AbstractAdequate integrated management of potato leaf diseases can have an influence on pathogens which are challenging to control and cause main potato tuber diseases. Deriving from this hypothesis, an experiment was designed to evaluate the efficacy of foliar application of twelve fungicides on late blight in tubers, common scab, black scurf and silver scurf. In the period of 2013 to 2017, the fungicides were equally applied six times in registered doses during the growing seasons. Eight tuber disease indicators and eight yield characteristics were then evaluated in harvested tuber samples. Significant differences were found among the fungicides used. In comparison to the negative control (n. c.), the application of Infinito, Ranman Top and Revus Top reduced the quantity of tubers infected by late blight by 72% on average. The incidence of common scab increased by 65% of n. c. under the effect of copper oxychloride, while an average reduction by 19% of n. c. was observed with the use of Acrobat MZ WG, Curzate Gold and Consento. The occurrence of silver scurf was significantly enhanced by Curzate Gold and Consento (105% of n. c.), while it was suppressed by Revus Top and Revus (85% of n. c.). All fungicides were ineffective on black scurf yet had a positive influence on the potato yield and quality, with the choice of fungicide having a great impact on the quality of tubers. The application of any safe fungicide is important, especially in years similar to 2015, which was the most devastating recent year for potato production in terms of structural changes in yield of tubers.

AbstractThe technology of hybrid breeding in diploid potatoes creates opportunities to design novel and improved cultivation systems based on hybrid true potato seeds. A promising cultivation pathway to produce seed or ware tubers is by transplanting greenhouse-raised seedlings into the field. This study explored the effects of transplanting date and seedling age on tuber yield, using greenhouse-raised seedlings. Field trials with experimental hybrid genotypes were conducted in three consecutive years. In 2017 and 2018, 4- and 6-week-old seedlings were transplanted at four dates: March, April, May and June. In 2019, transplanting dates included April, May and June and seedling age was 5 weeks. In 2018, the March planting experienced severe frost during the initial field period resulting in crop failure. In 2017 and 2019, plants could withstand shorter and less severe frost events. Seedling age did not significantly affect tuber parameters. Transplanting in June resulted in lower marketable yield (> 28 mm) compared with earlier transplanting dates when crops were harvested in September. At full crop senescence, no differences in marketable yield were observed. The optimal transplanting window, taking into account weather-related risks, is approximately between early April and end May. For some genotypes, crop cycle length was observed to be a more important yield-determining factor than transplanting date.

Abstract Field trials to evaluate the performance of new varieties are an essential component of potato breeding. Besides the genetic differences, environmental factors can lead to variation in a trial. In variety trials, the observed differences amongst varieties should reflect genetic differences, without a large impact of the random or systematic variation in the field. One way to reduce within-field variation is to adjust the plot size and its shape in a trial. Two years of field trials in which individual plants in 90-plant plots of both diploid hybrid and tetraploid varieties were measured provided data to derive relationships between LSD% and plot size and shape. We provide a method to estimate the equations to calculate the expected variation when using different plot dimensions in a relatively homogeneous trial field for tuber yield, tuber volume, tuber count, tuber shape and the standard deviations of tuber volume and shape. Compared with the yield traits, the variation for tuber shape was relatively small. The effect of plot shape was minor. With these equations, breeders can determine what plot dimensions are needed to reach the desired precision for each trait.

Abstract Experiments were carried out in 2012 and 2013 to answer two basic questions in the testing of potato blackleg causing agents before and after harvest. Firstly, what is the spatial distribution of symptomatic plants in the field. Secondly, what is the distribution of infected tubers over the crates and the resulting detection probability using the standard method of collecting 200 tubers from the top crates in storage. In both years, ten farmers were equipped with a global positioning system (Garmin GPSMAP 62) and asked to register the position of blackleg diseased plants every time they scouted their potato lot for diseases. To answer the second question, potatoes marked with four nails (only visible internally after harvest) and potatoes with a different skin colour were added to one-hectare (ha) fields of seed potatoes in different patterns of aggregation ranging from random, to aggregated distribution, up to one big hotspot prior to harvest. The invisibly marked tubers were used for the unbiased collection of twenty 200-tuber samples from the storage crates, while the coloured skin tubers were used to ascertain, when the potatoes were graded, the distribution of ‘infected’ potatoes over the storage crates. The experiment was carried out with 0.05 and 0.1% disease incidence, in 2012 and 2013, respectively. Twenty two out of 26 fields proved to have a random pattern of diseased plants at harvest, which indicates that the blackleg diseased plants came into the field as infected seed potatoes. Two of the four aggregated patterns detected, started out as random distributions but became aggregated later in time, indicating spread in the field. A random spatial pattern in the field at harvest proved to result in a uniform distribution of infected tubers in the crates and, consequently, sampling of only the top crates for the 200-tuber sample does not introduce any bias. Fifty percent of the infected farmer lots were detected by the Nederlandse Algemene Keuringsdienst inspectors performing their official field surveys, which was a better performance than the 18% detection obtained by the standard 200-tuber sampling method. Only 6 out of 80 samples from the ‘infected’ lots with 0.05% disease incidence level, and 22 out of 80 samples at the 0.1% disease incidence level were detected by the latter method. It was concluded that intensifying the field survey would be cheaper and more successful than enlarging the tuber sample size to increase the probability for detection of infected seed lots.

