The genetic causes of congenital heart diseases especially cardiac valve disorders are mostly unknown. During the last decade, the zebrafish became an excellent and established model organism (1) to uncover these genetic defects and (2) to elucidate the underlying molecular pathomechanisms. We recently isolated the zebrafish mutation ping pong ( png m683 ) in a large-scale ENU-mutagenesis screen for recessive lethal mutations that perturb cardiac function. png mutant zebrafish embryos show pathologically developed cardiac valves. Due to malformation of the cardiac AV valves, png mutant zebrafish embryos exhibit vigorous regurgitation of blood between the atrium and the ventricle. Furthermore, as a result of the cardiac valve malformation and cardiac dysfunction png mutants die at day 6 post fertilization. Expression of several factors known to be crucial for the proper development and formation of the atrio-ventricluar canal (e.g. notch1b, bmp4 or versican) is significantly altered in png mutant zebrafish hearts. By a positional cloning approach we demonstrate that the ping pong phenotype is caused by a promotor mutation in a zebrafish gene encoding for a novel component of the “transcriptional mediator complex”. This mediator complex is a multi-protein complex that acts as a transcriptional coactivator and transduces informations from transcription factors to the RNA polymerase II. png is strongly expressed in zebrafish as well as human cardiomyocytes. Furthermore, sequence alignments demonstrate the evolutionary conservation of the ping pong gene product. Gene specific knock-down studies by means of modified antisense oligonucleotides reveal a phenocopy of the png mutant phenotype whereas injection of the gene-specific mRNA in png mutant embryos restores the mutant phenotype indicating that png is indeed responsible for the observed phenotype. The zebrafish evolved as an excellent model organism to study the molecular signalling pathways involved in cardiac valve formation. By detailed characterization of the zebrafish line ping pong we will obtain new insights into these molecular mechanisms especially the transcriptional control of valve formation and therefore the pathomechanisms of human cardiac valve disorders.

Abstract Currently drilling of horizontal wells is a common enhanced oil recovery method. Geosteering services are often used for accurate well placement, which makes it possible to achieve a significant increase in production at relatively low cost. This paper describes the result of using seismic data in three-dimensional visualization for high-quality geosteering using a deep boundary detection tool and multilayer inversion in real time. Crossing the top of the reservoir while drilling horizontal sections at the current oilfield is unacceptable, due to the presence of reactive mudstones. In case of crossing the top of reservoir, further work on running and installing the liner becomes impossible due to instability and may lead to well collapse. Based on prewell analysis of the structural data, the well was not supposed to approach the top of the target formation along the planned profile. However, while preparing geosteering model and analyzing seismic data it became possible to reveal that risk, elaborate its mitigation and eventually increase the length of the horizontal section. Such integrated analysis made it possible to maintain the wellbore within the target reservoirs, as well as to update the structural bedding of the top based on the multilayer inversion results.

Abstract The Well X in Nong Yao field, is an infill-well designed for the Gulf of Thailand which presented several interesting challenges due to its complexity, tortuosity, and potential collision risks with other wells. This paper demonstrates the application of a Real-time Advanced Survey Correction (RASC) with Multi Station Analysis (MSA) to correct the Measurement While Drilling (MWD)'s azimuth. The Well X is a 3D Complex design with a high drilling difficulty index (DDI) at 6.9, high tortuosity of 316 degree, and which has an aggressive build on inclination and azimuthal U-turning well path. The well also creates difficult doglegs severity (DLS) up to 5.5deg/100ft, which is near the limit of the flexibility required to achieve the horizontal landing point. The conventional MWD survey, with proximity scanning with the nearby Well A, demonstrates high risk with a calculated Oriented Separation Factor (OSF) of 1.01. The RASC-MSA method is applied with a clearly defined workflow during execution in real-time and provide significant improvement in calculated OSF. RASC-MSA is applied for every 1,000 ft interval drilling below the 9.625in casing shoe. The workflow ensures that the directional driller follows the corrected survey along the well path and especially in the last 300 ft before reaching the electrical submersible pump (ESP) tangent section. The result from RASC-MSA, indicated a 29 ft lateral shift on the left side of the MWD standard surveys. Without this technique, Well X has a high potential to collide with Well A and Well B (Figure 1) as the actual OSF may less than 1 while drilling. The final 3D Least Distance proximity scanning with Well A shows a minimum OSF = 1.35, which is a 30% improvement compared to the conventional MWD survey. Another nearby well, Well B, indicates a minimum OSF=1.66 and passed the anti-collision OSF rule. In consideration of the drilling efficiency, availability, cost effectiveness and time saving, the RASC-MSA analysis to correct the MWD's azimuth are applied and the separation factor can be improved by 30%. In conclusion, the collision risk management technique applied successfully met the complex challenges of Well X, which was successfully drilled and safely delivered. This paper introduced the laboratory model of Real-time monitor system based on the 3D Visualization for calefaction furnace, depicted the process of the model.In this paper we created a virtual environment and transport the real-time data which we collected from the locale to the virtual scene,to realize the real time monitor on the real environment.Through simulating in the lab,the effect of this system was realistic at the same time it arrived at the goal of better monitoring with better real-time.

For improved prevention and reduction of marine disasters, China’s marine authorities and emergency response agencies require a solution that provides risk assessment, early warning, and decision-making support. This paper proposes a comprehensive approach to disaster assessment that involves automated long-term operation, a spatial information visualization method and systematic integration. The proposed approach provides functions for numerical ocean models with forecast results, automated processing of massive data, multiple disaster/element coupled assessment, and multidimensional display and expression. With regard to storm surge disasters, the approach proposed in this paper adopts a four-tier structure and the functions of each tier are described separately. The original data are comprised of a combination of statistical analysis data and real-time data obtained from the unstructured grid Finite Volume Community Ocean Model. Automated data processing methods and assessment theories incorporating an indicator system and weighted parameters are used for the assessment. By applying 2D/3D visualization technology, assessment results are displayed via several modes for ease of operation and comprehension. The validity of the approach was verified by applying it to Typhoon Hato (No. 1713). Compared with the results of the post-disaster investigation, the assessment results of the proposed approach proved the reliability of the system.
