A single foliar spray with 2.5 mM glycinebetaine (GB) was applied on potato (Solatium tuberosum L.) cvs. Matilda, Sabina and Saturna grown in a sand bed infested with Streplomyces scabies (Thaxter) Waksman & Henrici at tuber initiation. A period of drought was maintained after GB treatment to enhance scab infection. Logit models were used to compare scab incidence in all harvested tubers (n = 622) and the type of scab in the symptomatic tubers (n = 382), whereas the tuber surface areas covered with scab lesions were compared using ANOVA. A 10 g increase in tuber weight increased the odds of scab incidence by 11.3%. Scab incidence was lowest in Sabina and highest in Matilda. Raised scab and superficial scab were common in all cultivars, but pitted scab developed only in a few tubers of Sabina and Matilda. Among the scabby tubers, the incidence of raised scab was highest in Sabina and lowest in Saturna. The mean surface area covered with scab was 32.8% in Matilda, 11.7% in Saturna and 7.5% in Sabina. Treatment with GB slightly reduced the severity of scab symptoms, as shown by the reduction in the proportion of tubers with raised scab among the scabby tubers (odds 0.60). No effect on the incidence of scab or the tuber area covered with scab lesions was detected following GB treatment.

Abstract Introduction Since toddlers explore with their hands, contact burns continue to be a major pediatric problem. The purpose of this report is to review a pediatric burn unit’s 8-year experience with contact burns of the hand. Methods After IRB approval, a review of pediatric contact hand burns that occurred between 2006–2014 was performed. We examined the causes and outcomes in pediatric contact hand burns in a single pediatric burn program. Results In the 8-year span, 535 children suffered contact burns to the hand (67 per year). The majority suffered hands burns from an oven or stove (120). The other etiologies included burns from a fireplace (76), clothing iron (65), curling or straightening iron (50), and firepit or campfire (46). The mean age at time of injury was 2.62 years old, with a range of 2 months old to18 years old. Male children (339) typically burned their hands more than females (197). Locations of injury included the palmar surface, dorsal surface, fingers tips/thumb, wrist or a combination of several different areas. Most children burned the palmar aspect of their hand (384) compared to the dorsal aspect (61). These burns typically cover small total body surface areas (mean 1.08% TBSA), with only 2% of burns comprising >5% TBSA. Approximately, 84% of these patients did not need surgery, but 86 (16%) had skin grafting (usually full-thickness) and 26% needed a secondary surgery. Of those that needed more than two, the average number of procedures was 3.6. Approximately 4.1% of patients needed a tertiary surgery. Causes for tertiary surgeries included contractures and graft loss. Out of twenty-two patients that needed a third surgery, 59% were due to graft loss and 41% were due to contractures. Conclusions Contact burns to the hand continue to be a major problem for toddlers. Children are most likely to burn themselves on an oven or stove, fireplace, clothing iron or curling/straightening iron. The palmar surface of the hand is the most likely site. While most children do not require surgery, approximately 16% require grafting. A significant number of those patients need reconstructive surgery. Clearly, current prevention efforts have failed to reduce these injuries. Applicability of Research to Practice Palm burns are common in young children. Efforts should focus on preventing these injuries.

SummarySix field experiments, three each during 1982–3 and 1983–4, were conducted on a sandy loam soil to study the effect of varying levels of nitrogen, phosphorus and potassium, in the absence and presence of farmyard manure (FYM) (30 t/ha), on the number of tubers and yield of potato in three grades. Increase in nitrogen, phosphorus and potassium application, in the absence or presence of FYM, did not significantly affect the total number of tubers/m2 but did affect the number of tubers in different grades. An increase in nitrogen and potassium significantly decreased the number of tubers/m2 in small (< 25 g) and increased in medium (25–75 g) and large (> 75 g) grades at 45, 60, 75 and 90 days after planting. Increase in the application of phosphorus increased the number of tubers/m2 in the small grade and decreased it in the large grade but did not affect the number in the medium grade. Increase in nitrogen and potassium application decreased the tuber yield in the small grade and increased it in the medium and large grades. Applied phosphorus increased the yield in the small and medium grades and decreased it in the large grade. The increase in the yield of tubers with increase in nitrogen and potassium application was found to be caused by an increase in the number of tubers in the medium and large grades at the expense of the small grade; however, with applied phosphorus the increase in yield was due to increase in the weight of individual tubers within the small and medium grades. FYM application decreased the number of tubers in the small grade and increased it in the medium and large grades. The response of potato to nitrogen increased and to phosphorus and potassium decreased with the application of FYM.

