An attempt has been made to develop a simple, sensitive and rapid high performance liquid chromatographic (HPLC) method of analysis for sparfloxacin using 35% acetonitrile in buffer solution as mobile phase. Buffer solution (40 mM of sodium dihydrogen phosphate in de-ionized water) was used as solvent to dissolve sparfloxacin and 0.05mg/ml stock solution was prepared. Sparfloxacin solution was scanned with UV-spectrophotometer and the absorption maximum (λmax) was found at 292 rim. This method was successfully applied to four tablet dosage forms of sparfloxacin encoded as ?, ?, ?, and ?, from four different companies and the result was found to be satisfactory and reproducible. A rapid and reproducible reverse phase high performance liquid chromatographic method has been developed for the estimation of lamotrigine in its pure form as well as in pharmaceutical dosage forms. Chromatography was carried out on a Luna C18column using a mixture of potassium dihydrogen phosphate buffer (pH 7.3) and methanol in a ratio of 60:40 v/v as the mobile phase at a flow rate of 1.0 mL/min. The detection was done at 305 nm. The retention time of the drug was 6.1 min. The method produced linear responses in the concentration range of 10 to 70 μg/mL of lamotrigine. The method was found to be reproducible for analysis of the drug in tablet dosage forms.

A simple, selective, accurate High Performance Liquid Chromatographic (HPLC) method was developed and validated for the analysis of Fenspiride hydrochloride in bulk and tablet dosage forms. Chromatographic separation was achieved isocratically using a C18 reverse phase column [Inertsil C18 column (250×4.6mm, 5μm)] utilizing a mobile phase containing 10mM Ammonium acetate: Acetonitrile (50:50 v/v) at a flow rate of 1 mL/min. The eluents were monitored at wavelength of 210 nm for a run time of 7 minutes at ambient temperature. The average retention time of the drug was found to be 4.6 minutes. The developed method was validated as per ICH guidelines to ascertain the reproducibility of the method. The method was found to be linear in the concentration range of 10-50 μg/mL with a good correlation coefficient of 0.998. The limit of detection (LOD) and limit of quantification (LOQ) were 0.007 and 0.021 μg/mL and the percentage recovery and assay were found to be 99.315 and 98.97%. Specificity with placebo by 3 D plots showed that the method was specific and free from interfering substances. Therefore, the fully validated method was good enough to carry out routine analysis of Fenspiride in bulk and tablet formulations.

Objective: To develop and validate reverse phase-high performance liquid chromatographic method for estimation of olanzapine in bulk and tablet dosage form.Methods: Chromatographic analysis was performed on XTerra C18 (150×3.5 mm inner diameter, 5 μm) column using a mobile phase consisting of buffer (potassium dihydrogen phosphate) and methanol (45:55% v/v) with a flow rate of 0.6 ml/minutes. The detection was carried out at 247 nm.Results: The calibration curve of olanzapine was linear in the range of 30-70 μg/ml. The mean % assay of marketed formulation was found to be 100.2%, and % recovery was observed in the range of 98-102%. Relative standard deviation for the precision study was found <2%.Conclusion: The developed method is simple, precise and rapid, making it suitable for estimation of olanzapine in bulk and tablet dosage form.Keywords: Olanzapine, Reverse phase-high performance liquid chromatographic, Validation.

A simple, sensitive, precise and specific reverse phase high performance liquid chromatographic method was developed and validated for the determination of Telmisartan in bulk and tablet dosage forms. It was found that the excipient in the tablet dosage forms does not interfere in the quantification of active drug by proposed method. The HPLC separation was carried out by reverse phase chromatography on RP18, column (250×4.6mm) with a mobile phase composed of 0.025M potassium dihydrogen phosphate : acetonitrile : methanol (45:50:5) at a flow rate of 1ml/min. The detection was monitored at 216 nm. The calibration curve for Telmisartan was linear from 100 to 500 ng/ml. The interday and intraday precision was found to be within limits. LOD and LOQ for Telmisartan were found to be 27 ng/ml and 83 ng/ml. Accuracy and reproducibility were also found within range. The proposed method has adequate sensitivity, reproducibility and specificity for the determination of Telmisartan in bulk and its tablet dosage forms.

A simple, sensitive, precise, selective reverse phase high performance liquid chromatographic method was developed and validated for erlotinib hydrochloride in tablet dosage form.(0.02M)The separation was achieved on C18 column (150mm×4.6mm.i.d.,5.0μm) using potassium dihydrogen phosphate: acetonitrile in the ratio 50:50v/v as mobile phase having pH 4.5 was adjusted with methanol and flow rate 1ml/min. Detection was carried out using a UV detector at 248nm. The column temperature was adjusted at 30ᵒC. The method was validated for precision, linearity and range, stability and robustness. The developed and validated method was successfully applied for the quantitative analysis of ERLONAT tablets. The total chromatographic analysis time per sample was about 7min with Erlotinib eluting at 6.547min.Validation studies demonstrated that this HPLC method is simple, specific, rapid, reliable and reproducible. The standard curves were linear over the concentration ranges, 88.32- 132.48μg/ml for erlotinib. The high recovery confirms the suitability of the proposed method for the determination of Erlotinib in ERLONAT tablets. The results of analysis have been validated according to ICH guideline requirements. The method can be applied for Erlotinib hydrochloride tablets.

