Avaliou-se, in vitro, a capacidade de crescimento em 39ºC e 42ºC, a produção de enzimas hidrolíticas e a atividade hemolítica de 21 cepas clínicas e de referência de sete espécies de Candida spp, Candida dubliniensis e Candida krusei demonstraram menor potencial de virulência e Candida albicans maior.

Abstract This study analyzed the antifungal activity of phytoconstituents from linalool on Candida spp. strains, in vitro, isolated from patients with clinical diagnoses of oral candidiasis associated with the use of a dental prosthesis. Biological samples were collected from 12 patients using complete dentures or removable partial dentures and who presented mucous with diffuse erythematous or stippled features, indicating a clinical diagnosis of candidiasis. To identify fungal colonies of the genus Candida, samples were plated onto CHROMagar Candida®. The antifungal activity of linalool, a monoterpene unsaturated constituent of basil oil, was performed using the broth microdilution technique. Then, the minimum inhibitory concentration (MIC), the two subsequent stronger concentrations and the positive controls were subcultured on Sabouraud Dextrose Agar plates to determine the minimum fungicidal concentration (MFC). The experiments were performed in triplicate and nystatin was used as a positive control in all tests. Diagnoses of oral candidiasis were verified in eight patients (66.6%) and the most prevalent fungal species was Candida albicans (37.5%), followed by Candida krusei (25.0%); and Candida tropicalis (4.2%). The best antifungal activity of linalool was observed on Candida tropicalis (MIC = 500 mg/mL), followed by Candida albicans (MIC = 1.000 mg/mL), and Candida krusei (MIC = 2.000 mg/mL).Under the study conditions and based on the results obtained, it can be concluded that the Candida strains tested were susceptible to linalool.

       The objective of this study is to test In Vitro the twenty chemical compounds that contains Schiff base or oxazepine, indoline, imidazolo units in concentrations( 50, 100, 150) mg / dl as antifungal activity, against three pathogenic Candida species  that occur in humans. We tested one isolates of    ) Candida albicans ,Candida glabrata and Candida krusei). All these species affect human health . The study was carried out in the Laboratory of Public Health , directly of health for the period from May 2016 to April 2017  , Candida spp isolates used in this study were collected from patients admitted at  some private clinical in  Kirkuk city . All isolates were identified using CHROM agar and stored at -70 0C. Preparation of Schiff base ( 1-7 ) from amino pyridine derivatives with aromatic aldehyde by nucleophilic addition reactions preparation of 1,3 – oxazepine 4,7 – dione ( 8-13 ) were carried out by cyclization of appropriate Schiff bases with malic anhydride and phthalic anhydride , preparation of compounds ( 14 – 17) from reaction of a mixture aminopyridine derivative with potassium hydroxide then chloroacetic acid was  added , preparation of compounds ( 18 – 20 ) from reaction of amino pyridine derivative and 4- phenyl phenacyl bromide , all these compounds were characterized by melting points and FT.IR spectroscopy. Some of them were characterized by H1- NMR and C13-NMR spectroscopy .Some compounds contain Schiff base  group in compounds ( 1-7) showed inhibitory effect Candida albicans and Candida krusei  .This study demonstrates that the three Candida species were resistant to a range of compounds (8-13) containing oxazepine and as antifungal against , while the compound(14-17) contain Indolin show low inhabition zon for Candida albicans and the compound (18-20) contain imidazo group showed inhibited  effect against three Candida species.

Introducción: la candidemia es una enfermedad grave, con elevada morbi-mortalidad y cuyo tratamiento no siempre conduce a la cura. La distribución de especies de Candida y la sensibilidad antifúngica varía según el área geográfica, incluso entre centros de salud de una misma región. Objetivo: establecer la distribución de especies de Candida y su sensibilidad in vitro frente a diferentes antifúngicos. Materiales y métodos: se realizó un estudio descriptivo, prospectivo y transversal entre noviembre de 2013 y mayo de 2014 de casos de candidemias en una institución de salud de Valledupar, Colombia. Las Concentraciones Inhibitorias Mínimas (CIM) se determinaron según el protocolo estándar M27-A3-S4. Resultados: Se estudiaron 40 aislados clínicos de Candida spp. obtenidos de sangre (97,5%) y médula ósea (2,5%). Del total, 15 (37,5%) fueron caracterizados como Candida tropicalis, 13 (32,5%) del Complejo Candida albicans, cinco (12,5%) del Complejo Candida glabrata, tres (7,5%) como Candida guilliermondii, tres (7,5%) del Complejo Candida parapsilosis y uno (2,5%) como Candida krusei. No se observó resistencia a la anfotericina B ni al voriconazol en ninguno de los aislados, pero sí al fluconazol en uno (6,6%) de Candida tropicalis y uno (33,3%) del Complejo Candida parapsilosis y a la caspofungina en el aislado de Candida krusei y en uno (20%) del Complejo Candida glabrata. Conclusiones: la epidemiología local de las levaduras causantes de candidemia mostró mayor prevalencia de especies no-albicans, entre las que se encontró resistencia a los antifúngicos evaluados, lo que es relevante para la elección e instauración de un tratamiento eficaz.

