Traditional Chinese herbal medicines (TCHMs) were tested for their ability of antiquorum sensing. Water extracts ofRhubarb, Fructus gardeniae, and Andrographis paniculatashow antiquorumsensing activity when usingChromobacterium violaceumCV12472 as reporter; the sub-MIC concentrations of these TCHMs were tested against AHL-dependent phenotypic expressions of PAO1. Results showed significant reduction in pyocyanin pigment, protease, elastase production, and biofilm formation in PAO1 without inhibiting the bacterial growth, revealing that the QSI by the extracts is not related to static or killing effects on the bacteria. The results indicate a potential modulation of bacterial cell-cell communication,P. aeruginosabiofilm, and virulence factors by traditional Chinese herbal medicine. This study introduces not only a new mode of action for traditional Chinese herbal medicines, but also a potential new therapeutic direction for the treatment of bacterial infections, which have QSI activity and might be important in reducing virulence and pathogenicity of pathogenic bacteria.

Postpartum treatments have been used by local women in Indonesia for some time now. One commonly used postpartum treatment is the consumption of a traditional herbal medicine called PHM-1 during the first 40 days after childbirth. In addition, a second medicine known as PHM-2 is taken for days 41-80. However, sufficient scientific evidence about the benefits of this postpartum herbal medicine does not exist. In this study, the antibacterial abilities of PHM-1 and PHM-2 therefore was evaluated and showed positive potential for both. Low IC50 values were obtained against pathogenic bacteria such as Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Rhodopseudomonas palustris, Ralstonia pickettii, and Staphylococcus epidermidis and compared with chloramphenicol as a positive control. The antibacterial activity of both PHM-1 and PHM-2 against R. pickettii had the highest inhibitory activity as evidenced by the lowest IC50 values of 11.16 and 6.26 μg/ml, respectively. Furthermore, an in vivo acute toxicity test showed that PHM-1 produced a low LD50 value of 3.28 g/kg BW for both of male and female mice. These results support the use of both PHM-1 and PHM-2 as antibacterial treatments for postpartum women.

Use of herbal medicine faces constraint particularly imparting knowledge in identifying whether a product is microbiologically fit for health or not. There has been relatively less research on microbial quality of the herbal medicines in Nepal. In this context, this research has focused on microbial quality of different herbal medicines. A total of twenty one herbal medicines were collected from different sales outlet of Kathmandu. The microbial load in herbal medicine was determined by aerobic plate count method and bacterial isolates were identified based on morphological, cultural and biochemical tests. Out of twenty one herbal medicines analyzed, all were found free from pathogenic bacteria and indicator organism of fecal contamination. However , Bacillus spp. were isolated from ten herbal medicines. The microbial load on Nutrient Agar was found within the range of 1.20x103- 6.06x105 cfu/ ml (or g). Altogether six different Bacillus spp were identified and the most predominant was Bacillus subtilis. In vitro antibacterial activity of the herbal medicines, from which microorganisms were not detected, were determined against six test bacteria by cup plate method. Out of eleven different herbal medicines, five showed the zone of inhibition against all test bacteria and at least two test bacteria were inhibited by each of the herbal medicines. The highest zone of inhibition was 30 mm shown by Chitrakaharitaki Churna of concentration 100mg/ml against Pseudomonas aeruginosa. Febrile malaria and asymptomatic malaria parasitemia substantially decrease iron absorption in single-meal, stable isotope studies in women and children, but to date there is no evidence of decreased efficacy of iron-fortified foods in malaria-endemic regions. Without inadequate malarial surveillance or health care, giving iron supplements to children in areas of high transmission could increase morbidity and mortality. The most likely explanation is the appearance of non-transferrin-bound iron (NTBI) in the plasma. NTBI forms when the rate of iron influx into the plasma exceeds the rate of iron binding to transferrin. Two studies in women have reported substantially increased NTBI with the ingestion of iron supplements. Our studies confirm this, but found no significant increase in NTBI on consumption of iron-fortified food. It seems likely that the malarial parasite in hepatocytes can utilize NTBI, but it cannot do so in infected erythrocytes. NTBI however may increase the sequestration of parasite-infected erythrocytes in capillaries. Bacteremia is common in children with severe malaria and sequestration in villi capillaries could lead to a breaching of the intestinal barrier, allowing the passage of pathogenic bacteria into the systemic circulation. This is especially important as frequent high iron doses increase the number of pathogens in the intestine at the expense of the barrier bacteria.