A simple, sensitive, accurate and rapid reverse phase high performance liquid chromatographic method is developed for the simultaneous estimation of ramipril, aspirin and atorvastatin in pharmaceutical preparations. Chromatography was performed on a 25cm×4.6 mm i.d, 5µm particle, C18 column with Mixture of (A) acetonitrile methanol (65:35) and (B) 10 mM sodium dihydrogen phosphate monohydrate (NaH2PO4.H2O) buffer and mixture of A:B (60:40 v/v) adjusted to pH 3.0 with o-phosphoric acid (5%v/v) was used as a mobile phase at a flow rate of 1.5 ml min-1. UV detection was performed at 230 nm. Total run time was less then 12 min; retention time for Ramipril, aspirin and Atorvastatin were 3.620, 4.920 min and 11.710 min respectively. The method was validated for accuracy, precision, linearity, specificity and sensitivity in accordance with ICH guidelines. Validation revealed that the method is specific, rapid, accurate, precise, reliable, and reproducible. Calibration plots were linear over the concentration ranges 05-50 µg mL-1for Ramipril, 05-100 µgmL-1for aspirin and 02-20 µg mL-1for atorvastatin. Limits of detection were 0.014, 0.10 and 0.0095 ng mL-1limits of quantification were 0.043, 0.329 and 0.029 ng mL-1for ramipril aspirin and atorvastatin respectively. The high recovery and low coefficients of variation confirm the suitability of the method for simultaneous analysis of the all three drugs in the dosage forms. The validated method was successfully used for quantitative analysis of marketed pharmaceutical preparations.

A simple precise and rapid Reverse Phase High Performance Liquid Chromatographic (RP-HPLC) method has been developed for quantitative determination of Regorafenib in tablet dosage form. In this method Hypersil Gold (C18, 150mm× 4.6mm id, 3μ) column with mobile phase consisting of Trifluoroacetic acid (0.2% v/v) and Acetonitrile in the ratio of (50: 50 v/v) at 400C in an isocratic mode was used. The detection was carried out at 260 nm and 20μL injection volume was selected with the flow rate 1mL/min. The linearity range of Regorafenib shows concentration between 5-200 μg/mL. The regression coefficient obtained was 0.999. Retention time of Regorafenib was found to be 6.49 minutes. Acetonitrile and Water in the ratio of (3:1) was used as a diluent. The method was validated as per ICH guidelines and is simple, fast, accurate, precise and can be applied for routine quality control analysis of Regorafenib in tablet dosage form.

A simple, accurate, precise high-performance liquid chromatographic (HPLC) method has been developed and validated for the simultaneous determination of abacavir sulphate (ABA) and lamivudine (LAM) in combined dosage form. Separation was performed on a C18 column [Agilent ODS UG 5 column, 250 mm x 4.5 mm], with methanol: water (50:50 V/V) isocratic elution using a flow rate of 1mL/min. Good sensitivity was observed with UV detection at 277 nm. After method development, the interference of other active compounds and excipients, repeatability and linearity, were investigated. Retention times of LAM and ABA were found to be 3.3 and 6.3 min, respectively. The method was validated over the range from 2.5-12.5 μg/mL for LAM and 5-25 μg/mL for ABA with correlation coefficients of 0.9997 and 0.9996, respectively. This method was shown to be accurate, robust, selective, linear, and repeatable and can be successfully employed in routine quality control for the simultaneous analysis of ABA and LAM in tablets.

A reverse phase high performance liquid chromatographic method was developed for the simultaneous determination of simvastatin and ezetimibe in tablet dosage forms. The separation was effected on a C18 Supelcosil column (250 mm x 4.6 mm; 5µ) using a mobile phase consisting of 0.01 M ammonium acetate buffer and acetonitrile (35:65v/v) at a flow rate of 1 mL/min. The detection was made at 240 nm. The retention times for ezetimibe and simvastatin were 5.9 and 8.5 min respectively. Calibration curves were linear over the ranges of 0.5-40 µg/mL for simvastatin and 2.5-50 µg/mL for ezetimibe. The proposed method was validated as per the ICH and USP guidelines. The method is accurate and precise and found to be suitable for the quantitative analysis of both the drugs individually and in combination in tablet dosage forms.

A simple, economic, accurate, sensitive, specific and precise stability indicating reverse phase high performance liquid chromatographic [RP-HPLC] method for the determination of Regorafenib in pure and tablet dosage from was developed and validated. The chromatographic separation was carried out using Phenomenex Luna-C18 column (4.5x250 mm; 5 µm particle size) as a stationary phase and methanol: acetonitrile: water (55:25:20 v/v/v) as a mobile phase. The flow rate of 1 mL/min was used with PDA detection at 275 nm. The retention time of Regorafenib was 2.480 min. RP-HPLC method was developed with linearity range of 40-240 µg/mL of Regorafenib. The correlation coefficient [r2] was found to be 0.9999. The assay results obtained was in good agreement with the corresponding labeled amount by developed method within range of 98.83 ± 0.6937. Accuracy of the method was confirmed by recovery studies and the recoveries were found to be between 99.61 % and 100.22 %, the corresponding %RSD was found to be 0.2029. Precision, LOD, LOQ, specificity, robustness and ruggedness were performed as per ICH Q2(R1) guidelines and were within the acceptance criteria. This method can be conveniently used to detect the possible degradation product in the dosage form of Regorafenib during stability studies (acidic, alkaline, oxidative, thermal and photolytic). The method proved to be effective on the analysis of stressed marketed tablet formulation.
