Passion fruit (Passiflora edulis) is a tropical and subtropical plant that is widely cultivated in China due to its high nutritional value, unique flavor and medicinal properties. In August 2020, typical anthracnose symptoms with light brown and water-soaked lesions on Passiflora edulis Sims were observed, which result in severe economic losses. The incidence of this disease was approximately 30%. The pathogens from the infected fruit were isolated and purified by the method of tissue isolation. Morphological observations showed that the colony of isolate BXG-2 was gray to celadon and grew in concentric circles. The orange conidia appeared in the center after 14 days of incubation. The pathogenicity was verified by Koch’s postulates. The internal transcribed spacer (ITS), chitin synthase (CHS-1), actin (ACT), and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) were amplified by relevant PCR programs. The multi-gene (ITS, GAPDH, ACT, CHS-1) phylogeny analysis confirmed that isolate BXG-2 belongs to Colletotrichum fructicola. The inhibitory effect of six synthetic fungicides on the mycelial growth of the pathogen was investigated, among which difenoconazole 10% WG showed the best inhibitory effect against C. fructicola with an EC50 value of 0.5579 mg·L−1. This is the first report of anthracnose on Passiflora edulis Sims caused by Colletotrichum fructicola in China.

Green manures can replace or supplement mineral fertilization and add organic matter to the soils, ensuring greater sustainability to fruit growing in semiarid regions. Biological fixation, transfer and balance of nitrogen were determined on an irrigated yellow passion fruit orchard (Passiflora edulis Sims) intercropped separately with three cover crops: sunn hemp, Crotalaria juncea (L.); pigeon pea, Cajanus cajan (L.) Mill; and jack bean, Canavalia ensiformis (L.) DC. In a fourth treatment, legumes were not planted, but spontaneous vegetation was left to grow freely. The legumes were croped for 90 days in three lines (0.5 m apart) inside the passion fruit plant lines (2.5 m apart). Fixation and transfers were determined by the 15N natural abundance technique, using sunflower as a reference plant. The three planted legumes nodulated abundantly and fixed nitrogen in high proportions (between 50 and 90% of their N), forming symbiosis with bacteria naturally established in the soil. Jack bean produced more biomass than sunn hemp and pigeon pea, and as much as the spontaneous plants, of which 23% were legumes. The amounts of fixed N (150, 43, 30 and 29 kg ha-1) were determined mainly by the biomass of legumes. More than 40% of the N of passion fruit plants came from the biological nitrogen fixation of the intercropped jack bean, which provided an amount of N higher than that exported in the fruits, generating a positive balance of more than 100 kg ha-1. Therefore, it is recommended to intercrop jack bean in irrigated passion fruit orchards.

AbstractPseudorabies, caused by pseudorabies virus (PRV) variants, has broken out among commercial PRV vaccine-immunized swine herds and resulted in major economic losses to the pig industry in China since late 2011. However, the mechanism of virulence enhancement of variant PRV is currently unclear. Here, a recombinant PRV (rPRV HN1201-EGFP-Luc) with stable expression of enhanced green fluorescent protein (EGFP) and firefly luciferase as a double reporter virus was constructed on the basis of the PRV variant HN1201 through CRISPR/Cas9 gene-editing technology coupled with two sgRNAs. The biological characteristics of the recombinant virus and its lethality to mice were similar to those of the parental strain and displayed a stable viral titre and luciferase activity through 20 passages. Moreover, bioluminescence signals were detected in mice at 12 h after rPRV HN1201-EGFP-Luc infection. Using the double reporter PRV, we also found that 25-hydroxycholesterol had a significant inhibitory effect on PRV both in vivo and in vitro. These results suggested that the double reporter PRV based on PRV variant HN1201 should be an excellent tool for basic virology studies and evaluating antiviral agents.

AbstractAleutian mink disease (AMD), which is caused by Aleutian mink disease virus (AMDV), is an important contagious disease for which no effective vaccine is yet available. AMD causes major economic losses for mink farmers globally and threatens some carnivores such as skunks, genets, foxes and raccoons. Aptamers have exciting potential for the diagnosis and/or treatment of infectious viral diseases, including AMD. Using a magnetic beads-based systemic evolution of ligands by exponential enrichment (SELEX) approach, we have developed aptamers with activity against AMDV after 10 rounds of selection. After incubation with the ADVa012 aptamer (4 μM) for 48 h, the concentration of AMDV in the supernatant of infected cells was 47% lower than in the supernatant of untreated cells, whereas a random library of aptamers has no effect. The half-life of ADVa012 was ~ 32 h, which is significantly longer than that of other aptamers. Sequences and three dimensions structural modeling of selected aptamers indicated that they fold into similar stem-loop structures, which may be a preferred structure for binding to the target protein. The ADVa012 aptamer was shown to have an effective and long-lasting inhibitory effect on viral production in vitro.

