ECF pulp and paper mill effluents. Cause-effect relationships, however, are difficult to establish.