Background: Probiotic bacteria have been emerging as a trustworthy choice for the prevention and treatment of Candida spp. infections. This study aimed to develop and characterize an orodispersible film (ODF) for delivering the potentially probiotic Enterococcus faecium CRL 183 into the oral cavity, evaluating its in vitro antifungal activity against Candida albicans. Methods and Results: The ODF was composed by carboxymethylcellulose, gelatin, and potato starch, and its physical, chemical, and mechanical properties were studied. The probiotic resistance and viability during processing and storage were evaluated as well as its in vitro antifungal activity against C. albicans. The ODFs were thin, resistant, and flexible, with neutral pH and microbiologically safe. The probiotic resisted the ODF obtaining process, demonstrating high viability (>9 log10 CFU·g−1), up to 90 days of storage at room temperature. The Probiotic Film promoted 68.9% of reduction in fungal early biofilm and 91.2% in its mature biofilm compared to the group stimulated with the control film. Those results were confirmed through SEM images. Conclusion: The probiotic ODF developed is a promising strategy to prevent oral candidiasis, since it permits the local probiotic delivery, which in turn was able to reduce C. albicans biofilm formation.

INTRODUÇÃO: A candidíase é uma das infecções fúngicas mais frequentes entre os pacientes infectados pelo vírus da imunodeficiência humana. O presente estudo objetivou a caracterização das leveduras do gênero Candida de distintas amostras clínicas, provenientes de pacientes HIV - positivos, assim como a determinação do perfil de suscetibilidade in vitro a cinco drogas antifúngicas. MÉTODOS: A caracterização dos isolados de Candida sp foi realizada através da metodologia clássica, testes bioquímicos (zimograma e auxanograma) e morfológicos (prova do tubo germinativo e microcultivo em lâmina). Também, foram realizadas a técnica genotípica (PCR) e identificação pelo método comercial API 20C AUX (BioMeriéux). Para a determinação do perfil de suscetibilidade in vitro, foram utilizadas cinco drogas antifúngicas (cetoconazol, fluconazol, itraconazol, voriconazol e anfotericina B), através do método comercialmente disponível - Etest. RESULTADOS: Foram identificados 105 isolados de leveduras do gênero Candida provenientes de 102 pacientes infectados pelo vírus HIV. Destes, foram caracterizadas 82 (78,1%) Candida albicans, 8 (7,6%) Candida parapsilosis, 8 (7,6%) Candida tropicalis, 4 (3,8%) Candida krusei, 2 (1,9%) Candida glabrata e 1 (1%) Candida guilliermondii. CONCLUSÕES: Considerando o perfil geral de sensibilidade, 60% dos isolados foram suscetíveis a todos os antifúngicos testados, porém as espécies C. tropicalis e C. krusei demonstraram uma tendência a valores mais elevados de CIMs para os azóis do que os encontrados paraC. albicans, sugerindo resistência.

We validated thein vivoefficacy of orally administered toremifene againstC. albicans and S. aureusbiofilm formation in a rat subcutaneous catheter model. Combined, our results demonstrate the potential of toremifene as a broad-spectrum oral antibiofilm compound.

Background: Oral infection caused by Candida spp. is a major healthcare problem in dental and oral care. Treatment failure has been reported in cases of oral candidiasis as a result of resistance to common antifungals. Aim and Objective: In this study, the in vitro and in vivo activities of extract of Tithonia diversifolia against virulence factor-borne and antifungal resistant-Candida albicans were investigated. Candida albicans was isolated from the saliva of patients attending a tertiary hospital in Ekiti State. Methodology: Standard methods were used to determine the presence of virulence factors in the isolates. In vitro and in vivo anti-candidal activities of the hydro-ethanolic extract of T. diversifolia were also tested on the test fungus. Results: The virulence factors have varying percentage of occurrence in all the isolates with catalase having the highest. Itraconazole and nystatin were not effective against the isolates. Out of the six isolates selected (based on antifungal resistance) only three produced strong biofilm. The reduction in the population of the test organisms by the extract was time and concentration dependent. At the end of candidal challenge and treatment assays, extract of T. diversifolia has lower anti-candidal property compared to nystatin. Conclusion: This study has shown that C. albicans associated with the mouth carries virulence factors and are resistant to common antifungals. In this work, we noticed antifungal effects of hydro-alcoholic extract of T. diversifolia on C. albicans associated with oral infections.

Candida sp. é um gênero de levedura que faz parte da microbiota do corpo humano e de animais. É considerado o principal grupo de fungos patógenos oportunistas. O uso de plantas medicinais é empregado cada vez mais nas diversas áreas. A planta Cymbopogon citratus (DC). Stapf é popularmente conhecida no Brasil por Capim-cheiroso, Capim-cidreira, Capim-santo, Capim-limão, Capim-cidró, Capim-cidrão. Este estudo teve como objetivo avaliar a atividade antifúngica do óleo essencial de Cymbopogon citratus frente a leveduras do gênero Candida sp.. O óleo essencial foi obtido por hidrodestilação das folhas pelo método do arraste a vapor. A atividade antifúngica foi verificada pelas técnicas de disco-difusão, concentração inibitória mínima e concentração letal mínima. Foram utilizadas as cepas padrão ATCC de Candida albicans, Candida dubliniensis, Candida tropicalis, Candida glabrata, Candida Krusei, Candida parapsilosis e Candida utilis. O óleo essencial apresentou atividade antifúngica frente a todas as espécies. A faixa de inibição varia de 0,281 µg/mL a 1,125 µg/mL e concentração letal varia de 0,562 µg/mL a 1,125 µg/mL. Esses achados contribuem para um futuro emprego da planta C. citratus como fitoterápico em tratamento preventivo ou alternativo contra infecções fúngicas.