O alagamento no solo é um fenômeno que reduz a sobrevivência, o crescimento e o desenvolvimento de plantas, podendo ocasionar queda na produtividade quando essas são agricultáveis. O maracujazeiro (Passiflora edulis Sims) é uma frutífera comercial com propriedades medicinais cuja produção incentiva a irrigação; e não são encontrados estudos que apresentem efeitos do excesso de água no solo sobre estas plantas. O objetivo deste trabalho foi verificar alterações morfoanatômicas e fisiológicas no desenvolvimento inicial de plântulas de maracujazeiro expostas a diferentes níveis de água no solo. As plântulas foram organizadas em três tratamentos: solo com capacidade de campo, pré-submergido e alagado. Após sete dias, foram feitas análises de trocas gasosas, potencial hídrico, quantificação de prolina e cortes anatômicos de raízes. O solo alagado influencia as trocas gasosas e afeta negativamente a fotossíntese. Não houve alterações no potencial hídrico nem na quantificação de prolina entre os tratamentos. Os tratamentos pré-submergido e alagado apresentaram aerênquima e aumento no diâmetro das raízes e na espessura das barreiras apoplásticas e do córtex. O maracujazeiro apresentou alterações fisiológicas e anatômicas que permitiram sua sobrevivência em condições pré-submergidas e alagadas, o que significa que plântulas jovens dessa espécie suportam maiores quantidades de água no solo.

Este trabalho foi desenvolvido com os objetivos de avaliar os efeitos imediato e latente da temperatura de secagem e de dois tipos de embalagem sobre a qualidade fisiológica de sementes de maracujá-amarelo após três e seis meses de armazenamento. Empregaram-se quatro níveis de temperatura do ar de secagem (ambiente à sombra, 30, 35 e 40 ºC), um nível de fluxo de ar, 48 m³ min-1 m-2, e teores de água inicial e final de cerca de 35 e 10 % b.u., respectivamente. Os tratamentos de secagem foram realizados em protótipo de secador de leito fixo, em camada delgada, com fluxo de ar ascendente. Avaliou-se a qualidade fisiológica por meio de testes de germinação e vigor. Depois da secagem, as sementes foram embaladas e armazenadas em câmara do tipo B.O.D. a 15±1 ºC. As temperaturas utilizadas na secagem das sementes não influenciam a germinação. O armazenamento por três e seis meses não afetou a qualidade fisiológica das sementes. A secagem das sementes de mamão pode ser realizada à sombra ou em temperaturas de 30 a 40 ºC sem afetar a qualidade fisiológica das mesmas.

No Cerrado brasileiro, há uma grande diversidade de cores, tamanhos e aromas de frutos em acessos silvestres de P. edulis. Estes acessos também são importantes fontes de resistência a doenças, podendo ser incorporados em programas de melhoramento genético do maracujazeiro azedo. Neste trabalho, objetivou-se estimar a variabilidade genética existente em acessos silvestres e comerciais de P. edulis utilizando-se de marcadores RAPD. O DNA genômico de cada acesso foi extraído e amplificado com treze iniciadores decâmeros (OPD-04, OPD-07, OPD-08, OPD-16, OPE-18, OPE-20, OPF-01, OPF-14, OPG-05, OPG-08, OPH-04, OPH-12 e OPH-16) para a obtenção dos marcadores RAPD. Os marcadores obtidos foram convertidos em uma matriz de dados binários, a partir da qual foram estimadas as distâncias genéticas entre os acessos e realizadas análises de agrupamento e de dispersão gráfica. Um total de 187 marcadores foi gerado, sendo que apenas 28 (14,97%) deles foram monomórficos. As distâncias genéticas entre os 15 acessos de maracujazeiro variaram de 0,091 a 0,496. Os marcadores moleculares demonstraram a alta variabilidade genética dos acessos de P. edulis, sendo que os acessos de frutos amarelos apresentaram maior distanciamento em relação aos de frutos roxos. Menores distâncias genéticas foram verificadas entre os acessos de mesma origem geográfica.
