AbstractDielectric substances exhibit great potential for high-power capacitors due to their high stability and fast charge–discharge; however, a long-term challenge is to enhance energy density. Here, we propose a poly(vinylidene fluoride) (PVDF) composite utilizing BaTiO3 nanoparticle@TiO2 nanosheet (BT@TO ns) 2D nanohybrids as fillers, aiming at combining the interfacial strategy of using a core–shell filler and the electron scattering of a 2D filler to improve the energy density. With 4 wt% filler, the composite possesses the largest breakdown strength (Eb) of 561.2 MV m−1, which is significantly enhanced from the 407.6 MV m−1 of PVDF, and permittivity of 12.6 at 1 kHz, which is a 23% increase from that of PVDF. A superhigh energy density of 21.3 J cm−3 with an efficiency of 61% is obtained at 550 MV m−1. The 2D BT@TO ns-filled composite exhibits a higher energy density than composites filled with core–shell 1D BT@TO nws or non-core–shell 0D BT, 1D TO, or 2D TO particles. The Eb and energy density improvements are attributed to the buffer layer-based interface engineering and enhanced area scattering of electrons caused by the 2D hybrids, an effect similar to that of a ping-pong paddle to scatter electric field-induced charge migrations in composites. Thus, an effective hybrid strategy is presented for achieving high-performance polymer composites that can be used in energy storage devices.

Abstract Over the last decades, light-emitting diodes (LED) have replaced common light bulbs in almost every application, from flashlights in smartphones to automotive headlights. Illuminating nightly streets requires LEDs to emit a light spectrum that is perceived as pure white by the human eye. The power associated with such a white light spectrum is not only distributed over the contributing wavelengths but also over the angles of vision. For many applications, the usable light rays are required to exit the LED in forward direction, namely under small angles to the perpendicular. In this work, we demonstrate that a specifically designed multi-layer thin film on top of a white LED increases the power of pure white light emitted in forward direction. Therefore, the deduced multi-objective optimization problem is reformulated via a real-valued physics-guided objective function that represents the hierarchical structure of our engineering problem. Variants of Bayesian optimization are employed to maximize this non-deterministic objective function based on ray tracing simulations. Eventually, the investigation of optical properties of suitable multi-layer thin films allowed to identify the mechanism behind the increased directionality of white light: angle and wavelength selective filtering causes the multi-layer thin film to play ping pong with rays of light.

Homeostasis plays a central role in our understanding how cells and organisms are able to oppose environmental disturbances and thereby maintain an internal stability. During the last two decades there has been an increased interest in using control engineering methods, especially integral control, in the analysis and design of homeostatic networks. Several reaction kinetic mechanisms have been discovered which lead to integral control. In two of them integral control is achieved, either by the removal of a single control species E by zero-order  kinetics ("single-E controllers"), or by the removal of two control species  by second-order kinetics ("antithetic or dual-E control"). In this paper we show results when the control species E 1  and E 2  in antithetic control are removed enzymatically by ping-pong or ternary-complex mechanisms. Our findings show that enzyme-catalyzed dual-E controllers can work in two control modes. In one mode, one of the two control species is active, but requires zero-order kinetics in its removal. In the other mode, both controller species are active and both are removed enzymatically. Conditions for the two control modes are put forward and biochemical examples with the structure of enzyme-catalyzed dual-E controllers are discussed.

PIWI-interacting RNAs (piRNAs) are small single-stranded RNAs that can repress transposon expression via epigenetic silencing and transcript degradation. They have been identified predominantly in the ovary and testis, where they serve essential roles in transposon silencing in order to protect the integrity of the genome in the germline. The potential expression of piRNAs in somatic cells has been controversial. In the present study we demonstrate the expression of piRNAs derived from both genic and transposon RNAs in the intersegmental muscles (ISMs) from the tobacco hawkmoth Manduca sexta. These piRNAs are abundantly expressed, ∼27 nt long, map antisense to transposons, are oxidation resistant, exhibit a 5’ uridine bias, and amplify via the canonical ping-pong pathway. An RNA-seq analysis demonstrated that 19 piRNA pathway genes are expressed in the ISMs and are developmentally regulated. The abundance of piRNAs does not change when the muscles initiate developmentally-regulated atrophy, but are repressed coincident with the commitment of the muscles undergo programmed cell death at the end of metamorphosis. This change in piRNA expression is correlated with the repression of several retrotransposons and the induction of specific DNA transposons. The developmentally-regulated changes in the expression of piRNAs, piRNA pathway genes, and transposons are all regulated by 20-hydroxyecdysone, the steroid hormone that controls the timing of ISM death. Taken together, these data provide compelling evidence for the existence of piRNA in somatic tissues and suggest that they may play roles in developmental processes such as programmed cell death.

