Modifikasi pH air penyuling pada proses ekstraksi nilam secara destilasi uap telah dilakukan. Daun nilam yang digunakan berasal dari Banjamegara, Jawa Tengah. Variasi pH air penyuling yang digunakan yaitu 7, 9, 10 dan 12 diperoleh melalui penambahan NaOH 1 N. Komponen minyak nilam dianalisis menggunakan GC-MS. Hasil analisis menunjukkan bahwa rendemen minyak atsiri nilam dengan menggunakan air penyuling pH 7,9, 10 dan 12 masing-masing yaitu 1,232 %, 1,717 %, l,071% dan 0,420 % dari penyulingan 2 kg nilam kering dengan metode destilasi uap. Hasil uji fisika menunjukkan karakteristik minyak atsiri nilam dengan pH air penyuling 7, 9, 10 dan 12 yaitu berwarna kuning kecoklatan (jernih), berwarna kuning kecoklatan (keruh), berwarna kuning kecoklatan (keruh) dan merah kecoklatan. Nilai indeks bias masing-masing yaitu 1,505, 1,504, 1,504 dan 1,506. Hasil uji berat jenis dari masing-masing minyak nilam yaitu 0,962, 0,963,0.967 dan0,978.Komponen utama minyak atsiri nilam yang diperoleh dari penyulingan dengan air pH 7 yaitu alpha-guaiene (14,12 %), seychellene (8,16 %), alpha-patchoulene (5,94 %) Delta-guaiene (15,42 %), dan patchouli alcohol (34,5 %). Sedangkan minyak atsiri nilam dengan air penyulingan pada pH 9 yaitu alpha-guaiene (14,56 %), seychellene (7,97 %), alpha-patchoulene (5,89 %) Delta-guaiene (15,78 %), dan patchouli alcohol (35,62 %). Pada pH 10 dan 12 tidak terdeteksi adanya patchouli alcohol. Penelitian ini menunjukkan minyak nilam dengan kandungan patchouli alcohol tertinggi diperoleh dari hasil penyulingan dengan air penyuling pH 9.

Valeriana jatamansiJones is an important medicinal plant that grows wild in Himachal Pradesh, India. Molecular and biochemical diversity among 13 natural populations from Himachal Pradesh was assessed using RAPD and GC-MS to know the extent of existing variation. A total of seven genetically diverse groups have been identified based on RAPD analysis which corroborated well with the analysis based on chemical constituents. The essential oil yield ranged from 0.6% to 1.66% (v/w). A negative correlation between patchouli alcohol and viridiflorol, the two major valued constituents, limits the scope of their simultaneous improvement. However, other few populations like Chamba-II and Kandi-I were found promising for viridiflorol and patchouli alcohol, respectively. The analysis of chemical constitution of oil of the populations from a specific region revealed predominance of specific constituents indicating possibility of their collection/selection for specific end uses like phytomedicines. The prevalence of genetically diverse groups along with sufficient chemical diversity in a defined region clearly indicates the role of ecology in the maintenance of evolution of this species. Sufficient molecular and biochemical diversity detected among natural populations of this species will form basis for the future improvement.

Pogostemon cablin Benth. is a plant producing patchouli oil, which mostly consists of patchouli alcohol compound. Patchouli oil has great potentials in the world market because of its stability and high price. In this study, in vitro multiplication of Sidikalang variety of Acehnese patchouli shoots was done on solid and liquid Murashige & Skoog (MS) medium. This study aimed to determine the effect of cytokinins in various combinations of shoot multiplication and to compare the patchouli oil yield of in vitro and ex vitro culture. In vitro multiplication of Acehnese patchouli shoots by using solid MS medium with addition of 0.2 ppm benzyl aminopurine (BAP) and 0.2 ppm Kinetin resulted in shoot explants with an average growth index of 82.198 ± 0.690. Patchouli oil extraction was done on 7 weeks old in vitro shoot explants cultured on solid MS medium + 0.2 ppm BAP + 0.2 ppm Kinetin using water distillation method. In vitro shoots yielded 2.5% patchouli oil and contained ± 35% patchouli alcohol compound, whereas ex vitro shoots produced 4% patchouli oil and contained ± 25% patchouli alcohol compound. The qualitative analysis by using thin layer chromatography (TLC) showed that there were similarities in the number of spot and Rf value for each spot of ex vitro and in vitro patchouli oil.
