Effects of hyperprolactinaemia on reproduction in male mice

ABSTRACT Hyperprolactinaemia induced by pituitary isografts in male host mice was confirmed by radioimmunoassay, but plasma testosterone levels determined by radioimmunoassay in these mice showed no changes. Immunoenzyme electron microscopic observations revealed large spherical-shaped immunoreactive prolactin granules in pituitary grafts in male hosts, regardless of the sex of the donor mice, indicating the disappearance of sexual dimorphism in prolactin-producing cells in hyperprolactinaemic mice. In hyperprolactinaemic host mice the male accessory sex glands, particularly the seminal vesicle and the ventral prostate, exhibited considerable proliferation and significant increase in weight. These phenomena do not seem to be mediated by the increased action of testosterone. Such biological effects in host mice were much greater when the donor was female rather than male, and were more noticeable in C57BL mice than in C3H mice. J. Endocr. (1985) 107, 71–76

SUMMARY Plasma testosterone concentration and the weights of the seminal vesicles and ventral prostate gland were measured in normal and cryptorchid Fischer rats at 3, 4·5, 7·5 and 13·5 months of age, and in normal parabionts and cryptorchid parabionts of 13·5 months of age. Testosterone was measured individually by a protein-binding method. In normal rats, all parameters rose to a maximum at 7·5 months of age, then levelled off or declined at 13·5 months of age. In cryptorchid rats, a similar pattern at a lower level was found for accessory sex gland weight, but plasma testosterone levels showed a progressive decline from an above normal level at 3 months to a subnormal level at 13·5 months of age. Cryptorchid parabionts were less responsive to gonadotrophin stimulation from union with a castrated partner than normal parabionts. The present study showed that plasma testosterone levels in normal and cryptorchid rats are age-related. It also showed that the pattern of plasma testosterone levels observed in cryptorchid rats is different from that seen in normal rats. Accessory sex gland weight is also age-related but is not a reliable index of plasma testosterone levels.

C57BL male mice (Jackson Lab., Bar Harbor, Maine) weighing about 18 gms were randomly divided into three groups: group I was fed sweetened liquid alcohol diet (modified Schenkl) in which 36% of the calories were derived from alcohol; group II was maintained on a similar diet but alcohol was isocalorically substituted by sucrose; group III was fed regular mouse chow ad lib for five months. Liver and heart tissues were fixed in 2.5% cacodylate buffered glutaraldehyde, post-fixed in 2% osmium tetroxide and embedded in Epon-araldite.

Angiotensin-1-7 (Ang-(1-7)) counteracts angiotensin II through effects at Mas receptors (MasR). We demonstrated that sexual dimorphism of obesity-hypertension was associated with dysregulated production of Ang-(1-7). However, the role of MasR in sexual dimorphism of obesity-hypertension has not been examined. MasR deficient mice have also been reported to exhibit deficits in cardiac function. In this study, we hypothesized that deficiency of the MasR would differentially regulate obesity-hypertension in male versus ( vs ) female mice. In addition, we quantified effects of MasR deficiency on cardiac function in obese male mice. Male and female MasR +/+ and -/- mice were fed a low fat (LF, 10%kcal) or high fat (HF, 60% kcal) diet for 16 weeks, and blood pressure was quantified by radiotelemetry. As demonstrated previously, male MasR +/+ mice (24 hr diastolic blood pressure, DBP: LF, 90 ± 3; HF, 96 ± 2 mmHg; P<0.05), but not females (LF, 85 ± 1; HF, 85 ± 2 mmHg), developed hypertension in response to HF feeding. MasR deficiency converted female HF-fed mice to an obesity-hypertension phenotype (DBP: 92 ± 1 mmHg; P<0.05). Surprisingly, male HF-fed MasR -/- mice exhibited reduced DBP compared to HF-fed MasR +/+ males (90 ± 1 vs 96 ± 2 mmHg; P<0.05). To define mechanisms for reductions in DBP of HF-fed male MasR -/- mice, we performed cardiac magnetic resonance (CMR) imaging in both genotypes at 1 month of HF feeding. MasR -/- mice had significantly reduced ejection fraction (EF) compared to MasR +/+ mice at baseline (51.4 ± 2.5 vs 59.3 ± 2.1%; P<0.05) and after one month of HF-feeding (49.8 ± 2.4 vs 52.6 ± 1.9%; P<0.05). Further, CMR imaging demonstrated a thickening of the ventricle wall in MasR -/- mice with 1 month of HF-feeding. MasR +/+ , but not MasR -/- mice, exhibited diet-induced reductions in EF (by 16%; P<0.05) at 1 month of HF feeding, which were reversed by infusion of Ang-(1-7). These results demonstrate that MasR contributes to sexual dimorphism of obesity-hypertension. Ang-(1-7) protects females from obesity-hypertension through the MasR. In contrast, reductions in DBP in obese male mice with MasR deficiency may arise from deficits in cardiac function. These results suggest that MasR agonists may be effective therapies for obesity-associated cardiovascular conditions.

The relative importance of testosterone and prolactin in regulating growth and protein synthesis in rat accessory sex glands has been investigated. Protein synthesis was measured by incubating tissue minces in vitro with [35S]methionine and analysing labelled proteins on polyacrylamide gels containing sodium dodecyl sulphate. Plasma prolactin was assayed by radioimmunoassay. Results showed that castration for 8 days significantly reduced wet weights and total protein synthesis in the ventral prostate, dorsolateral prostate and caput epididymidis, but that these effects could be reversed by exogenous testosterone. Similarly, the specific incorporation of [35S]methionine into four polypeptides in the ventral prostate, two polypeptides in the dorsolateral prostate and two polypeptides in the caput epididymidis was lowered by castration but markedly stimulated by testosterone. Acute or chronic administration of 2-bromo-α-ergocryptine to animals in combination with testosterone had no significant effect on any of the parameters measured, although the drug reduced circulating prolactin to undetectable levels. In addition, exogenous prolactin given alone, or in combination with testosterone, to hypophysectomized rats had no effect on general or specific protein synthesis. The induction of hyperprolactinaemia in immature or mature rats with pituitary homographs had no effect on testosterone-stimulated growth of any accessory gland, although it caused a significant stimulation of total protein synthesis in the dorsolateral prostate and coagulating glands. However, this was a generalized effect as it did not increase the specific incorporation of [35S]methionine into androgen-dependent proteins. The results do not indicate a major role for prolactin in regulating androgen responsiveness of male accessory sex glands in the rat.
