ABSTRACTThermophilic bacteria are regarded as attractive production organisms for cost-efficient conversion of renewable resources to green chemicals, but their genetic accessibility is a major bottleneck in developing them into versatile platform organisms. In this study, we aimed to isolate thermophilic, facultatively anaerobic bacilli that are genetically accessible and have potential as platform organisms. From compost, we isolated 267 strains that produced acids from C5and C6sugars at temperatures of 55°C or 65°C. Subsequently, 44 strains that showed the highest production of acids were screened for genetic accessibility by electroporation. TwoGeobacillus thermodenitrificansisolates and oneBacillus smithiiisolate were found to be transformable with plasmid pNW33n. Of these,B. smithiiET 138 was the best-performing strain in laboratory-scale fermentations and was capable of producing organic acids from glucose as well as from xylose. It is an acidotolerant strain able to produce organic acids until a lower limit of approximately pH 4.5. As genetic accessibility ofB. smithiihad not been described previously, six otherB. smithiistrains from the DSMZ culture collection were tested for electroporation efficiencies, and we found the type strain DSM 4216Tand strain DSM 460 to be transformable. The transformation protocol forB. smithiiisolate ET 138 was optimized to obtain approximately 5 × 103colonies per μg plasmid pNW33n. Genetic accessibility combined with robust acid production capacities on C5and C6sugars at a relatively broad pH range makeB. smithiiET 138 an attractive biocatalyst for the production of lactic acid and potentially other green chemicals.

Inflammatory bowel disease (IBD) is a chronic intestinal disease associated with the inflammatory gastrointestinal tract and microbiome dysbiosis. Probiotics are a promising intervention, and several probiotics have been reported to positively affect IBD remission and prevention, particularly on ulcerative colitis (UC). However, there is still a limitation in the knowledge of effectiveness and safety of probiotics therapies for IBD. Exploring more potential probiotics helps to find extensive evidence for probiotic intervention. This study established a rapid method for probiotics candidate screening and finally screened out one strain with the best protective effect. Forty strains isolated from four different sources were used for this screening. Hemolysis tests and acute toxic test evaluated strain safety. Zebrafish were first treated with dextran sodium sulfate (DSS) for colitis induction, and every bacteria were individually added to the fish water subsequently. Results showed eight strains could lower the larvae mortality within 3 days under a 0.6% DSS concentration, including Lacticaseibacillus rhamnosus GG, L. rhamnosus NBRC3425, Bacillus smithii DSM4216, B. smithii XY1, Bacillus coagulans NBRC12583, Bacillus coagulans XY2, Lactobacillus parafarraginis XYRR2, and Bacillus licheniformis XYT3. Among eight, B. smithii XY1 was the only strain having the equal ability to alleviate neutrophil infiltration in the larvae intestine with that ability of prednisolone under a 0.5% DSS concentration. Bacillus smithii XY1 restored intestinal epithelial cell integrity after DSS damage, as well as regulated the gene expression inflammation-related factors, indicating its bio-function of inflammatory response alleviation.

Avoiding food-borne diseases by competitive exclusion agents is a proactive strategy. In the current paper, we report the use of Bacillus smithii TBMI12 spores as potential competitive exclusion agents. One group of mice was predosed for three successive days with 108 colony forming units of B. smithii TBMI12 spores followed by inoculation with 106 colony forming units of wild-type Salmonella enterica serotype Enteritidis cells. Microbial plate counts of the animals' livers and spleens showed that only 40% of the mice were infected with S. enterica serotype Enteritidis, while the control group was 100% infected. These results suggest that B. smithii TBMI12 spores may protect against infection by S. enterica serotype Enteritidis.
