Amla (Emblica officinalisGaertn.) has been used for many centuries in traditional Indian Ayurvedic formulations for the prevention and treatment of many inflammatory diseases. The present study evaluated the anti-inflammatory and anticoagulant properties of amla fruit extract. The amla fruit extract potentially and significantly reduced lipopolysaccharide (LPS)-induced tissue factor expression and von Willebrand factor release in human umbilical vein endothelial cells (HUVEC)in vitroat clinically relevant concentrations (1–100 μg/ml). In a leucocyte adhesion model of inflammation, it also significantly decreased LPS-induced adhesion of human monocytic cells (THP-1) to the HUVEC, as well as reduced the expression of endothelial-leucocyte adhesion molecule-1 (E-selectin) in the target cells. In addition, thein vivoanti-inflammatory effects were evaluated in a LPS-induced endotoxaemia rat model. Oral administration of the amla fruit extract (50 mg/kg body weight) significantly decreased the concentrations of pro-inflammatory cytokines, TNF-α and IL-6 in serum. These results suggest that amla fruit extract may be an effective anticoagulant and anti-inflammatory agent.

Background: Momordica charantia (MC) (bitter gourd) have shown the inhibition of NF-κβ and Leukotrienes expression in many inflammatory pathological conditions. Based on ist anti-inflammatory action this study aimed to Identify MC fruit dry powder (MCp) and MC fresh juice (MCj) action on airway inflammation in Guinea Pig model and also to find out the presence of alkaloids and flavonoides.Methods: 18 adult Guinea pigs of both sexes (excluding mating animals) were randomly divided into three groups with six animals in each namely Montelukast, MCj and MCp groups respectively. They were given with OD oral administration of concerned drugs for 1 -7 days. 6 control animals were exposed to 2% histamine aerosol for 120 seconds using histamine chamber prior to treatment with Montelukast, MCj and MCp to observe the normal Pre-Convulsive Dyspnea (PCD) in seconds. After six hours of daily OD oral dose of test drugs administration, all the groups’ were exposed to 2% Histamine aerosol one by one for 120 sec on day1 & 7 to observe PCD. The MCj and MCp were also tested with chemical assay, TLC to confirm the presence of alkaloids, flavonoides.Results: ON DAY 1 and 7: MCj and MCp showed significant decrease in PCD occurrence and it is statistically significant compared to the control. PCD blocking action of MCj group was effective in DAY 1. TLC and Chemical Assay were not supportive for bronchodilator action.Conclusions: MCj and MCp have promising preventive role in asthma. This study had expressed the positive protective role of Momordica charantia in asthmatic condition based on its known anti-inflammatory action.

Objective: Momordica charantia fruit extract and antidiabetic drug Nateglinide might be used simultaneously in the treatment of diabetes, so the objective of this study was to investigate pharmacokinetic herb-drug interactions of Momordica charantia fruit extract and pure charantin with nateglinide in rats. Methods: After oral co-administration of Momordica charantia fruit extract (250 mg/kg) and Charantin (10 mg/kg) with nateglinide in rats, drug concentration parameters peak plasma concentration (Cmax), time to reach peak plasma concentration (tmax), elimination half-life (t1/2), apparent volume of distribution (Vd), plasma clearance (Cl), and area under the curve (AUC) were calculated by using the non-compartment model. Results: NAT was absorbed into the circulatory system and reached its peak concentration approximately 2 h after being administered individually. tmax of groups co-administered NAT+MCE has been changed to 4h. A significant decrease in Cmax of NAT from 16.28 µg/ml to 11.37 µg/ml and 10.37 µg/ml with NAT with charantin and NAT with MCE groups, respectively. AUC of NAT decreased from 84.53 h/µg/ml to 53.63 h/µg/ml and 47.17 h/µg/ml by co-administration with Charantin and MCE respectively. Co-administration of nateglinide with Charantin and Momordica charantia fruit extract decreased systemic exposure level of nateglinide in vivo with decreasing Cmax and AUC and an increase in t1/2, Cl and Vd. Conclusion: From this study, it can be concluded that nateglinide, Momordica charantia fruit extract, and pure Charantin existed pharmacokinetic herb-drug interactions in the rat which has to be correlated with the anti-diabetic study. Further studies should be done to understand the effect of other herbal ingredients of Momordica charantia fruit extract on nateglinide as well as to predict the herb-drug interaction in humans.

Abstract Background Arthritis is a disorder of change in joint architecture and ligament degeneration. Rheumatoid arthritis is an autoimmune disorder in which body’s immune system targets own cells and degrade them. Martynia annua L. has been used in Indian traditional therapies for the treatment of epilepsy, many types of inflammations, respiratory infection, sore throat and wound. The objective of the present study was to determine anti-inflammatory and anti-arthritic activities of M. annua ethanolic fruit extract. Methods The extraction was performed using ethanol as a solvent followed by phytochemical investigation of M. annua ethanolic fruit extract. Antioxidant and anti-hyaluronidase enzyme inhibition activities was performed for the fruit extract. In vivo anti-inflammatory of fruit extract was performed on Calotropis procera latex (CPL) induced paw edema in rats using ibuprofen as standard. Inflammation was observed at 0, 1, 2, 4 and 6 h. In vivo anti-arthritic of fruit extract was performed on Complete Freund’s adjuvant (CFA) induced arthritis in rats. Arthritis was observed at 0, 7, 14 and 21 day. X-Ray study was also performed for inflammatory and arthritic paw of rats. Results The qualitative phytochemical screening of fruit extract showed presence of flavonoids, terpenoids, saponins, tannins, steroids, glycosides, proteins, carbohydrates, amino acids and polysaccharides. The antioxidant activity of fruit extract was 49.1 as compared to standard 45.73 at 100 μl dose. The anti-hyaluronidase enzyme inhibition activity of fruit extract was 84.60 as compared to standard 94.21 at 100 μl dose. Conclusion It is evident from the study that Martynia annua L. extract possess both antioxidant and hyalurinodase inhibition activity at dose dependent manner as well as anti-arthritic and anti-inflammatory potential.