O presente trabalho trata do desenvolvimento e estudo de um jogo digital educativo intitulado “Ping Pong Aritmético”, aplicado para auxiliar nos estudos das operações aritméticas nos primeiros anos do Ensino Fundamental anos finais. A pesquisa realizada é caracterizada como um método de abordagem exploratória descritiva. O recurso pedagógico foi utilizado por 230 alunos de três escolas da rede pública de Manaus - AM. A coleta de dados foi feita a partir da realização de observações, questionários e entrevistas e contou com a colaboração de 23 acadêmicos do PIBID Matemática da Universidade Federal do Amazonas - UFAM. O jogo foi inicialmente pensado para ser jogado de forma presencial, devido a pandemia, as regras foram adaptadas para que pudesse ser realizado por meio remoto. A aplicação do jogo evidenciou de inicio a dificuldade dos estudantes com as operações básicas e posteriormente uma melhora em relação a essas operações. O jogo teve uma avaliação positiva dos acadêmicos que se mostraram dispostos em fazer uso desse recurso em suas aulas. Por fim, a dinâmica em grupo permitiu, mesmo nesses tempos difíceis de pandemia, proporcionar uma espécie de “socialização a distância”, quebrando a monotonia das aulas online.

Arboviruses transmitted by mosquitoes are responsible for the death of millions of people each year. In addition to arboviruses, many insect-specific viruses (ISVs) have been discovered in mosquitoes in the last decade. ISVs, in contrast to arboviruses transmitted by mosquitoes to vertebrates, cannot replicate in vertebrate cells even when they are evolutionarily closely related to arboviruses. The alphavirus genus includes many arboviruses, although only a few ISVs have been discovered from this genus so far. Here, we investigate the interactions of a recently isolated insect-specific alphavirus, Agua-Salud alphavirus (ASALV), with its mosquito host. RNAi is one of the essential antiviral responses against arboviruses, although there is little knowledge on the interactions of RNAi with ISVs. Through knock-down of transcripts of the different key RNAi pathway (siRNA, miRNA and piRNA) proteins, we show the antiviral role of Ago2 (siRNA), Ago1 (miRNA), and Piwi4 proteins against ASALV in Aedes aegypti derived cells. ASALV replication increased in Dicer2 and Ago2 knock-out cells, confirming the antiviral role of the siRNA pathway. In infected cells, mainly ASALV-specific siRNAs are produced while piRNAs, with the characteristic nucleotide bias resulting from ping-pong amplification, are only produced in Dicer2 knock-out cells. Taken together, ASALV interactions with the mosquito RNAi response differs from arthropod-borne alphaviruses in some aspects, although they also share some commonalities. Further research is needed to understand whether the identified differences can be generalised to other insect-specific alphaviruses.

AbstractRNA helicases are enzymes that generally unwind double-stranded RNA using ATP hydrolysis energy, mainly involved in RNA metabolism, transcription, translation, and mRNA splicing. While the helicase core is crucial for RNA unwinding activity, N- and C-terminal extensions of specific helicases may contain an intrinsically disordered region for electrostatic interaction, resulting in the formation of droplets in the cytoplasm. However, how the disordered region of the RNA helicase contributes to RNA unwinding and dissociation remains unclear. Here, we focused on Bombyx mori Vasa, which unwinds truncated target transposon RNAs from the piRNA-induced silencing complex piRISC. In this study, we used single-molecule techniques to visualise how Vasa dynamically interacts with piRISC and investigate how Vasa oligomerization is involved in the process of piRNA amplification, named the ping-pong pathway. We found that Vasa’s oligomerization is required during these processes in vitro and in vivo, and that Vasa triggers the dissociation of truncated RNA in heterogeneous pathways. Our single-molecule results suggest that oligomerized Vasa guides the timing of the process regulating overall dissociation efficiency.
