<p>ABSTRAK</p><p>Nomuraea rileyi adalah salah satu jamur entomopatogen yangpotensial mengendalikan hama Helicoverpa armigera dan Spodopteralitura pada tanaman kapas, tembakau, dan jarak kepyar. Di lapanganpernah ditemukan larva hama H. armigera dan S. litura yang terinfeksisecara alami oleh N. rileyi yang mengindikasikan bahwa N. rileyiberpotensi sebagai agens hayati. Sebelum N. rileyi dikembangkan sebagaiagens hayati, maka perlu diketahui metode perbanyakannya pada mediabuatan. Penelitian bertujuan untuk mengetahui komposisi media tumbuhyang sesuai untuk perbanyakan N. rileyi dan pengujian patogenisitasnyaterhadap H. armigera dan S. litura. Penelitian dilakukan di LaboratoriumFitopatologi dan Laboratorium Patogen Serangga Balai PenelitianTanaman Tembakau dan Serat Malang mulai bulan Mei sampai denganNovember 2009. Penelitian ini terdiri atas 2 pengujian, yaitu pengujiankarakter biologi N. rileyi dan patogenisitas pada ulat H. armigera dan S.litura. Dalam pengujian karakter biologi jamur diuji 4 macam mediaperbanyakan, yaitu: (1) Sabouraud maltose agar + ekstrak yeast (SMAY),(2) Sabouraud maltose agar + ekstrak yeast + ekstrak beras (SMAYB), (3)Sabouraud maltose agar + ekstrak yeast + ekstrak kentang (SMAYK), dan(4) Media lengkap untuk N. rileyi (MLNr), serta 2 tingkat suhu inkubasi,yaitu 23±1 dan 27±1ºC. Perlakuan disusun dalam rancangan acak lengkap(RAL) dengan lima kali ulangan. Setiap media disiapkan di dalam 10cawan petri per perlakuan dan masing-masing diinokulasi dengan 10 5konidia/ml. Parameter yang diamati adalah laju pertumbuhan jamur danproduksi konidia. Sedangkan dalam pengujian patogenisitas konidia N.rileyi terhadap larva H. armigera dan S. litura dilakukan dengan metodepelumuran (painting), yaitu ulat diletakkan di atas konidia di dalam cawanpetri selama ± 10 detik kemudian dipindahkan ke vial-vial plastikberdiameter 2,5 cm berisi pakan daun kapas muda (± 1 cm 2 ) untuk H.armigera dan daun jarak kepyar untuk S. litura. Apabila pakan daun telahhabis, serangga diberi pakan buatan berbahan dasar tepung kedelai. Pakanbuatan diganti setiap 2 hari sampai ulat menjadi pupa. Selanjutnya ulatyang telah diperlakukan dengan jamur diinkubasi-kan pada suhu ruang(27°-29°C) selama ± 14 hari dan diamati perkembangan ulat maupunjamurnya setiap hari. Parameter yang diamati adalah mortalitas ulat H.armigera dan S. litura serta gejala mikosis pada ulat terinfeksi. Hasilpenelitian menunjukkan bahwa suhu inkubasi berpengaruh terhadap lajupertumbuhan N. rileyi. Pada suhu 23±1ºC N. rileyi tumbuh lebih cepat(7,42-8,23 mm/hari) pada semua komposisi media yang diuji (SMAY,SMAYK, SMAYB, dan MLNr) dibanding pada suhu 27±1ºC (0,99-1,26mm/hari). Produksi konidia N. rileyi lebih banyak pada suhu 27±1ºCdibanding pada 23 ± 1ºC, yaitu berturut-turut 24,7 x 10 8 konidia/ml dan17,9 x 10 8 konidia/ml masing-masing pada media SMAYK dan MLNr.Perbedaan komposisi media tumbuh tidak menyebabkan penurunanpatogenisitas pada konidia N. rileyi sebab mortalitas ulat H. armigeramaupun S. litura masing-masing mencapai 100%. Hasil penelitianmengindikasikan bahwa N. rileyi mudah diperbanyak secara massal padamedium agar dan virulensinya baik pada H. armigera dan S. litura.</p><p>Kata kunci : Nomuraea rileyi, epizootik, Helicoverpa armigera,Spodoptera litura, konidia, patogenisitas, mortalitas</p><p>ABSTRACT</p><p>Effect of medium composition on growth of entomo-pathogenic fungi Nomuraea rileyi (Farlow) Samson andits pathogenicity against Helicoverpa armigera andSpodoptera litura</p><p>N. rileyi is one of potential entomopathogenic fungi to controlcotton bollworm, Helicoverpa armigera, tobacco and Rhicinus caterpilar,Spodoptera litura. These fungi naturally infect those insect pests indicatingtheir potential to be used as natural control agent. Techniques of in vitroproduction of these fungi need to be developed to find out their potentialagainst the insect target. Study on effect of medium composition ongrowth of entomopathogenic fungi N. rileyi and its pathogenicity againstH. armigera and S. litura was carried out at Phytopathology and InsectPathology Laboratories of Indonesian Tobacco and Fiber Crops ResearchInstitute (IToFCRI) from May to November 2009. The objective of thestudy was to find out the suitable composition of medium for N. rileyi andits pathogenicity against H. armigera and S. litura. The study consisted oftwo tests. The first test was testing for biological characters, namely invitro growth rate and conidia production of N. rileyi on four differentcompositions of medium as followed: (1) Sabouraud maltose agar + yeastextract (SMAY), (2) Sabouraud maltose agar + yeast extract + rice extract(SMAYR), (3) Sabouraud