Production of the phytotoxin thaxtomin A by pathogenic Streptomyces spp. is essential for induction of common scab disease in potato. The disease can be significantly reduced by a range of chemicals applied as foliar sprays before tuber initiation. We tested a range of chemicals that had previously demonstrated varying capacities to reduce common scab for both disease suppression and their ability to inhibit thaxtomin A toxicity in both ‘Desiree’ and ‘Russet Burbank’ potato. Our results for disease suppression generally supported previous studies. Our tuber slice assays with thaxtomin A showed a strong correlation between the ability of the chemical to suppress common scab symptom development and the ability of the chemical to inhibit thaxtomin A toxicity. A Bayesian measurement error linear regression model was derived for each cultivar and trial and demonstrated a clear positive relationship between disease and thaxtomin-A-induced necrosis. The relationships obtained were much stronger than would have been obtained without adjustment for measurement error. This demonstrates that disease mitigation using chemical foliar sprays is strongly correlated with the ability of the chemical to inhibit thaxtomin A toxicity, suggesting this mechanism as a key mode of action for understanding this novel disease control strategy.

A novel strain of Streptomyces (named DS3024) was isolated from a potato field in Michigan in 2006. The taxonomy of the organism was determined by morphology, biochemistry, and genetic analysis. Analysis of the 16S ribosomal RNA gene sequence indicated that the organism was most similar to an isolate of Streptomyces sp., ME02-6979.3a, which is not pathogenic to potato tubers but is distinct from other known pathogenic Streptomyces spp. Strain DS3024 has genes that encode thaxtomin synthetase (txtAB), which is required for pathogenicity and virulence, and tomatinase (tomA), which is a common marker for many pathogenic Streptomyces spp. However, the nec1 gene (associated with virulence in most pathogenic Streptomyces spp.) was not detected. The new strain was capable of growth at pH 4.5, caused necrosis on potato tuber slices, and produced thaxtomin A. In greenhouse experiments, DS3024 caused scab symptoms on potato tubers similar to those caused by Streptomyces scabies on tubers of potato cv. Atlantic, which is scab susceptible. We propose that DS3024 is a new strain of Streptomyces capable of causing common scab on potato tubers. The prevalence of this strain of Streptomyces in potato-producing areas in the north-central United States has not been determined.

In a comparative skeletal muscle study Folkow and Halicka (Microvasc. Res. 1: 1-14, 1968) reported that the capillary filtration coefficient (CFC) of postural (red) muscle was two times the CFC of locomotor (white) muscle. It was concluded that the twofold difference in CFC was due solely to a difference in the perfused capillary surface areas (Sf) of red vs. white muscle. However, CFC is the product of capillary hydraulic conductivity (LP) and Sf. Hence their conclusion assumed that the average LP of red muscle capillaries is exactly equal to the average LP of white muscle capillaries. The following study was undertaken to test the validity of this assumption. The microocclusion procedures and analytical model described by Lee et al. (Circ. Res. 28: 358-370, 1971) and Gore [Am. J. Physiol. 242 (Heart Circ. Physiol. 11): H268-H287, 1982] were used to determine LP. Independent measurements of LP were recorded from single capillaries in red, anterior latissimus dorsi (ALD) and white, posterior latissimus dorsi (PLD) muscles of chickens anesthetized with L.A. Thesia. We found that the mean capillary hydraulic conductivity in postural muscle [(LP)ALD = 0.20 +/- 0.06 (SE) micrometers . s-1 . cmH2O-1 (n = 11)] was significantly different from the mean capillary hydraulic conductivity in locomotor muscle [(LP)PLD = 0.061 +/- 0.01 micrometers . s-1 . cmH2O-1 (n = 14)] (P less than 0.05). These results provide direct evidence that observed differences in red vs. white muscle CFC's may not be due solely to different perfused capillary surface areas but may also be due to differences in capillary hydraulic conductivity.

