CHANGES IN SECRETION OF LUTEINIZING HORMONE AND FOLLICLE-STIMULATING HORMONE INDUCED BY HYPOVOLAEMIA IN RATS

Changes in levels of LH and FSH in the circulation were examined during repeated blood sampling in untreated rats and gonadectomized male and female rats treated with oestrogen, progesterone and thyroxine. Blood depletion induced a significant increase in levels of LH in steroid-treated rats but the increase was abolished when the depleted blood volume was replaced with egg albumen. The rise in LH was less dramatic in male than in female animals. In untreated rats, levels of LH either decreased or did not change with repeated phlebotomy. In contrast, the levels of FSH either did not alter or were lowered in all situations.

ABSTRACT A preparation of ovine follicle stimulating hormone (FSH) which is approximately 50 times as active as NIH-FSH-S1 has been tested in hypophysectomized male and female rats and found to be a potent steroidogenic factor. Contamination of the FSH preparation with interstitial cell-stimulating hormone (ICSH, LH) was found to be very low (0.006–0.016 unit/mg) and could not account for the observed activity. In addition, treatment of the FSH preparation with 6 m urea and neuraminidase indicates that the steroidogenic activity is either an intrinsic property of the FSH molecule or a discrete entity which differs chemically from ICSH.

ABSTRACT Augmentation of testis weight caused by the follicle stimulating hormone (FSH) was studied in hypophysectomized immature rats treated concomitantly with supramaximal amounts of human chorionic gonadotrophin (HCG). Using a rat pituitary homogenate, responses were found to increase linearly with the logarithm of the doses given. In assays of rat pituitary homogenate against purified sheep FSH no significant divergence was observed. The effect of a submaximally active dose of FSH was not significantly altered by the concomitant administration of purified growth hormone, thyrotrophin, corticotrophin, prolactin or luteinizing hormone (LH). Serum from intact male and female rats caused significant growth of the testis, while serum from male animals was more active than that of female rats. However, when serum from hypophysectomized male and female rats was given, some testis growth still occurred in one of the two experimental series. In this case, the responses were smaller than those caused by serum from intact males but not smaller than those caused by serum from intact females. No difference in testis growth-promoting effect could be demonstrated between the sera obtained from hypophysectomized male and female rats.