maltose agar + yeast extract + potato extract(SMAYP), and (4) Completed medium for N. rileyi (MLNr). Alltreatments were designed in randomized complete design (RCD) with fivereplicates. Parameters observed were the growth rate of N. rileyi andconidia production. The second was testing on pathogenicity of N. rileyiproduced from all medium tested against H. armigera and S. litura larvae.Result showed that incubation temperature influenced the growth rate offungi. N. rileyi grew faster at 23±1ºC (7.42-8.23 mm/day) than that at27±1ºC (0.99-1.26 mm/day) on all media tested. Conidia production washigher at 27±1ºC than at 23±1ºC. Both SMAYP and MLNr were the bestmedia for producing N. rileyi conidia, which were 24.7 and 17.9 x 10 8conidia/ml, respectively. Pathogenicity of N. rileyi against H. armigeraand S. litura was not affected by composition of medium tested becausethe larval mortality of both insect pests was 100%. Entomopathogenic fungi (EPFs), Isaria fumosorosea and Beauveria bassiana, are efficient biological agents in the management of multiple arthropod pests. In this study, the effects of both EPF species on various life stages of Spodoptera litura (F.) (Lepidoptera: Noctuidae) and its natural enemy Rhynocoris marginatus (Fab.) (Hemiptera: Reduviidae) were determined under laboratory conditions. I. fumosorosea significantly ( P < .05) reduced the growth rate of the third and fourth instar larvae of S. litura. For relative consumption rate (RCR), the maximum impact was recorded for I. fumosorosea, which reduced the RCR of the larvae. The larvae of S. litura treated with I. fumosorosea showed significantly lower efficiency of conversion of ingested food (ECI) and the larval mortality rate (58.0%) was also higher compared with B. bassiana (33.3%). Similarly, I. fumosorosea had a significant effect on the pupal formation of S. litura; however, no significant effect was found on adult emergence percentage. To determine the effect of EPF-infected prey on the adult predator, their handling time, predatory rate, consumption rate, and the survival rate were recorded. No significant effect of EPF species on the predation rate was found. Furthermore, no significant difference was found in the survival rate of predators fed on either EPF-infected prey or healthy larvae. The interaction of these EPFs with a reduviid predator suggested that both EPF species, especially I. fumosorosea, could be used together with the predator to boost the biological control of S. litura in commercial crops.

Abstract Background Spodoptera litura Fabricius (Lepidoptera: Noctuidae) is an important pest causing severe damage to many cultivating plants such as corn worldwide, including Indonesia. This study was performed to obtain and identify entomopathogenic fungi (EPF) of S. litura collected from corn fields in 4 corn producing regions of Lampung, Indonesia, as well as to investigate the damage caused by this pest on corn in Lampung Province. Results Three corn fields in each region were selected for collecting soil samples. Soil samples were collected from 5 corn plant rhizospheres, at each field in six months of survey. Ten larvae of Tenebrio molitor Linnaeus (Coleoptera: Tenebrionidae) were laid on each soil sample as a bait, covered with a filter paper and incubated at room temperature. The emerging fungi from T. molitor cadaver were transferred onto Potato Dextrose Agar (PDA) medium and incubated for 7 days at room temperature. Pathogenicity test was determined against 3rd instar of S. litura larvae. Identification was performed based on the sequence of Internal Transcribed Spacer (ITS) Region. Observations on the corn damage caused by S. litura were conducted at all corn producing areas in Lampung. Twelve fungal isolates were obtained causing 0–75% of mortality of S. litura. Four fungal isolates (NKPT, SKHJ, SDHJ and RAHJ), which caused mortality more than 20%, were further identified. One isolate (NKPT) was confirmed as Beauveria bassiana and the other 3 isolates (SKHJ, SDHJ and RAHJ) were Aspergillus oryzae. S. litura generally caused slight damages to the corn which was found in every observation year performed during 2010–2019. Medium plant damage was observed in 2010–2012 and 2018–2019, severe damage was found in 2011 and crop failure was recorded in 2018. Conclusions Aspergillus oryzae and B. bassiana were the EPF recorded infecting S. litura in corn in Lampung Province. This was the first report on the isolates of A. oryzae as EPF of S. litura in Indonesia. Slight damages with S. litura were always recorded in every observation year but not for those of medium and severe damages and crop failure.