AbstractIn organic potato production, the need for varieties with durable late blight resistance developed through classical breeding programmes is urgent. Besides late blight resistance, other variety characteristics needed in organic potato production are early canopy closure for weed suppression and good tuber dormancy to eliminate the need for (chemical) sprouting inhibition during storage, amongst others. This paper is a unique example of collaboration between researchers, farmers and professional breeders of both large, medium and small breeding companies. The aim of the resulting breeding project, Bioimpuls, was to provide a substantial impulse to both the organic and conventional potato breeding sector by enlarging the access to various sources of late blight resistance. The Bioimpuls activities include providing true seed populations for variety selection with five available sources of R-genes against Phytophthora infestans, early and advanced introgression breeding with six new R-genes, and education and communication. The results achieved over the 11-year period (2009–2019) are analysed. Many true seed populations containing multiple resistance genes are produced and selected, and a constant flow of breeding clones is entering the evaluation and positioning trials of companies. However, it will still take a considerable amount of time before varieties with stacked resistance genes will replace the new resistant single gene varieties entering the market in the next few years. Five out of six new sources of R-genes need more years of backcrossing before they are ready for commercial use. Bioimpuls successfully introduced a training course for farmer breeders, and published a manual for potato breeding.

AbstractTo contribute to the development of a novel cropping system for potato grown from greenhouse-derived seedlings from hybrid true potato seeds, planting density trials were carried out under normal Dutch agronomic conditions. For two consecutive years, 5-week-old seedlings of two experimental genotypes were transplanted into farmers’ potato production fields at two contrasting locations: a flat-bed system on sandy soil and a traditional ridge system on clay soil. Planting densities were 6.25, 12.5, 25, 50, 100 and 200 plants/m2 in the flat-bed system, and 3.125, 4.688, 6.25, 12.5, 25 and 50 plants/m2 in the ridge system. In general, increasing planting density of hybrid seedlings per area decreased tuber fresh weight per plant and reduced the number of tubers per plant. On a per hectare basis, an increased planting density resulted in increased total tuber yield and number of tubers up to very high densities, but finally both parameters levelled off. Highest total tuber yields harvested were 107 and 45 Mg/ha for the flat-bed and ridge system, respectively. On flat-beds, the optimal planting density for total yield was 50 plants/m2. On ridges, planting density interacted with year and genotype, resulting in an optimum planting density of 25 plants/m2 to reach the maximum total yield. Obtained yields in the commercial size classes Baby Baker (20 < size class ≤ 35 mm) and Seed Tubers (28 < size class ≤ 50 mm) were in general very high on the flat-beds, with a maximum Seed Tuber yield of 64 Mg/ha at 50 plants/m2. The current study showed that transplanted hybrid seedlings are feasible alternatives for seed-tuber-grown systems for certain potato outlets.

AbstractTranscriptomic changes in plants during drought stress give insights into the mechanisms with which plants stabilise their metabolic processes in order to cope with the drought condition. In potato, understanding such drought-induced transcriptomic changes is critical because prolonged field drought interferes with tuber formation and bulking period of potato development, which eventually affects yield. We hypothesised that phenotypic drought responses of potato genotypes may be linked to differences in transcriptomic changes. Using an RNA sequencing approach, we investigated such transcriptomic changes in leaves of three cultivars (Biogold, Hansa and Lady Rosetta) under drought. We found more differentially expressed genes (DEGs) in the tolerant cultivars, Lady Rosetta and Biogold, than in the sensitive cultivar (Hansa). The differential gene expression trend reflected the phenotypic drought responses of the cultivars. For instance, we found in both Biogold and Lady Rosetta but not in Hansa, an upregulation of genes involved in carbohydrate metabolism (e.g., Alpha-glucosidase), flavonoid biosynthesis (e.g., Flavanone 3 beta-hydroxylase), lipid biosynthesis/transfer (e.g., nonspecific Lipid Transfer Proteins), heat shock proteins and secondary metabolites like phenolics and lignins. Furthermore, a prolonged drought stress resulted in reduced DEGs in Biogold and Hansa, but not in Lady Rosetta that also maintained its tuber yield under such prolonged stress suggesting a more robust drought tolerance. Our findings suggest that a synergistic expression of genes involved in several different aspects of drought response is required in order to obtain a robust tolerance.
