Rhizoctonia solani (teleomorph: Thanatephorus spp.) is a plant pathogenic fungus which had a wide host range. It is best known to cause many diseases in plants such as collar rot, root rot, damping- off, sheath blight, stem canker, web blight and wire stem throughout the world. In vitro study of the various isolates of R. solani for morphological and cultural characters and results revealed that this experiment the various isolates of R. solani differed in colony characters and showed black brown colored cultures with 90.0 mm colony diameter on 7th day of incubation under uniform environmental condition. The highest growth reported was isolates CHIRs-5 and sclerotial formation was recorded in all the isolates of R. solani. Morphology of sclerotia varied from 1.9×1.5 mm of DCHIRs-1 and 1.8×1.5 mm of RUDPRs-2.

Nine isolates of Trichoderma spp. were isolated from different agro- ecological regions of Nepal viz; Jumla, Palpa, Chitwan, Tarahara, Banke, Illam and Salyan and screened against Sclerotium rolfsii Sacc. Adreded soil borne phytopathogen causing collar rot of chickpea in chickpea; In-vitro efficacy of nine fungal antagonist (Trichoderma spp.) against Sclerotium rolfsii were screened. Pot experiment was done to find out the effective management of S. rolfsi through Tricoderma using different methods i.e. Seed treatment, soil drenching and soil application. All the tested isolates of Trichoderma spp. were found effective on mycelial growth inhibition and sclerotial parasitization of S. rolfsii. Trichoderma isolated from Palpa district showed maximum growth inhibition (%) of pathogen periodically after 48(93.78%), 72(96.00%), 96(97.96%) and 120(100.00%) hours of inoculation. Parasitized sclerotium showed minimum sclerotial germination on agar plates. Moreover, Trichoderma species isolated from Palpa districts showed second best percent mycelial growth inhibition periodically at 72(25.00%), 120(29.16%), 168(29.16%) and 216(29.16%).In pot experiment at 40 days after sowing, Seedling height was maximum in soil drenching with 30g per 100ml of water (22.27cm) and Mortality percentage of seedlings was least or highest disease control was observed in seed treated with 109cfu/ml (0.000%).

AbstractWith the increasing demand for antimicrobial agents and the spread of antibiotic resistance in pathogens, the exploitation of plant oils to partly replace antibiotic emerges as an important source of fine chemicals, functional food utility and pharmaceutical industries. This work introduces a novel catalytic method of plant oils hydroxylation by Fe(III) citrate monohydrate (Fe3+-cit.)/Na2S2O8 catalyst. Methyl (9Z,12Z)-octadecadienoate (ML) was selected as an example of vegetable oils hydroxylation to its hydroxy-conjugated derivatives (CHML) in the presence of a new complex of Fe(II)-species. Methyl 9,12-di-hydroxyoctadecanoate 1, methyl-9-hydroxyoctadecanoate 2 and methyl (10E,12E)-octadecanoate 3 mixtures is produced under optimized condition with oxygen balloon. The specific hydroxylation activity was lower in the case of using Na2S2O8 alone as a catalyst. A chemical reaction has shown the main process converted of plantoils hydroxylation and (+ 16 Da) of OH- attached at the methyl linoleate (ML-OH). HPLC and MALDI-ToF-mass spectrometry were employed for determining the obtained products. It was found that adding oxidizing agents (Na2S2O8) to Fe3+ in the MeCN mixture with H2O would generate the new complex of Fe(II)-species, which improves the C-H activation. Hence, the present study demonstrated a new functional method for better usage of vegetable oils.Producing conjugated hydroxy-fatty acids/esters with better antipathogenic properties. CHML used in food industry, It has a potential pathway to food safety and packaging process with good advantages, fundamental to microbial resistance. Lastly, our findings showed that biological monitoring of CHML-minimum inhibitory concentration (MIC) inhibited growth of various gram-positive and gram-negative bacteria in vitro study. The produced CHML profiles were comparable to the corresponding to previousstudies and showed improved the inhibition efficiency over the respective kanamycin derivatives.

Este estudo verificou a eficiência da aplicação de diferentes moléculas em reduzir o comprimento relativo da lesão (CRL) da queima das bainhas em arroz. Plantas dos cultivares BR-Irga 409 e Labelle foram cultivadas em solução nutritiva e inoculadas com Rhizoctonia solani, no estádio de máximo perfilhamento. Às 24 horas antes da inoculação, as bainhas das plantas foram pulverizadas com soluções de silicato de potássio (SP), silicato de potássio + fósforo (SP+F), Acibenzolar-S-Metil (ASM), fungicida Carbendazim, quitosana desacetilada (QD), etileno (ET) e fosfito de potássio (FP). Plantas cujas bainhas foram pulverizadas com água destilada serviram como testemunhas. O efeito das moléculas contidas nesses produtos no crescimento micelial de R. solani foi testado in vitro. Para BR-Irga 409, o CRL foi menor com a aplicação do FP, em relação aos demais tratamentos, exceto o Carbendazim. A aplicação do Carbendazim reduziu em 86,1% o CRL, em relação à testemunha. O CRL foi significativamente menor no cultivar BR-Irga 409 do que no 'Labelle', com aplicação do FP. O crescimento micelial de R. solani foi reduzido apenas pelo FP e Carbendazim, em comparação com os demais tratamentos. Não houve diferença significativa entre os tratamentos testemunha, SP e SP+F para a concentração de Si nas bainhas das plantas dos dois cultivares.

Rhizoctonia damping-off and crown and root rot caused by Rhizoctonia solani are major diseases of sugar beet (Beta vulgaris L.) worldwide, and growers in the United States rely on fungicides for disease management. Sensitivity of R. solani to fungicides was evaluated in vitro using a mycelial radial growth assay and by evaluating disease severity on R. solani AG 2-2 inoculated plants treated with fungicides in the greenhouse. The mean concentration that caused 50% mycelial growth inhibition (EC50) values for baseline isolates (collected before the fungicides were registered for sugar beet) were 49.7, 97.1, 0.3, 0.2, and 0.9 μg ml−1 and for nonbaseline isolates (collected after registration and use of fungicides) were 296.1, 341.7, 0.9, 0.2, and 0.6 μg ml−1 for azoxystrobin, trifloxystrobin, pyraclostrobin, penthiopyrad, and prothioconazole, respectively. The mean EC50 values of azoxystrobin, trifloxystrobin, and pyraclostrobin significantly increased in the nonbaseline isolates compared with baseline isolates, with a resistant factor of 6.0, 3.5, and 3.0, respectively. Frequency of isolates with EC50 values >10 μg ml−1 for azoxystrobin and trifloxystrobin increased from 25% in baseline isolates to 80% in nonbaseline isolates. Although sensitivity of nonbaseline isolates of R. solani to quinone outside inhibitors decreased, these fungicides at labeled rates were still effective at controlling the pathogen under greenhouse conditions.
