Tanaman nilam (Pogostemon cablin Benth.) merupakan salah satu tanaman penghasil minyak atsiri atau yang lebih dikenal dengan nama Patchouli Alcohol (PA). PA merupakan senyawa yang terbesar di dalam minyak nilam sebagai penentu mutu minyak nilam, kadar PA dalam minyak nilam menurut SNI 06-2385-2006 yaitu minimal 30%. Sampel minyak nilam yang digunakan adalah minyak nilam yang berasal dari Aceh Tamiang. Tujuan penelitian ini dilakukan untuk mengetahui kadar PA sebelum dan sesudah adsorbsi menggunakan Nanomontmorillonite (Nano-MMT) sebagai adsorben dengan menggunakan metode kualitatif dengan uji warna dan metode kuantitatif dengan analisis Gas Cromatography Mass Spectrometry (GC-MS) untuk menentukan kadar PA, mengingat Nano-MMT adalah jenis tanah liat (clay) yang dapat digunakan sebagai adsorben yang mengadsorpsi kontaminan metalik dengan baik dikarenakan Nano-MMT mempunyai muatan negatif yang dapat mengikat spesies yang bermuatan positif kontaminan metalik seperti logam berat. Berdasarkan hasil analisis diketahui bahwa kadar PA sebelum diadsorpsi 7,74% b/v, dan kadar PA setelah diadsorpsi berkisar antara 4,47%-7,86% b/v, kadar tersebut belum sesuai dengan SNI. Hal ini disebabkan karena banyak hal diantaranya terdapat komponen pengotor berupa kontaminan metalik seperti logam, terjadinya oksidasi, tercampurnya minyak nilam dengan bahan lain, pencemaran wadah penyulingan minyak, dan waktu penyimpanan yang relatif lama.

Abstract BackgroundPatchouli alcohol is an effective component of the medicinal plant patchouli. Similar to other secondary metabolites, its synthesis is likely also regulated by transcription factors. Although the biosynthetic pathway of patchouli alcohol has been characterized, the regulatory mechanism of patchouli alcohol biosynthesis has not been fully revealed.ResultsThis study combined the transcriptome data of patchouli leaves treated with different hormones and WGCNA to establish a coexpression network. The modules correlated to patchouli alcohol content were identified, and PcMYB25 played a crucial role in regulating patchouli alcohol biosynthesis. The overexpression of PcMYB25 can promote the expression of patchouli alcohol synthase (PTS), thereby increasing the content of patchouli alcohol.Conclusions This is the first report that MYB25 regulates the secondary metabolism of patchouli. These experimental results lay the foundation for further analysis of the regulatory mechanism of patchouli alcohol synthesis.

The study aims to determine the price fluctuations and their effects on patchouli farm and patchouli oil production in Aceh Province. The Research Method used survey method with an explanatory research as an approach. The location of the study was conducted in Aceh Province, in Aceh Jaya Regency and Aceh Barat Regency. The fluctuations of patchouli oil price have an affect to the motivation of farmers in developing patchouli farming. Farmers in the study location tend to do patchouli farming when the price of patchouli oil is high, instead farmers do not do patchouli farming when the price of patchouli oil decreases. The fluctuations in patchouli oil prices determined by the quality of patchouli oil and market demand have a dominant influence on the development of production and the productivity of patchouli in Aceh. Standardization of patchouli oil quality at the agro-industry level will increase the price of patchouli oil at the farmers level and increase farmers' income. The price of patchouli oil fluctuates depends on Patchouli Alcohol (PA) levels. This is the main reason for traders at the district and provincial levels to reduce prices at the farm level. Aceh Patchouli has oil quality that qualified with basic export standards. But there is still a need for cultivation techniques and better application of refining technology to improve the quality of patchouli oil at the farmer level so that it can increase the price of patchouli oil at the farmer level while  simultaneously increasing the income and welfare of farmers. The phenomenon of patchouli oil price fluctuations in high has an important role in the development of patchouli oil agro-industry.

Background. Nonsmall-cell lung cancer (NSCLC) is the main type of lung cancer, whose morbidity and mortality rank first among malignant tumors. More than 70% of NSCLC patients are diagnosed at locally advanced or advanced stage, missing the best operation period. Chemotherapy and targeted therapy are important means for the treatment of advanced NSCLC, but various side effects seriously affect the curative effect and the life quality of NSCLC patients. Our previous clinical practice has shown that Mufangji Decoction, a classic traditional Chinese medicine, has a significant curative effect in the treatment of NSCLC, but the specific mechanism is not clear. This study intends to explore the potential mechanism of Mufangji Decoction and its active ingredient patchouli alcohol against NSCLC and to provide a scientific basis for the prevention and treatment of NSCLC by traditional Chinese medicine. Methods. The in vivo and in vitro experiments were performed to evaluate the antitumor effects and investigate the underlying mechanism of Mufangji Decoction and its active ingredient patchouli alcohol. Network pharmacology was applied to analyze the effective ingredients and potential targets or signaling pathways of Mufangji Decoction. Results. Our current study shows that Mufangji Decoction can effectively inhibit the growth of subcutaneous transplantation of NSCLC. The following network pharmacological analysis and in vivo experiment suggest that patchouli alcohol is one of the main active ingredients of Mufangji Decoction and exerts antitumor effects. Further mechanism investigation reveals that the antitumor effect of patchouli alcohol is related to the induction of Akt/mTOR signaling pathway-mediated autophagy in NSCLC cells. Conclusion. Mufangji Decoction and its active ingredient patchouli alcohol might exert their antitumor effects in NSCLC partly through regulating Akt/mTOR-mediated autophagy, providing the evidence that traditional Chinese medicine might be a key approach for NSCLC treatment via targeting the Akt/mTOR signal axis.