Object Meningiomas of the lower clivus and foramen magnum are among the most challenging of all neurosurgical lesions. Debate continues regarding the most appropriate approach to this eloquent anatomical region. This anatomical study was undertaken to measure and compare the area of surgical exposures of the lower clivus achieved using the retrosigmoid and the extreme-lateral transcondylar (ELT) approaches. Methods Thirteen embalmed cadaveric heads were dissected bilaterally via the retrosigmoid approach on one side and the ELT approach on the other. The circumference of the area of exposure was delineated using beaded pins placed into the dura. After removal of the brain, the longest longitudinal and transverse axes of the pinned areas were measured and surface area calculated. The area of surgical exposure was also expressed as a percentage of the total area of the lower clivus. Normalized and adjusted surface areas were calculated using the bimastoid diameter. The areas of exposure were compared using the two-tailed paired Student t-test. The mean area of exposure required using the retrosigmoid approach was 19.8 ±14.7 mm2 (range 6–49 mm2) and that using the ELT approach was 27.8 ±22.8 mm2 (range 10–90 mm2). The mean percentage of the lower clivus exposed by the retrosigmoid approach was 14.9 ±3.6% (range 10–22%) and that exposed by the ELT approach was 20.5 ± 4.9% (range 10–25%). The ELT approach provided significantly greater area of operative exposure and allowed a significantly higher percentage of lower clivus and foramen magnum exposure than did the retrosigmoid approach (p <0.05). Normalized and adjusted surface areas, taking into consideration the bimastoid diameter, were also statistically significant in favor of the ELT approach. Conclusions The ELT approach provided a significantly greater area of exposure than did the retrosigmoid approach.

The phytotoxin thaxtomin, produced by plant pathogenic Streptomyces species, is the only known pathogenicity determinant for common scab diseases of potato and other root and tuber crops. Genes encoding thaxtomin synthetase (txtAB) are found on a pathogenicity island characteristic of genetically diverse plant pathogenic Streptomyces species. In this study, an SYBR Green quantitative real-time polymerase chain reaction (PCR) assay using primers designed to anneal to the txtAB operon of Streptomyces was developed to quantify pathogenic bacterial populations in potatoes and soil. The real-time PCR assay was specific for pathogenic Streptomyces strains. The detection limit of the assay was 10 fg of the target DNA, or one genome equivalent. Cycle threshold (Ct) values were linearly correlated with the concentration of the target DNA (correlation coefficient R2 = 0.99) and were not affected by the presence of plant DNA extracts, indicating the usefulness of the assay for quantitative analyses of the pathogenic bacteria in plant tissues. The amount of pathogenic Streptomyces DNA in total DNA extracts from 1 g asymptomatic and symptomatic tubers was quantified using the assay and ranged from 101 to 106 pg. A standard curve was established to quantify pathogenic Streptomyces in soil. Using the standard curve, numbers of pathogenic Streptomyces colony forming units were extrapolated to range from 103 to 106 per gram of soil from potato fields where common scab was found. This real-time PCR assay using primers designed from the txtAB operon allows rapid, accurate, and cost effective quantification of pathogenic Streptomyces strains in potato tubers and in soil.

Potato common scab (CS) is an economically important crop disease that is caused by several members of the genus Streptomyces. In this study, we characterized the plant-pathogenic Streptomyces spp. associated with CS-infected potato tubers harvested in Newfoundland, Canada. A total of 17 pathogenic Streptomyces isolates were recovered from potato scab lesions, of which eight were determined to be most similar to the known CS pathogen S. europaeiscabiei. All eight S. europaeiscabiei isolates were found to produce the thaxtomin A phytotoxin and to harbor the nec1 virulence gene, and most also carry the putative virulence gene tomA. The remaining isolates appear to be novel pathogenic species that do not produce thaxtomin A, and only two of these isolates were determined to harbor the nec1 or tomA genes. Of the non-thaxtomin-producing isolates, strain 11-1-2 was shown to exhibit a severe pathogenic phenotype against different plant hosts and to produce a novel, secreted phytotoxic substance. This is the first report documenting the plant-pathogenic Streptomyces spp. associated with CS disease in Newfoundland. Furthermore, our findings provide further evidence that phytotoxins other than thaxtomin A may also contribute to the development of CS by Streptomyces spp.

Abstract An antegrade approach is frequently used in catheter-directed thrombolysis to remove deep-vein thrombosis. However, the antegrade approach is difficult when accessing veins with small diameters, therefore, understanding the variation of deep calf vein is important. This study measured the diameters and surface areas of the proximal and distal posterior tibial vein, peroneal vein, and anterior tibial vein in order to determine which are preferable for venous access. This study dissected 132 legs from Korean and Thai cadavers. The proximal and distal posterior tibial vein, peroneal vein, and anterior tibial vein were scanned and measured. The mean diameter and surface area were largest for the proximal tibial vein, at 6.34 mm and 0.312 cm2, respectively, followed by the anterior tibial vein (5.22 mm and 0.213 cm2), distal posterior tibial vein (3.29 mm and 0.091 cm2), and peroneal vein (3.43 mm and 0.081 cm2). The proximal posterior tibial vein and anterior tibial vein have large diameters and surface areas, which make them ideal for applying an antegrade approach in catheter-directed thrombolysis. The distal posterior tibial vein and peroneal vein are not recommended due to their smaller surface areas and also the anatomical variations therein.
