Vasoactive intestinal polypeptide (VIP) is implicated in the modulation of vagal effects on the heart rate. In this study, the impact of acute and chronic atropine administration on VIP levels in rat heart atria was investigated in relation to heart rate in the course of vagus nerves stimulation. Anaesthetised control and atropinised (10 mg/kg/day for 10 days) rats pretreated with metipranolol and phentolamine that were either given or not a single dose of atropine were subjected to bilateral vagus nerve stimulation (30 min: 0.7 mA, 20 Hz, 0.2 ms). VIP concentrations in the atria were determined after each stimulation protocol. In control rats with or without single atropine administration, the heart rate upon vagal stimulation was higher than in atropinised animals with or without single atropine dose, respectively. VIP concentrations in the control atria were significantly decreased after the stimulation; the decrease was comparable both in the absence and presence of a single dose of atropine. Compared to controls, VIP levels were significantly decreased after chronic atropine treatment and they were not further reduced by vagal stimulation and single atropine administration. Administration of VIP antagonist completely abolished the differences in the heart rate upon vagal stimulation between control and atropinised groups. In conclusion, the data indicate that chronic atropine administration affects VIP synthesis in rat heart atria and consequently it modifies the heart rate regulation.

It is well known that the GABAergic and noradrenergic systems play an important role in blood pressure and heart rate regulation. Benzodiazepines and beta-carbolines, respectively, increase or decrease the probability of chloride-channel opening induced by GABA. The aim of this study was to determine, in conscious rats, the interaction existing between the central alpha2-adrenoceptor stimulation induced by clonidine and the facilitation or impairment of benzodiazepine receptor activity through the administration of either diazepam, a benzodiazepine receptor agonist, or methyl 6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate (DMCM), an inverse benzodiazepine agonist. Clonidine (5-10 µg, intracerebroventricularly) reduced heart rate and increased mean blood pressure by activation of central alpha2-adrenoceptors. Diazepam (2 mg/kg, intravenously (i.v.)) induced an increase in heart rate, while DMCM (0.3 mg/kg, i.v.) elicited a bradycardic effect. The bradycardic effects induced by both clonidine and DMCM were antagonized by the prior administration of methylatropine (1.5 mg/kg, i.v.). DMCM (0.3 mg/kg, i.v.) prevented the clonidine effects on heart rate and mean blood pressure, while diazepam (2 mg/kg, i.v.) failed to modify these effects. Our results suggest that the bradycardic effects of clonidine are mediated by a vagal stimulation and are related to the activation of a GABAergic pathway.Key words: blood pressure, clonidine, diazepam, methyl 6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate (DMCM), heart rate.

Mechnikov Objective to study the features of autonomic regulation of sinus rhythm features of autonomic regulation of cardiac sinus rhythm in patients with Takotsubo syndrome (TTS) during inpatient stay in hospital. Materials and methods. The study included 25 female patients (the median age 57.5 ± 28.5 years) with TTS, diagnosis was based on the InterTAK criteria. On the 5-7th day after admission, the heart rate variability (HRV) was assessed both at rest and cardiac autonomic function tests: the deep breathing test (DBT) and active orthostatic test (AOT) with the usage of a computer diagnostic system «Valenta». RESULTS According to HRV data, initially at rest, the majority of the studied patients predominantly had a parasympathetic impact on the heart rate regulation: in 70% - increased parasympathetic influence, in 17% - increased sympathetic influence, in 13% - a balanced heart rate regulation. When performing a DBT, the predominance of the parasympathetic influence on the heart rate was noted both in spectral parameters and according to the data of time analysis (HF  during DBT  increased 1.8 times  up to  489.6 ± 114.2 ms2 (p <0.05) ; LF  decreased 1.7 times  up to 254.1 ± 70.3 ms2 (p <0.05); VLF  increased 2.8 times up to 402.3 ± 101.8 ms2 (p <0, 05); TP  increased 1.4 times up to 1145.9 ± 251.3 ms2; CV increased 1.3 times up to 6.5 ± 0.7% (p <0.05); SDNN increased 1.3 times up to - 58.7 ± 6.6 ms (p <0.05). HRV measured during an AOT  showed a paradoxical  evaluation  in the parasympathetic impact on the heart rate (HF increased by 1.8 times and reached - 488.0 ± 104.7 ms2 (p <0.05); LF decreased by 1.1 times up to  408 , 4 ± 103.6 ms2; VLF increased 1.2 times  up to  166.4 ± 52.2 ms2; TP increased 1.3 times  up to 1062.8 ± 181.1 ms2 (p <0.05); CV increased 1.6 times  up to 8.0 ± 0.9% (p <0.05); SDNN increased 1.4 times  up to 62.6 ± 7.5 ms (p <0.05)). Conclusions During the index hospital inpatient stays the majority of patients with TTS have a vegetative imbalance due to increased parasympathetic influence on the heart rate, both at rest and  during a deep breathing test and an active orthostatic test. This fact is quite unexpected and requires further study.