Abstract Patchouli oil comes from Tinombala village which has never been tested for the Indonesian National Standard (SNI). The purpose of this study was to determine the quality of patchouli oil by characterizing parameters of physical and chemical properties including color, refractive index, specific gravity, solubility in alcohol, acid number, ester number, iron content (Fe), and patchouli alcohol and alpha copaene content.. Sampling was done by random sampling. The results show a specific gravity of 25/25oC sample A = 0.9564, B = 0.9658, and C = 0.9590, refractive index (nD20) sample A = 1.5111, B = 1.5085 and C = 1.5086, the solubility in alcohol using ethanol of the three samples is >1:10 while using methanol 1:10, the acid number of the sample A=21.0375, B=9.1162 and C=19.6350, the ester number of the sample A=28,05, B=21.04 and C=21.04, iron (Fe) content of sample A=1.84210 mg/L, B=0.186842 mg/L and C=6.78950 mg/L, patchouli content alcohol sample A=,16,96%, B=15,52% and C=14,55% and for alpha copaene not detected. The results showed that the quality of patchouli oil in Tinombala village did not meet the Indonesian National Standard (SNI). Only the specific gravity, refractive index, and iron content are by the quality requirements, while the acid number and ester number and the main component determining the quality of patchouli oil, namely the patchouli alcohol content are still below the Indonesian National Standard, which is below 30%.

Abstract The purpose of this study was to determine the interaction effect of the use of mycorrhizal biofertilizers and varieties to increase the production of organic patchouli on the Entisol soil of Aceh Besar. This study used a factorial randomized block design with three replications. The first factor observed was mycorrhizal biofertilizer from the various genus, namely Glomus mosseae, Gigaspora sp. and the mixed genus of Glomus mosseae and Gigaspora sp. While the second factor is the use of varieties, namely Tapak Tuan and the Lhokseumawe variety. The results showed that the best interactions on the growth and production, P205 uptake, and oil content were found in the mixed mycorrhizal genus (Glomus mosseae + Gigaspora sp.) with the Tapak Tuan variety. From the patchouli alcohol content test results, the best results were obtained in a combination of mixed mycorrhizal biofertilizers and the Tapak Tuan variety by 30%.

Abstract Patchouli alcohol (PA) content is an important indication in the world patchouli oil trade. Testing the PA content of patchouli oil usually takes a long time and expensive, so that it greatly inhibits the activity of buying and selling patchouli oil in the community. Therefore the main objective of this study is to develop NIRS technology to estimate the PA content in Aceh patchouli oil purified rapidly and appropriately using PLSR and PCR methods. The sample in this study was patchouli oil which was purified into 20 types based on PA content. Laboratory test using GC-MS. Near infrared (NIR) spectrum of Patchouli oil samples were acquired and recorded using self-developed FT-IRIPTEK T-1516. The results showed that NIRS technology with PLSR and PCR methods provides a solution to solve the problem of PA content prediction in Aceh’s patchouli oil. SNV pretreatment is proven to improve prediction accuracy and robustness of the prediction model that built. The PLSR-SNV method succeeded in obtaining a prediction model that was categorized as very good model performance, while PCR-SNV method succeeded in obtaining a prediction model that was categorized as good model performance.

Patchouli oil contains patchouli alcohol as the main component which is a tricyclic tertiary alcohol sesquiterpene compound that is insoluble in water. Microemulsion is a droplet dispersion technology that combines an insoluble liquid with another liquid. The role of surfactants in helping the dispersion process of microemulsion formulation is by reducing the interfacial tension. Microemulsion is a supersolvents carrier, so that both hydrophilic and lipophilic can be administered through a microemulsion system. The purpose of this study was to formulate patchouli oil into a stable microemulsion preparation. This research was conducted by making 4 preparations of patchouli oil microemulsion containing 5% (F1), 10% (F2), and 15% (F3) patchouli oil, and microemulsion without patchouli oil (F0). Evaluation of microemulsion preparations includes organoleptic test, pH measurement, density determination, viscosity test, freeze and thaw test, centrifugation test, and particle size analysis. The test results showed that the microemulsion containing 5% patchouli oil had the most stable characteristics compared to other formulas.

Considering the current dramatic and fatal situation due to the high spreading of SARS-CoV-2 infection, there is an urgent unmet medical need to identify novel and effective approaches for prevention and treatment of Coronavirus disease (COVID 19) by re-evaluating and repurposing of known drugs. For this, tomatidine and patchouli alcohol have been selected as potential drugs for combating the virus. The hit compounds were subsequently docked into the active site and molecular docking analyses revealed that both drugs can bind the active site of SARS-CoV-2 3CLpro, PLpro, NSP15, COX-2 and PLA2 targets with a number of important binding interactions. To further validate the interactions of promising compound tomatidine, Molecular dynamics study of 100 ns was carried out towards 3CLpro, NSP15 and COX-2. This indicated that the protein-ligand complex was stable throughout the simulation period, and minimal backbone fluctuations have ensued in the system. Post dynamic MM-GBSA analysis of molecular dynamics data showed promising mean binding free energy 47.4633 ± 9.28, 51.8064 ± 8.91 and 54.8918 ± 7.55 kcal/mol, respectively. Likewise, in silico ADMET studies of the selected ligands showed excellent pharmacokinetic properties with good absorption, bioavailability and devoid of toxicity. Therefore, patchouli alcohol and especially, tomatidine may provide prospect treatment options against SARS-CoV-2 infection by potentially inhibiting virus duplication though more research is guaranteed and secured.