Abstract Background The tobacco cutworm, Spodoptera litura [Fab.] (Lepidoptera: Noctuidae), is a devastating insect pest of several crops. Entomopathogenic nematodes (EPNs) of the families Heterorhabditidae and Steinernematidae are used as an alternative control measure in lieu of the hazardous synthetic chemical applications. Results A survey of naturally occurring EPNs was conducted across the province of Cotabato, Philippines, covering a total of 5 municipalities with 25 villages. Galleria-baiting technique was employed to recover nematodes from peanut and grassland soils. Out of 50 soil samples collected, only 5 samples harbored nematodes, indicating a recovery of 10%. Preliminary morphological data identified only one EPN under the genera Heterorhabditis (1 isolate), whereas 4 were facultative necromenic nematodes from the genera Metarhabditis (2 isolates) and Oscheius (2 isolates). Analysis of D2D3 segments of the 28S rDNA confirmed high sequence similarity to Heterorhabditis indica, Metarhabditis rainai, Oscheius insectivora, and Oscheius sp. This is the first record of H. indica and M. rainai in the entire region, whereas the first record for Oscheius spp. in the Philippines. Furthermore, the biocontrol potential of the local H. indica infective juvenile (IJ) populations (PIGCD1) isolated from peanut was assessed against the tobacco cutworm, S. litura, under laboratory conditions. The mean percentage mortality caused by H. indica on S. litura at 7 different concentrations ranged from 0-100% at 24 h post inoculation. The lethal concentration (LC50) required to kill 50% of the S. litura larvae population with H. indica was 7.13±1 (IJs/larva). Conclusions The use of Galleria-baiting method is a convenient approach to detect EPNs including other facultative necromenic nematodes from the soils. Obtained data indicated that the local H. indica isolate can be a promising alternative measure to suppress the economically important insect pest, S. litura, and this may provide significant outlook to establish the biocontrol program in the country.

Spodoptera litura Fabricius (Lepidoptera: Noctuidae) atau dikenal dengan ulat grayak merupakan hama penting pada tanaman kedelai dan beberapa jenis tanaman penting di Indonesia. Serangan S. litura dapat mengakibatkan kerusakan, bahkan kehilangan hasil pada tanaman kedelai. Gejala serangan berupa daun berlubang karena larva memakan jaringan daun hingga menyisakan epidermis dan tulang daun. Hama ini dilaporkan menyerang  tanaman kedelai di sentra-sentra produksi di Indonesia yaitu Aceh, Jawa Barat, Jawa Tengah, Jawa Timur, Nusa Tenggara Barat, Sulawesi Selatan dan dan Sulawesi Tengah. Upaya pengendalian hama ulat grayak yang dilakukan petani  adalah menggunakan pestisida sintetik, namun karena dampak negatif penggunaannya terhadap kesehatan manusia dan  keseimbangan ekosistem alam, maka perlu adanya alternatif pengendalian yang dampak negatifnya rendah terhadap lingkungan. Beberapa teknologi pengendalian yang telah diteliti dan diketahui efektifitasnya antara lain; penggunaan Spodoptera litura nuclear polyhedrosis virus (SlNPV) (50-100%), cendawan entomopatogen Beauvaria bassiana (51-93%), Metarhizium anisopliae (93-100%), Nomuraea rileyi  dan Lecanicillium lecanii (80-85%), Parasitoid (13-56%), predator Forficula auricularia (96%), nematoda entomopatogen Steinernematidae (30-51%), pestisida nabati (&gt;30%), tanaman perangkap dan varietas tahan Aplikasi yang tepat akan mendukung perkembangbiakan spesies tersebut di alam sehingga akan terjadi siklus rantai makanan yang seimbang dan berkelanjutan. Komponen pengendalian ini dapat dimasukkan dalam pengendalian terpadu, sehingga serangan S. litura dapat ditekan, tanaman berproduksi optimal, keseimbangan ekosistem dapat dipertahankan,  residu pestisida dapat diturunkan dan sistem pertanian berkelanjutan tercapai.
